    LDP-IDS: Local Differential Privacy for Infinite Data Streams

    Streaming data collection is essential to real-time data analytics in various IoTs and mobile device-based systems, which, however, may expose end users' privacy. Local differential privacy (LDP) is a promising solution to privacy-preserving data collection and analysis. However, existing few LDP studies over streams are either applicable to finite streams only or suffering from insufficient protection. This paper investigates this problem by proposing LDP-IDS, a novel ww-event LDP paradigm to provide practical privacy guarantee for infinite streams at users end, and adapting the popular budget division framework in centralized differential privacy (CDP). By constructing a unified error analysi for LDP, we first develop two adatpive budget division-based LDP methods for LDP-IDS that can enhance data utility via leveraging the non-deterministic sparsity in streams. Beyond that, we further propose a novel population division framework that can not only avoid the high sensitivity of LDP noise to budget division but also require significantly less communication. Based on the framework, we also present two adaptive population division methods for LDP-IDS with theoretical analysis. We conduct extensive experiments on synthetic and real-world datasets to evaluate the effectiveness and efficiency pf our proposed frameworks and methods. Experimental results demonstrate that, despite the effectiveness of the adaptive budget division methods, the proposed population division framework and methods can further achieve much higher effectiveness and efficiency.Comment: accepted to SIGMOD'2

    LoPub: High-Dimensional Crowdsourced Data Publication with Local Differential Privacy

    High-dimensional crowdsourced data collected from numerous users produces rich knowledge about our society. However, it also brings unprecedented privacy threats to the participants. Local differential privacy (LDP), a variant of differential privacy, is recently proposed as a state-of-the-art privacy notion. Unfortunately, achieving LDP on high-dimensional crowdsourced data publication raises great challenges in terms of both computational efficiency and data utility. To this end, based on Expectation Maximization (EM) algorithm and Lasso regression, we first propose efficient multi-dimensional joint distribution estimation algorithms with LDP. Then, we develop a Local differentially private high-dimensional data Publication algorithm, LoPub, by taking advantage of our distribution estimation techniques. In particular, correlations among multiple attributes are identified to reduce the dimensionality of crowdsourced data, thus speeding up the distribution learning process and achieving high data utility. Extensive experiments on realworld datasets demonstrate that our multivariate distribution estimation scheme significantly outperforms existing estimation schemes in terms of both communication overhead and estimation speed. Moreover, LoPub can keep, on average, 80% and 60% accuracy over the released datasets in terms of SVM and random forest classification, respectively

    Increased expression of MMP9 is correlated with poor prognosis of nasopharyngeal carcinoma

    <p>Abstract</p> <p>Introduction</p> <p>The aim of the present study was to analyze the expression of matrix metalloproteinase 9 (<it>MMP9</it>) in nasopharyngeal carcinoma (NPC) and its correlation with clinicopathologic features, including the survival of patients with NPC.</p> <p>Methods</p> <p>Using real-time PCR, we detected the mRNA expression of <it>MMP9 </it>in normal nasopharyngeal tissues and nasopharyngeal carcinoma (NPC) tissues. Using immunohistochemistry analysis, we analyzed <it>MMP9 </it>protein expression in clinicopathologically characterized 164 NPC cases (116 male and 48 female) with age ranging from 17 to 80 years (median = 48.4 years) and 32 normal nasopharyngeal tissues. Cases with greater than or equal to 6 and less than 6 of the score value of cytoplasmic <it>MMP9 </it>immunostaining were regarded as high expression and low expression, respectively. The relationship between the expression levels of <it>MMP9 </it>and clinical features was analyzed.</p> <p>Results</p> <p>The expression level of <it>MMP9 </it>mRNA was markedly greater in NPC tissues than that in the nasopharyngeal tissues. Immunohistochemical analysis revealed that the protein expression of <it>MMP9 </it>detected in NPC tissues was higher than that in the nasopharyngeal tissues (<it>P </it>= 0.004). In addition, high levels of <it>MMP9 </it>protein were positively correlated with the status of lymph node metastasis (N classification) (<it>P </it>= 0.002) and clinical stage (<it>P </it>< 0.001) of NPC patients. Patients with higher <it>MMP9 </it>expression had a significantly shorter overall survival time than did patients with low <it>MMP9 </it>expression. Multivariate analysis suggested that the level of <it>MMP9 </it>expression was an independent prognostic indicator (<it>P </it>= 0.008) for the survival of patients with NPC.</p> <p>Conclusion</p> <p>High level of <it>MMP9 </it>expression is a potential unfavorable prognostic factor for patients with NPC.</p

    Protective effect of Macleaya cordata isoquinoline alkaloids on lipopolysaccharide-induced liver injury in broilers

