    Global genetic variation of select opiate metabolism genes in self-reported healthy individuals

    CYP2D6 is a key pharmacogene encoding an enzyme impacting poor, intermediate, extensive and ultrarapid phase I metabolism of many marketed drugs. The pharmacogenetics of opiate drug metabolism is particularly interesting due to the relatively high incidence of addiction and overdose. Recently, trans-acting opiate metabolism and analgesic response enzymes (UGT2B7, ABCB1, OPRM1 and COMT) have been incorporated into pharmacogenetic studies to generate more comprehensive metabolic profiles of patients. With use of massively parallel sequencing, it is possible to identify additional polymorphisms that fine tune, or redefine, previous pharmacogenetic findings, which typically rely on targeted approaches. The 1000 Genomes Project data were analyzed to describe population genetic variation and statistics for these five genes in self-reported healthy individuals in five global super- and 26 sub-populations. Findings on the variation of these genes in various populations expand baseline understanding of pharmacogenetically relevant polymorphisms for future studies of affected cohorts.Peer reviewe

    Modus Vivendi Beyond the Social Contract: Peace, Justice, and Survival in Realist Political Theory

    This essay examines the promise of the notion of modus vivendi for realist political theory. I interpret recent theories of modus vivendi as affirming the priority of peace over justice, and explore several ways of making sense of this idea. I proceed to identify two key problems for modus vivendi theory, so conceived. Normatively speaking, it remains unclear how this approach can sustain a realist critique of Rawlsian theorizing about justice while avoiding a Hobbesian endorsement of absolutism. And conceptually, the theory remains wedded to a key feature of social contract theory: political order is conceived as based on agreement. This construes the horizontal tensions among individual or group agents in society as prior to the vertical, authoritative relations between authorities and their subjects. Political authority thereby appears from the start as a solution to societal conflict, rather than a problem in itself. I argue that this way of framing the issue abstracts from political experience. Instead I attempt to rethink the notion of modus vivendi from within the lived experience of political conflict, as oriented not primarily toward peace, but political survival. With this shift of perspective, the idea of modus vivendi shows us, pace Bernard Williams, that the ā€œfirst political questionā€ is not how to achieve order and stability, but rather: what can I live with

    Evaluating the performance of ionic liquid coatings for mitigation of spacecraft surface charges

    To reduce the impact of charging effects on satellites, cheap and lightweight conductive coatings are desirable. We mimic space-like charging environments in ultra-high vacuum (UHV) chambers during deposition of charges via the electron beam of a scanning electron microscope (SEM). We use the charge induced signatures in SEM images of a thin ionic liquid (IL) film on insulating surfaces such as glass, to assess the general performance of such coatings. In order to get a reference structure in SEM, the samples were structured by nanosphere lithography and coated with IL. The IL film (we choose BMP DCA, due to its beneficial physical properties) was applied ex situ and a thickness of 10 to 30 nm was determined by reflectometry. Such an IL film is stable under vacuum conditions. It would also only lead to additional mass of below 20 mg/m2^2. At about 5 A/m2ā‰ˆ3ā‹…1019^2 \approx 3\cdot10^{19} e/(sā‹…\cdotm2^2), a typical sample charging rate in SEM, imaging is possible with no noticeable contrast changes over many hours; this electron current density is already 6 orders of magnitudes higher than "worst case geosynchronous environments" of 3ā‹…10āˆ’63\cdot10^{-6} A/m2^2. Measurements of the surface potential are used for further insights in the reaction of IL films to the electron beam of a SEM.     Graphene Oxide alpha Bi2O3 Composites for Visible Light Photocatalysis, Chemical Catalysis and Solar Energy Conversion

