    Indole contributes to tetracycline resistance via the outer membrane protein OmpN in Vibrio splendidus

    As an interspecies and interkingdom signaling molecule, indole has recently received attention for its diverse effects on the physiology of both bacteria and hosts. In this study, indole increased the tetracycline resistance of Vibrio splendidus. The minimal inhibitory concentration of tetracycline was 10 mu g/mL, and the OD600 of V. splendidus decreased by 94.5% in the presence of 20 mu g/mL tetracycline; however, the OD600 of V. splendidus with a mixture of 20 mu g/mL tetracycline and 125 mu M indole was 10- or 4.5-fold higher than that with only 20 mu g/mL tetracycline at different time points. The percentage of cells resistant to 10 mu g/mL tetracycline was 600-fold higher in the culture with an OD600 of approximately 2.0 (higher level of indole) than that in the culture with an OD600 of 0.5, which also meant that the level of indole was correlated to the tetracycline resistance of V. splendidus. Furthermore, one differentially expressed protein, which was identified as the outer membrane porin OmpN using SDS-PAGE combined with MALDI-TOF/TOF MS, was upregulated. Consequently, the expression of the ompN gene in the presence of either tetracycline or indole and simultaneously in the presence of indole and tetracycline was upregulated by 1.8-, 2.54-, and 6.01-fold, respectively, compared to the control samples. The combined results demonstrated that indole enhanced the tetracycline resistance of V. splendidus, and this resistance was probably due to upregulation of the outer membrane porin OmpN

    COMPREHENSIVE UNDERSTANDING THE INHIBITORS AND ENABLERS OF KNOWLEDGE TRANSFER IN ERP ASSIMILATIONS: A MULTI-CASE STUDY

    In the enterprise resource planning (ERP) assimilation process, organizations are increasingly concerned about how to ensure employees to have sufficient ERP knowledge effectively. However, limited attention has been directed toward examining knowledge transfer in the assimilation stage systematically. In particular, a significant omission is to understand the key enablers and inhibitors of employees’ learning intention who may receiver knowledge passively in the mandatory setting. We employed a multi-case study method in this exploratory research by interviewing 33 ERP users at all levels in nine big-size firms in China. Results of this analysis suggested that causal ambiguity in new systems, incumbent system habit, and technostress significantly undermined recipients’ learning intentions. Meanwhile, perceived management support, relation embeddedness, and symbolic adoption were key determinants of increased their learning intentions. This study is arguably the first that attempts to look into passive knowledge transfer phenomenon in some depth, and extends prior researches in ERP lifecycle by shedding light on the joint influences of enablers and inhibitors in ERP assimilation context

    Coordinating a Supply Chain with a Loss-Averse Retailer under Yield and Demand Uncertainties

    This paper investigates the channel coordination of a supply chain (SC) consisting of a loss-averse retailer and a risk-neutral supplier under yield and demand uncertainties. Three existing contracts are analyzed. Our results demonstrate that the buyback (BB) and quantity flexibility (QF) contracts can not only coordinate the supply chain but also lead to Pareto improvement for each player, while the wholesale price (WP) contract fails to coordinate the chain due to the effects of double marginalization and risk preference. For comparison, a chain with a risk-neutral retailer is also analyzed. Furthermore, numerical examples are provided to demonstrate the effectiveness of the coordination contracts, and the impacts of loss aversion and random yield on the decision-making behaviors and system performance are then discussed

    Impact of Water Scarcity on the Fenhe River Basin and Mitigation Strategies

    This study produced a drought map for the Fenhe River basin covering the period from 150 BC to 2012 using regional historical drought records. Based on meteorological and hydrological features, the characteristics and causes of water scarcity in the Fenhe River basin were examined, along with their impact on the national economy and ecological environment. The effects of water scarcity in the basin on the national economy were determined from agricultural, industrial, and domestic perspectives. The impact on aquatic ecosystems was ascertained through an evolution trend analysis of surface water systems, including rivers, wetlands, and slope ecosystems, and subterranean water systems, including groundwater and karst springs. As a result of these analyses, strategies are presented for coping with water scarcity in this basin, including engineering countermeasures, such as the construction of a water network in Shanxi, and the non-engineering approach of groundwater resource preservation. These comprehensive coping strategies are proposed with the aim of assisting the prevention and control of water scarcity in the arid and semi-arid areas of China