    Objective This experiment aimed to explore the protective action of dietary supplementation with isoquinoline alkaloids (IA) from Macleaya cordata on lipopolysaccharide (LPS)-induced liver injury in broilers. Methods Total 216 healthy broilers were selected in a 21-d trial and assigned randomly to the following 3 treatments: control (CON) group, LPS group, and LPS+IA group. The CON and LPS groups were provided with a basal diet, whereas the LPS+IA group received the basal diet supplemented with 0.6 mg/kg Macleaya cordata IA. Broilers in LPS and LPS+IA groups were intraperitoneally injected with LPS (1 mg/kg body weight) at 17, 19, and 21 days of age, while those in CON group were injected with equivalent amount of saline solution. Results Results showed LPS injection caused systemic and liver inflammation in broilers, inhibited immune function, and ultimately lead to liver injury. By contrast, supplementation of IA ameliorated LPS-induced adverse change in serum parameters, boosted immunity in LPS+IA group. Furthermore, IA suppressed the elevation of hepatic inflammatory cytokines and caspases levels induced by LPS, as well as the expressions of genes related to the toll-like receptor 4 (TLR4)/myeloid differentiation primary response 88 (MyD88)/nuclear factor-kappa B (NF-ÎşB) pathway. Conclusion Dietary inclusion of 0.6 mg/kg Macleaya cordata IA could enhance immune function of body and inhibit liver damage via inactivating TLR4/MyD88/NF-ÎşB signaling pathway in broilers

    Medical treatment and long-term outcome of chronic atrial fibrillation in the aged with chest distress: a retrospective analysis versus sinus rhythm

    Although “chest distress” is the most frequent complication in the aged with chronic atrial frbrillation (AF) in clinical practice, there are few data on the association between chronic AF and coronary artery disease (CAD) in the aged in terms of medical treatment and long-term outcome. We assessed coronary artery lesions in such patients and evaluated the efficacy of medical treatment in long-term follow-ups. Of 315 elderly patients (mean age: 77.39 ± 6.33 years) who had undergone coronary angiography for chest distress, 297 exhibited sinus rhythm (SR), whereas 18 patients exhibited chronic AF. Patients with AF were followed for 4.22 ± 2.21 years. Average diastolic blood pressure (DBP) of AF patients was observed to be markedly less than that of patients with SR (57.33 ± 6.87 mmHg vs 71.08 ± 10.54 mmHg, t-test: P < 0.01). Compared with SR patients, severe stenosis of the coronary artery in AF patients was reduced (73.06% vs 44.44%, Chi-square test: P < 0.01). AF patients with chest distress had high CHADS2 score (3.72 ± 1.27), but only 33.3% patients received oral anticoagulants, and such patients had a significantly lower rate of revascularization (21.43% vs 55.63%, Chi-square test: P < 0.01), and higher rate of all-cause death (22.22% vs 4.38%, Chi-square test: P < 0.01) and thromboembolism (16.67% vs 1.68%, Chi-square test: P < 0.01) in the long-term follow-ups compared with SR patients. Chest distress in the aged with AF was related to insufficient coronary blood supply that was primarily due to a reduced DBP rather than to occult CAD. Adequate and safe medical therapy was difficult to achieve in these patients. Such patients typically have a poor prognosis, and optimal therapeutic strategies to treat them are urgently needed

    Elevated expression of CDK4 in lung cancer

    <p/> <p>Background</p> <p>The aim of the present study was to analyze the expression of Cyclin-dependent kinase 4 (<it>CDK4</it>) in lung cancer and its correlation with clinicopathologic features. Furthermore, the involvement of <it>CDK4</it>-mediated cell cycle progression and its molecular basis were investigated in the pathogenesis of lung cancer.</p> <p>Methods</p> <p>Using immunohistochemistry analysis, we analyzed <it>CDK4 </it>protein expression in 89 clinicopathologically characterized lung cancer patients (59 males and 30 females) with ages ranging from 36 to 78 years and compared them to 23 normal lung tissues. Cases with cytoplasmic and nuclear <it>CDK4 </it>immunostaining score values greater than or equal to 7 were regarded as high expression while scores less than 7 were considered low expression. The correlation between the expression level of <it>CDK4 </it>and clinical features was analyzed. Furthermore, we used lentiviral-mediated shRNA to suppress the expression of CDK4 and investigate its function and molecular mechanism for mediating cell cycle progression.</p> <p>Results</p> <p>The expression level of <it>CDK4 </it>protein was significantly increased in lung cancer tissues compared to normal tissues (<it>P </it>< 0.001). In addition, high levels of <it>CDK4 </it>protein were positively correlated with the status of pathology classification (<it>P </it>= 0.047), lymph node metastasis (<it>P </it>= 0.007), and clinical stage (<it>P </it>= 0.004) of lung cancer patients. Patients with higher <it>CDK4 </it>expression had a markedly shorter overall survival time than patients with low <it>CDK4 </it>expression. Multivariate analysis suggested the level of <it>CDK4 </it>expression was an independent prognostic indicator (<it>P </it>< 0.001) for the survival of patients with lung cancer. Use of lentiviral-mediated shRNA to inhibit the expression of <it>CDK4 </it>in lung cancer cell line A549 not only inhibited cell cycle progression, but also dramatically suppressed cell proliferation, colony formation, and migration. Furthermore, suppressing <it>CDK4 </it>expression also significantly elevated the expression of cell cycle regulator <it>p21</it></p> <p>Conclusion</p> <p>Overexpressed <it>CDK4 </it>is a potential unfavorable prognostic factor and mediates cell cycle progression by regulating the expression of <it>p21 </it>in lung cancer</p

