Phase I trial of the DLL3/CD3 bispecific T-cell engager BI 764532 in DLL3-positive small-cell lung cancer and neuroendocrine carcinomas
Poorly differentiated neuroendocrine carcinomas such as small-cell lung cancer (SCLC) have poor survival and high relapse rates. DLL3 is found on these carcinomas and has become a target of increasing interest in recent years. The bispecific DLL3/CD3 T-cell engager BI 764532 has been shown to induce complete tumor regression in a human T cell-engrafted mouse model. Here, we describe the study design of a first-in-human, phase I, multicenter, open-label, non-randomized, dose-escalation study in patients with SCLC or other DLL3-positive neuroendocrine carcinomas. The study will determine the maximum tolerated dose and evaluate safety, tolerability, pharmacokinetics and preliminary efficacy of BI 764532 monotherapy
Iron Overload in Allogeneic Hematopoietic Cell Transplantation Outcome: A Meta-Analysis
AbstractAn elevated ferritin level before allogeneic hematopoietic cell transplantation (HCT) is an adverse prognostic factor for overall survival (OS) and nonrelapse mortality. Because ferritin is an imperfect surrogate of iron stores, the prognostic role of iron overload remains unclear. We conducted a patient-level meta-analysis of 4Â studies that used magnetic resonance imaging to estimate pre-HCT liver iron content (LIC). An elevated LIC was not associated with a significant increase in mortality: the hazard ratio (HR) for mortality associated with LICÂ >Â 7Â mg/g dry weight (primary endpoint) was 1.4 (PÂ = .18). In contrast, ferritin >1000Â ng/mL was a significant prognostic factor (HR for mortality, 1.7; PÂ = .036). There was, however, no significant association between ferritin > 2500 and mortality. This meta-analysis suggests that iron overload, as assessed by LIC, is not a strong prognostic factor for OS in a general adult HCT population. Our data also suggest that ferritin is an inadequate surrogate for iron overload in HCT
Sex stereotypes influence adults' perception of babies' cries
Background: Despite widespread evidence that gender stereotypes influence human parental behavior, their potential effects on adultsâ perception of babiesâ cries have been overlooked. In particular, whether adult listeners overgeneralize the sex dimorphism that characterizes the voice of adult speakers (men are lower-pitched than women) to their perception of babiesâ cries has not been investigated.
Methods: We used playback experiments combining natural and re-synthesised cries of 3 month-old babies to investigate whether the interindividual variation in the fundamental frequency (pitch) of cries affected adult listenersâ identification of the babyâs sex, their perception the babyâs femininity and masculinity, and whether these biases interacted with their perception of the level of discomfort expressed by the cry.
Results: We show that low-pitched cries are more likely to be attributed to boys and high-pitched cries to girls, despite the absence of sex differences in pitch. Moreover, low-pitched boys are perceived as more masculine and high-pitched girls are perceived as more feminine. Finally, adult men rate relatively low-pitched cries as expressing more discomfort when presented as belonging to boys than to girls.
Conclusion: Such biases in caregiversâ responses to babiesâ cries may have implications on childrenâs immediate welfare and on the development of their gender identity
First-in-Human Dose Escalation Trial To Evaluate the Clinical Safety and Efficacy of an Anti-MAGEA1 Autologous Tcr-Transgenic T Cell Therapy in Relapsed and Refractory Solid Tumors
RATIONALE OF THE TRIAL: Although the use of engineered T cells in cancer immunotherapy has greatly advanced the treatment of hematological malignancies, reaching meaningful clinical responses in the treatment of solid tumors is still challenging. We investigated the safety and tolerability of IMA202 in a first-in-human, dose escalation basket trial in human leucocyte antigen A*02:01 positive patients with melanoma-associated antigen A1 (MAGEA1)-positive advanced solid tumors.
TRIAL DESIGN: The 2+2 trial design was an algorithmic design based on a maximally acceptable dose-limiting toxicity (DLT) rate of 25% and the sample size was driven by the algorithmic design with a maximum of 16 patients. IMA202 consists of autologous genetically modified cytotoxic CD8
CONCLUSION: In conclusion, IMA202 had a manageable safety profile, and it was associated with biological and potential clinical activity of MAGEA1-targeting genetically engineered TCR-T cells in a poor prognosis, multi-indication solid tumor cohort
Non-affirmative Theory of Education as a Foundation for Curriculum Studies, Didaktik and Educational Leadership
This chapter presents non-affirmative theory of education as the foundation for a new research program in education, allowing us to bridge educational leadership, curriculum studies and Didaktik. We demonstrate the strengths of this framework by analyzing literature from educational leadership and curriculum theory/didaktik. In contrast to both socialization-oriented explanations locating curriculum and leadership within existing society, and transformation-oriented models viewing education as revolutionary or super-ordinate to society, non-affirmative theory explains the relation between education and politics, economy and culture, respectively, as non-hierarchical. Here critical deliberation and discursive practices mediate between politics, culture, economy and education, driven by individual agency in historically developed cultural and societal institutions. While transformative and socialization models typically result in instrumental notions of leadership and teaching, non-affirmative education theory, previously developed within German and Nordic education, instead views leadership and teaching as relational and hermeneutic, drawing on ontological core concepts of modern education: recognition; summoning to self-activity and Bildsamkeit. Understanding educational leadership, school development and teaching then requires a comparative multi-level approach informed by discursive institutionalism and organization theory, in addition to theorizing leadership and teaching as cultural-historical and critical-hermeneutic activity. Globalisation and contemporary challenges to deliberative democracy also call for rethinking modern nation-state based theorizing of education in a cosmopolitan light. Non-affirmative education theory allows us to understand and promote recognition based democratic citizenship (political, economical and cultural) that respects cultural, ethical and epistemological variations in a globopolitan era. We hope an American-European-Asian comparative dialogue is enhanced by theorizing education with a non-affirmative approach
Fundamental frequency of neonatal crying: Does body size matter?
Summary: The objective of this study was to determine the influence of fetal growth on the fundamental frequency (F0) of neonatal crying in a group of healthy full term infants. The spontaneous cries of 131 infants were audio recorded during the first week of life, and subsequently submitted to acoustic analyses. The individual cry utterances produced by each infant were measured for minimum, mean, and maximum F0. The infants were placed into one of three groupings (low, average, high) based on body size indices according to the ponderal index (PI), the ratio of body weight to body length (BW/L), and body weight (BW) alone. The F0 features of infants in each sub-grouping of body size were compared and contrasted. The results indicated that features of cry F0 were found to decrease marginally as a function of increased body size, with significant group differences confined to maximum F0. The BW index appeared to be the most sensitive measure in differentiating infant groups according to body size. In general neonatal body size appears to have a slight, although non-significant influence on the vocal F0 of crying in healthy full term infants. Any body size related changes in cry F0 are likely to be found for maximum F0 and may reflect stress-related variations in nervous system activation