    Altered insular functional connectivity correlates to impaired vigilant attention after sleep deprivation: A resting-state functional magnetic resonance imaging study

    ObjectivesThis study used resting-state functional magnetic resonance imaging (rs-fMRI) scans to assess the dominant effects of 36 h total sleep deprivation (TSD) on vigilant attention and changes in the resting-state network.Materials and methodsTwenty-two healthy college students were enrolled in this study. Participants underwent two rs-fMRI scans, once in rested wakefulness (RW) and once after 36 h of TSD. We used psychomotor vigilance tasks (PVT) to measure vigilant attention. The region-of-interest to region-of-interest correlation was employed to analyze the relationship within the salience network (SN) and between other networks after 36 h of TSD. Furthermore, Pearson’s correlation analysis investigated the relationship between altered insular functional connectivity and PVT performance.ResultsAfter 36 h of TSD, participants showed significantly decreased vigilant attention. Additionally, TSD induced decreased functional connectivity between the visual and parietal regions, whereas, a significant increase was observed between the anterior cingulate cortex and insula. Moreover, changes in functional connectivity in the anterior cingulate cortex and insula showed a significant positive correlation with the response time to PVT.ConclusionOur results suggest that 36 h of TSD impaired vigilant visual attention, resulting in slower reaction times. The decrease in visual-parietal functional connectivity may be related to the decrease in the reception of information in the brain.     Factors that affect the growth and photosynthesis of the filamentous green algae, Chaetomorpha valida, in static sea cucumber aquaculture ponds with high salinity and high pH

    Chaetomorpha valida, dominant filamentous green algae, can be harmful to sea cucumber growth in aquaculture ponds of China. In order to understand the environmental factors affecting the growth of C. valida in sea cucumber aquaculture ecosystems, a combination of field investigations and laboratory experiments were conducted. Field surveys over one year revealed that C. valida survived in sea cucumber aquaculture ponds in salinities ranging from 24.3 ± 0.01‰ to 32.0 ± 0.02‰ and a pH range of 7.5 ± 0.02–8.6 ± 0.04. The high salinity and pH during the period of low C. valida biomass from January to May lay the foundation for its rapid growth in the following months of June to October. Many factors interact in the field environment, thus, laboratory experiments were conducted to determine the isolated effects of pH and salinity on C. valida growth. In laboratory experiments, samples were incubated under different salinity and pH conditions at 25 °C, with a light intensity of 108 ÎŒmol photon·m−2·s−1, and a photoperiod of 12 L:12 D. Results showed that salinity and pH significantly affect the growth and Fv/Fm (quantum yield of photosynthesis) of C. valida (p < 0.01). C. valida grew the longest at a salinity of 34‰ and a pH of 8.0. At 34‰ salinity, C. valida grew to 26.44 ± 5.89 cm in 16 days. At a pH of 8.0, C. valida grew to 67.96 ± 4.45 cm in 32 days. Fv/Fm was 0.635 ± 0.002 at a salinity of 32‰, and 0.550 ± 0.006 to 0.660± 0.001 at pH 7.0 to 8.5. Based on these results, we conclude that C. valida can bloom in sea cucumber ponds due to the high salinity and pH of coastal sea waters, which promote growth and maintain the photosynthetic activity of C. valida

    A Pontine Region is a Neural Correlate of the Human Affective Processing Network

    The in vivo neural activity of the pons during the perception of affective stimuli has not been studied despite the strong implications of its role in affective processing. To examine the activity of the pons during the viewing of affective stimuli, and to verify its functional and structural connectivity with other affective neural correlates, a multimodal magnetic resonance imaging methodology was employed in this study. We observed the in vivo activity of the pons when viewing affective stimuli. Furthermore, small-world connectivity indicated that the functional connectivity (FC) between the pons and the cortico-limbic affective regions was meaningful, with the coefficient λ being positively associated with self-reported emotional reactivity. The FC between the pons and the cortico-limbic-striatal areas was related to self-reported negative affect. Corroborating this finding was the observation that the tract passing through the pons and the left hippocampus was negatively related to self-reported positive affect and positively correlated with emotional reactivity. Our findings support the framework that the pons works conjunctively with the distributed cortico-limbic-striatal systems in shaping individuals' affective states and reactivity. Our work paves the path for future research on the contribution of the pons to the precipitation and maintenance of affective disorders.published_or_final_versio