    Analysis of single-cell RNAseq identifies transitional states of T cells associated with hepatocellular carcinoma

    BACKGROUND: Exhausted T cells and regulatory T cells (Tregs) comprise diverse subsets of tumor immunosuppressive microenvironment that play key roles in tumor progress. Understanding subset diversity in T cells is a critical question for developing cancer immunotherapy. METHODS: A total of 235 specimens from surgical resections of hepatocellular carcinoma (HCC) patients were examined for infiltration of exhausted T cell (Tex) in tumor and adjacent tissue. We conducted deep single-cell targeted immune profiling on CD3 RESULTS: We observed transitional differentiation of exhausted CD8 CONCLUSIONS: T cell exhaustion is a progressive process, and the gene-expression profiling displayed T cell exhaustion and anergy are different. Accordingly, it is possible that functional exhaustion is caused by the combination effects of passive defects and overactivation in stress response. The results help to understand the dynamic framework of T cells function in cancer which is important for designing rational cancer immunotherapies

    Effect of replacing dicalcium phosphate with mono-dicalcium phosphate to supplement phosphorus on laying performance, phosphorus-calcium metabolism and bone metabolism of aged laying hens

    The objective of this study was to evaluate the effect of replacing dicalcium phosphate (DCP) with mono-dicalcium phosphate (MDCP) to formulate low-phosphorus (P) diets on laying performance, egg quality, phosphorus-calcium metabolism, and bone metabolism of 69–78-week-old aged laying hens. Hy-Line Brown laying hens (n = 1,350, 69 weeks old) were randomly assigned to six treatments, each with five replicates of 45 hens. A corn–soybean meal–based diet was formulated to contain 0.12% non-phytate phosphorus (NPP), 3.81% calcium (Ca), and 1,470 FTU/kg phytase. The control group (CON) was supplemented with DCP inorganic phosphorus (Pi) at the NPP level of 0.20% (dietary NPP levels of 0.32%). Test groups (T1–T5) were supplemented with MDCP Pi at NPP levels of 0.07%, 0.11%, 0.15%, 0.18, and 0.20% (dietary NPP levels of 0.19, 0.23, 0.27, 0.30, and 0.32%, respectively). Calcium carbonate levels were adjusted to ensure all experimental diets contained the same Ca levels (3.81%). The feeding trial lasted 10 weeks, with hens increasing in age from 69 to 78 weeks. When supplemented with 1,470 FTU/kg phytase, extra DCP Pi or MDCP Pi did not affect (p &gt; 0.05) laying performance (day laying rate, average egg weight, feed intake, feed-to-egg mass ratio, broken egg rate), egg quality (eggshell strength, albumen height, haugh units), or serum P, Ca, copper (Cu), iron (Fe), zinc (Zn), and manganese (Mn) levels. However, when laying hens were fed MDCP Pi (NPP levels of 0.07 to 0.20%), yolk color improved (p = 0.0148). The tibia breaking strength was significantly higher (p &lt; 0.05) in the 0.18 and 0.20% NPP MDCP Pi groups than in the 0.20% NPP DCP Pi group. The breaking strength, Ca content, and P content of tibia in 0.11% and 0.15% NPP MDCP Pi hens were not significantly (p &gt; 0.05) different from those in 0.20% NPP DCP Pi hens. Hens fed 0.07% NPP MDCP Pi had higher (p &lt; 0.01) serum levels of osteoprotegerin (OPG), type-I collagen c-telopeptide (CTX-I), and tartrate-resistant acid phosphatase 5b (TRACP-5b) than those in all other groups. Serum levels of TRACP-5b and CTX-I in the 0.11% and 0.15% NPP MDCP Pi group were significantly lower than those in 0.18 and 0.20% NPP MDCP Pi groups and the 0.20% NPP DCP Pi group (p &lt; 0.0001). Hens fed 0.07% and 0.11% NPP MDCP Pi had higher (p &lt; 0.05) serum levels of parathyroid hormone (PTH) than those in all other groups. No differences were detected in serum calcitonin (CT), 1,25-dihydroxy-vitamin D3 (1,25-(OH)2D3), bone alkaline phosphatase (BAP), osteocalcin(OCN), and osteopontin (OPN) among all groups (p &gt; 0.05). The expression of P transporters type IIa Na/Pi cotransporter (NaPi-IIa) in 0.11% and 0.15% NPP MDCP Pi hens were higher than those in 0.20% NPP MDCP Pi group and 0.20% NPP DCP Pi group (p &lt; 0.05). The results indicated that both renal P reabsorption and bone resorption were involved in adapting to a low-P diet. In summary, when MDCP was used instead of DCP to supplement P, NPP levels could be reduced to 0.11% (dietary NPP level of 0.23%) without negative effects on laying performance and skeletal health of aged hens. In addition, MDCP was more beneficial than DCP for tibia quality. The results of the current study would provide references for the application of MDCP in low-P diets of aged laying hens

