    Upregulation of microRNA-25 associates with prognosis in hepatocellular carcinoma

    BACKGROUND: Accumulating evidence has shown that up-regulation of microRNA-25(miR-25) is associated with the prognosis of several types of human malignant solid tumors. However, whether miR-25 expression has influence on the prognosis of hepatocellular carcinoma (HCC) is still unknown. METHODS: The differentially expressed amount of the miR-25 was validated in triplicate by quantitative reverse-transcription polymerase chain reaction (qRT-PCR). Survival rate was analyzed by log-rank test, and survival curves were plotted according to Kaplan–Meier. Multivariate analysis of the prognostic factors was performed with Cox regression model. RESULTS: The expression of miR-25 was significantly upregulated in HCC tissues when compared with adjacent normal tissues (p<0.0001). Patients who had high miR-25 expression had a shorter overall survival than patients who had low miR-25 expression (median overall survival, 31.0 months versus 42.9 months, p=0.0192). The multivariate Cox regression analysis indicated that miR-25 expression (HR=2.179; p=0.001), TNM stage (HR=1.782; p=0.014), and vein invasion (HR=1.624; p=0.020) were independent prognostic factors for overall survival. CONCLUSION: Our data suggests that the overexpression of miR-25 in HCC tissues is of predictive value on poor prognosis.     Exploring Charge Dissociation in a Statistical Sample of Active-Layer Models of an Organic Solar Cell

    Charge dissociation in the active layer is one of the key factors for the power conversion efficiency of bulk heterojunction organic solar cells (OSCs). Numerous charge-transfer mechanisms have been proposed based on one of few microscopic models. Here, we would explore possible charge-transfer mechanisms for 155 models of donor/acceptor (D/A) interfaces, built via materials DCV-1 and C60 as the donor and acceptor, respectively. After the calculations of the key parameters related to the charge dissociation and a statistical analysis for the correlation between these parameters were carried out, we can obtain a more robust description of the charge dissociation in practical OSCs. The complicated relationship among the key parameters not only illustrates the important correlation between the D/A stacking pattern and charge-transfer mechanism but also suggests that different charge-transfer mechanisms take place more likely depending on the specific arrangements of the donor and acceptor

    PASNet: Polynomial Architecture Search Framework for Two-party Computation-based Secure Neural Network Deployment

    Two-party computation (2PC) is promising to enable privacy-preserving deep learning (DL). However, the 2PC-based privacy-preserving DL implementation comes with high comparison protocol overhead from the non-linear operators. This work presents PASNet, a novel systematic framework that enables low latency, high energy efficiency & accuracy, and security-guaranteed 2PC-DL by integrating the hardware latency of the cryptographic building block into the neural architecture search loss function. We develop a cryptographic hardware scheduler and the corresponding performance model for Field Programmable Gate Arrays (FPGA) as a case study. The experimental results demonstrate that our light-weighted model PASNet-A and heavily-weighted model PASNet-B achieve 63 ms and 228 ms latency on private inference on ImageNet, which are 147 and 40 times faster than the SOTA CryptGPU system, and achieve 70.54% & 78.79% accuracy and more than 1000 times higher energy efficiency.Comment: DAC 2023 accepeted publication, short version was published on AAAI 2023 workshop on DL-Hardware Co-Design for AI Acceleration: RRNet: Towards ReLU-Reduced Neural Network for Two-party Computation Based Private Inferenc

    Genome-wide identification and comprehensive analysis of tubby-like protein gene family in multiple crops

    IntroductionThe highly conserved tubby-like proteins (TLPs) play key roles in animal neuronal development and plant growth. The abiotic stress tolerance function of TLPs has been widely explored in plants, however, little is known about comparative studies of TLPs within crops.MethodsBioinformatic identification, phylogenetic analysis, Cis-element analysis, expression analysis, Cis-element analysis, expression analysis and so on were explored to analysis the TLP gene family of multiple crops.ResultsIn this study, a comprehensive analysis of TLP genes were carried out in seven crops to explore whether similar function of TLPs in rice could be achieved in other crops. We identified 20, 9, 14, 11, 12, 35, 14 and 13 TLP genes in Glycine max, Hordeum vulgare, Sorghum bicolor, Arabidopsis thaliana, Oryza sativa Japonica, Triticum aestivum, Setaria italic and Zea mays, respectively. All of them were divided into two groups and ten orthogroups (Ors) based on amino acids. A majority of TLP genes had two domains, tubby-like domain and F-box domain, while members of Or5 only had tubby-like domain. In addition, Or5 had more exons and shorter DNA sequences, showing that characteristics of different Ors reflected the differentiated function and feature of TLP genes in evolutionary process, and Or5 was the most different from the other Ors. Besides, we recognized 25 cis-elements in the promoter of TLP genes and explored multiple new regulation pathway of TLPs including light and hormone response. The bioinformatic and transcriptomic analysis implied the stresses induced expression and possible functional redundancy of TLP genes. We detected the expression level of 6 OsTLP genes at 1 to 6 days after seed germination in rice, and the most obvious changes in these days were appeared in OsTLP10 and OsTLP12.DiscussionCombined yeast two-hybrid system and pull down assay, we suggested that the TLP genes of Or1 may have similar function during seed germination in different species. In general, the results of comprehensive analysis of TLP gene family in multiple species provide valuable evolutionary and functional information of TLP gene family which are useful for further application and study of TLP genes
