    The Economics of Efficient Phosphorus Abatement in a Watershed

    This study presents a method to determine efficient environmental targets at a watershed level. Efficient targets are devised by estimating abatement cost and cost of environmental damages and minimizing their sum. The method was applied to a case study of phosphorus pollution in a watershed in Oklahoma. Several cumulative scenarios with alternative abatement options were simulated and efficient targets were determined. As the number of abatement options at disposal to agricultural sources increased, their optimal abatement expanded relative to the abatement at the point source.     The evolution of mass loaded supernova remnants: II. Temperature dependent mass injection rates

    We investigate the evolution of spherically symmetric supernova remnants in which mass loading takes place due to conductively driven evaporation of embedded clouds. Numerical simulations reveal significant differences between the evolution of conductively mass loaded and the ablatively mass loaded remnants studied in Paper I. A main difference is the way in which conductive mass loading is extinguished at fairly early times, once the interior temperature of the remnant falls below ~ 107 K. Thus, at late times remnants that ablatively mass load are dominated by loaded mass and thermal energy, while those that conductively mass load are dominated by swept-up mass and kinetic energy.     Restoring mucosal barrier function and modifying macrophage phenotype with an extracellular matrix hydrogel: potential therapy for ulcerative colitis

    Background & Aims: Despite advances in therapeutic options, more than half of all patients with ulcerative colitis (UC) do not achieve long-term remission, many require colectomy, and the disease still has a marked negative impact on quality of life. Extracellular matrix (ECM) bioscaffolds facilitate the functional repair of many soft tissues by mechanisms that include mitigation of pro-inflammatory macrophage phenotype and mobilization of endogenous stem/progenitor cells. The aim of the present study was to determine if an ECM hydrogel therapy could influence outcomes in an inducible rodent model of UC. Methods: The dextran sodium sulfate (DSS)-colitis model was used in male Sprague Dawley rats. Animals were treated via enema with an ECM hydrogel and the severity of colitis was determined by clinical and histologic criteria. Lamina propria cells were isolated and the production of inflammatory mediators was quantified. Mucosal permeability was assessed in-vivo by administering TRITC-dextran and in-vitro using transepithelial electrical resistance (TEER). Results: ECM hydrogel therapy accelerated healing and improved outcome. The hydrogel was adhesive to colonic tissue, which allowed for targeted delivery of the therapy, and resulted in a reduction in clinical and histologic signs of disease. ECM hydrogel facilitated functional improvement of colonic epithelial barrier function and the resolution of the pro-inflammatory state of tissue macrophages. Conclusions: The present study shows that a nonsurgical and nonpharmacologic ECM-based therapy can abate DSS-colitis not by immunosuppression but by promoting phenotypic change in local macrophage phenotype and rapid replacement of the colonic mucosal barrie

    Evidence that distinct states of the integrin alpha6beta1 interact with laminin and an ADAM

    Integrins can exist in different functional states with low or high binding capacity for particular ligands. We previously provided evidence that the integrin alpha6beta1, on mouse eggs and on alpha6-transfected cells, interacted with the disintegrin domain of the sperm surface protein ADAM 2 (fertilin beta). In the present study we tested the hypothesis that different states of alpha6beta1 interact with fertilin and laminin, an extracellular matrix ligand for alpha6beta1. Using alpha6-transfected cells we found that treatments (e.g., with phorbol myristate acetate or MnCl2) that increased adhesion to laminin inhibited sperm binding. Conversely, treatments that inhibited laminin adhesion increased sperm binding. Next, we compared the ability of fluorescent beads coated with either fertilin beta or with the laminin E8 fragment to bind to eggs. In Ca2+-containing media, fertilin beta beads bound to eggs via an interaction mediated by the disintegrin loop of fertilin beta and by the alpha6 integrin subunit. In Ca2+-containing media, laminin E8 beads did not bind to eggs. Treatment of eggs with phorbol myristate acetate or with the actin disrupting agent, latrunculin A, inhibited fertilin bead binding, but did not induce laminin E8 bead binding. Treatment of eggs with Mn2+ dramatically increased laminin E8 bead binding, and inhibited fertilin bead binding. Our results provide the first evidence that different states of an integrin (alpha6beta1) can interact with an extracellular matrix ligand (laminin) or a membrane-anchored cell surface ligand (ADAM 2)

    NRG Oncology-Radiation Therapy Oncology Group Study 1014: 1-Year Toxicity Report From a Phase 2 Study of Repeat Breast-Preserving Surgery and 3-Dimensional Conformal Partial-Breast Reirradiation for In-Breast Recurrence.

    PURPOSE: To determine the associated toxicity, tolerance, and safety of partial-breast reirradiation. METHODS AND MATERIALS: Eligibility criteria included in-breast recurrence occurring \u3e1 year after whole-breast irradiation, \u3c3 \u3ecm, unifocal, and resected with negative margins. Partial-breast reirradiation was targeted to the surgical cavity plus 1.5 cm; a prescription dose of 45 Gy in 1.5 Gy twice daily for 30 treatments was used. The primary objective was to evaluate the rate of grade ≥3 treatment-related skin, fibrosis, and/or breast pain adverse events (AEs), occurring ≤1 year from re-treatment completion. A rate of ≥13% for these AEs in a cohort of 55 patients was determined to be unacceptable (86% power, 1-sided α = 0.07). RESULTS: Between 2010 and 2013, 65 patients were accrued, and the first 55 eligible and with 1 year follow-up were analyzed. Median age was 68 years. Twenty-two patients had ductal carcinoma in situ, and 33 had invasive disease: 19 ≤1 cm, 13 \u3e1 to ≤2 cm, and 1 \u3e2 cm. All patients were clinically node negative. Systemic therapy was delivered in 51%. All treatment plans underwent quality review for contouring accuracy and dosimetric compliance. All treatment plans scored acceptable for tumor volume contouring and tumor volume dose-volume analysis. Only 4 (7%) scored unacceptable for organs at risk contouring and organs at risk dose-volume analysis. Treatment-related skin, fibrosis, and/or breast pain AEs were recorded as grade 1 in 64% and grade 2 in 7%, with only 1 ( CONCLUSION: Partial-breast reirradiation with 3-dimensional conformal radiation therapy after second lumpectomy for patients experiencing in-breast failures after whole-breast irradiation is safe and feasible, with acceptable treatment quality achieved.     Impact of Systematic Errors in Sunyaev-Zel'dovich Surveys of Galaxy Clusters

    Future high-resolution microwave background measurements hold the promise of detecting galaxy clusters throughout our Hubble volume through their Sunyaev-Zel'dovich (SZ) signature, down to a given limiting flux. The number density of galaxy clusters is highly sensitive to cluster mass through fluctuations in the matter power spectrum, as well as redshift through the comoving volume and the growth factor. This sensitivity in principle allows tight constraints on such quantities as the equation of state of dark energy and the neutrino mass. We evaluate the ability of future cluster surveys to measure these quantities simultaneously when combined with PLANCK-like CMB data. Using a simple effective model for uncertainties in the cluster mass-SZ flux relation, we evaluate systematic shifts in cosmological constraints from cluster SZ surveys. We find that a systematic bias of 10% in cluster mass measurements can give rise to shifts in cosmological parameter estimates at levels larger than the 1σ1\sigma statistical errors. Systematic errors are unlikely to be detected from the mass and redshift dependence of cluster number counts alone; increasing survey size has only a marginal effect.