    Effects of different wheat bran fermentation sources on growth performance, nutrient digestibility, serum antioxidant capacity and fecal microbiota in growing pigs

    The present study aimed to evaluate the application of different wheat bran fermentation sources in growing pigs. A total of 320 pigs (43 ± 0.21 kg), were randomly allocated to 5 groups in a 21-d trial. The control group was fed a basal diet (CON) containing raw wheat bran, and the other four treatments were fed the diets in which the raw wheat bran in the basal diet was substituted with Aspergillus niger (WBA), Bacillus licheniformis (WBB), Candida utilis (WBC), and Lactobacillus plantarum (WBL) fermented wheat bran, respectively. The results showed that compared to the CON group, the crude fiber and pH values were decreased (p &lt; 0.05), while the gross energy (GE), crude protein (CP), and lactic acid values were increased (p &lt; 0.05) in all the wheat bran fermented by different strains. Compared with other treatments, feeding B. licheniformis fermented wheat bran had higher final weight, average daily gain, as well as lower feed-to-gain ratio. Compared with CON group, pigs fed with fermented wheat bran diets had higher dry matter, CP, and GE availability, serum total protein, albumin and superoxide dismutase levels, and fecal Lactobacillus counts, as well as lower malondialdehyde level and fecal Escherichia coli count. Collectively, our findings suggested that feeding fermented wheat bran, especially B. licheniformis fermented wheat bran, showed beneficial effects on the growth performance, nutrient digestibility, serum antioxidant capacity, and the gut microbiota structure of growing pigs

    Macleaya cordata isoquinoline alkaloids attenuate Escherichia coli lipopolysaccharide-induced intestinal epithelium injury in broiler chickens by co-regulating the TLR4/MyD88/NF-ÎşB and Nrf2 signaling pathways

    This study sought to explore the effects and potential mechanisms of dietary supplementation with isoquinoline alkaloids (IA) from Macleaya cordata to alleviate lipopolysaccharide (LPS)-induced intestinal epithelium injury in broilers. A total of 486 1-day-old broilers were assigned at random to a control (CON) group, LPS group, and LPS+IA group in a 21-d study. The CON and LPS groups received a basal diet, while the LPS+IA group received a basal diet supplemented with 0.6 mg/kg IA. At 17, 19, and 21 days of age, the LPS and LPS+BP groups were injected intraperitoneally with LPS, and the CON group was intraperitoneally injected equivalent amount of saline solution. The results manifested that LPS injection caused intestinal inflammation and lipid peroxidation, disrupted intestinal barrier and function, and increased the abundance of harmful microorganisms. However, dietary IA supplementation alleviated LPS-induced adverse changes in intestinal morphology, apoptosis, mucosal barrier integrity, cecum microorganisms, and homeostasis disorder by decreasing inflammatory cytokines and enhancing antioxidant-related genes expressions; inhibited LPS-induced increases in TLR4 and NF-ÎşB expressions and decreases in Nrf2 and GPX1 genes expressions. Our findings indicated that Macleaya cordata IA addition attenuated LPS-induced intestinal epithelium injury and disorder of intestinal homeostasis by enhancing the anti-inflammatory and antioxidant capacity of broiler chickens possibly via co-regulating TLR4/MyD88/NF-ÎşB and Nrf2 signaling pathways