    The growing challenges of environmental purification by solar photocatalysis, precious metal free catalysis and photocurrent generation in photovoltaic cells are receiving the utmost global attention. Here we demonstrate the one pot green chemical synthesis of a new stable heterostructured, eco friendly, multifunctional micro composite consisting of amp; 945; Bi2O3 micro needles intercalated with anchored graphene oxide GO micro sheets 1.0 wt for the above mentioned applications in a large economical scale. The bare amp; 945; Bi2O3 micro needles display twice as better photocatalytic activities than commercial TiO2 Degussa P25 while the GO hybridized composite exhibit 4 6 times enhanced photocatalytic activities than neat TiO2 photocatalyst in the degradation of colored aromatic organic dyes crystal violet and rhodamine 6G under visible light irradiation 300 W tungsten lamp . The highly efficient activity is associated with the strong surface adsorption ability of GO for aromatic dye molecules, the high carrier acceptability and efficient electron hole pair separation in Bi2O3 by individual adjoining GO sheets. Introduction of Ag nanoparticles 2.0 wt further enhances the photocatalytic performance of the composite over 8 folds due to a plasmon induced electron transfer process from Ag nanoparticles via GO sheets into the conduction band of Bi2O3. The new composites are also catalytically active. They catalyze the reduction of 4 nitrophenol to 4 aminophenol in presence of borohydride ions. Photoanodes assembled from GO amp; 945; Bi2O3 and Ag GO amp; 945; Bi2O3 composites display an improved photocurrent response power conversion efficiency 20 higher over those prepared without GO in dye sensitized solar cells DSSCs

    An observational prospective study of topical acidified nitrite for killing methicillin-resistant Staphylococcus aureus (MRSA) in contaminated wounds

    Background Endogenous nitric oxide (NO) kills bacteria and other organisms as part of the innate immune response. When nitrite is exposed to low pH, NO is generated and has been used as an NO delivery system to treat skin infections. We demonstrated eradication of MRSA carriage from wounds using a topical formulation of citric acid (4.5%) and sodium nitrite (3%) creams co-applied for 5 days to 15 wounds in an observational prospective pilot study of 8 patients. Findings Following treatment with topical citric acid and sodium nitrite, 9 of 15 wounds (60%) and 3 of 8 patients (37%) were cleared of infection. MRSA isolates from these patients were all sensitive to acidified nitrite in vitro compared to methicillin-sensitive S. aureus and a reference strain of MRSA. Conclusions Nitric oxide and acidified nitrite offer a novel therapy for control of MRSA in wounds. Wounds that were not cleared of infection may have been re-contaminated or the bioavailability of acidified nitrite impaired by local factors in the tissue

    A comprehensive multidisciplinary care pathway for hip fractures better outcome than usual care?

    Introduction: Hip fracture surgery is among the most performed surgical procedures in elderly patients. Mortality rates are high, however, and patients often fail to live independently following a hip fracture. To improve outcome, multidisciplinary care pathways have been initiated, but longer-term results are lacking. Aim of this study was to compare functional outcome and living situation six months after hip fracture treatment with and without a care pathway. Patients and methods: A multicentre prospective controlled trial was conducted with three hospitals: in one hospital patients were treated with a care pathway, in the other hospitals patients received usual care. All patients aged >= 60 years witha hipfracture were asked to participate. Besides basic characteristics, health-related quality of life (EQ-5D) and performance scores of activities of daily living (Katz Index and Lawton IADL) were assessed. Differences in scores were analysed using linear regression. Propensity score adjustment was used to correct for differences between the care pathway and the usual care group. Missing data were imputed. Results: No differences in rate of return to prefracture ADL level were found between patients in the care pathway group and the usual care group. The percentage of participants in the same situation as before the fracture was the same in both treatment groups (81%). There were no significant differences in quality of life, activities of daily living or mortality (15% vs 10%, p = 0.17), but hospital stay in the care pathway group was significantly shorter (median 7 vs 10 days). Discussion: Treatment of elderly patients with a hip fracture is commonly organised in care pathways. Although short-term advantages are reported, positive effects on longer-term functional results could not be proven in our study. This study confirmed a shorter hospital stay in the care pathway group, which potentially may lead to a reduction in costs. Conclusions: Functional outcome and living situation six months after a hip fracture is the same for patients treated with or without a care pathway. (C) 2021 The Author(s). Published by Elsevier Ltd
