    p53 Family: Role of Protein Isoforms in Human Cancer

    TP53, TP63, and TP73 genes comprise the p53 family. Each gene produces protein isoforms through multiple mechanisms including extensive alternative mRNA splicing. Accumulating evidence shows that these isoforms play a critical role in the regulation of many biological processes in normal cells. Their abnormal expression contributes to tumorigenesis and has a profound effect on tumor response to curative therapy.     Understanding Contextual Differences in Residential LTC Provision for Cross-National Research : Identifying Internationally Relevant CDEs

    Long-term care (LTC) reflects a growing emphasis on person-centered care (PCC), with services oriented around individuals' needs and preferences. Addressing contextual and cultural differences across countries offers important insight into factors that facilitate or hinder application of PCC practices within and across countries. This article takes an international lens to consider country-specific contexts of LTC, describing preliminary steps to develop common data elements that capture contextual differences across LTC settings globally. Through an iterative series of online, telephone, and in-person sessions, we engaged in in-depth discussions with 11 colleague experts in residential LTC and coauthors from six countries (China and Hong Kong, England, Sweden, Thailand, Trinidad and Tobago, and the United States). Our discussions yielded rich narrative describing a vast range in types of LTC settings, leading to our development of a working definition of residential LTC. Scope of services, funding, ownership, and regulations varied greatly across countries and across different residential LTC settings within countries. Moving forward, we recommend expanding our activities to countries that reflect different stages of residential LTC development. Our goal is to contribute to a larger initiative underway by the WE-THRIVE consortium to establish a global research measurement infrastructure that advances PCC internationally.Peer reviewe

    Analysis of the crystallization process of a biopharmaceutical compound in the presence of impurities using process analytical technology (PAT) tools

    BACKGROUND. The crystallization of biopharmaceuticals can be problematic since, because the biosynthesis of these compounds is very difficult to control, they can contain a significant amount of impurities that have to be eliminated. In fact, impurities can lead to changes in the properties of the drug that can significantly reduce its effectiveness or even put the user in danger. The aim of this work is to exploit process analytical technology (PAT) tools to study the crystallization step of vitamin B12 crude product extracted via fermentation. RESULTS. Linear cooling crystallization experiments were performed using different conditions. The effects of solvent, cooling rate, seeding and purity of the initial material on the final size distribution and purity of the crystals were investigated through the use of UV/Vis spectroscopy, focused beam reflectance measurement (FBRM) and the CryPRINS software (Crystallization Process Informatics System). It was found that impurities strongly inhibit the growth of vitamin B12 crystals, promoting nucleation and leading to poor final crystal size distribution. CONCLUSION. Slow cooling can help to increase the purity of the final product but also generates a broad crystal size distribution because of secondary nucleation. Preparing the solution with material already crystallized once and using purified seeds helped to obtain a narrower crystal size distribution and also to reduce breakage

    Improving skills in rounding off the whole number

    This study was conducted to address teaching and learning skills in rounding off a whole number. This study consisted of 15 years 4 students from the Kong Nan Chinese Primary School, Parit Raja, Johor, Malaysia. Initial survey to identify this problem was carried out by analyzing the exercise books and exercises in pre-test. Based on these analyses, a large number of students were not proficient in relevant skills. A ‘q’ technique was introduced as an approach in teaching and learning to help students master the skills of rounding whole numbers. In summary, this technique helps students to remember the sequence of processes and process in rounding numbers. A total of four sessions of teaching and learning activities that take less than an hour have been implemented specifically to help students to master this technique. Results of the implementation of these activities have shown very positive results among the students. Two post tests were carried out to see the effectiveness of techniques and the results shows that 100% of students were able to answer correctly at least three questions correctly. The t-test analysis was clearly showed the effectiveness of ‘q’ technique. This technique also indirectly helps to maintain and increase student interest in learning Mathematics. This is shown with the active involvement of students in answering questions given by the teacher

    Exploring the Effect of the Number of Hydrogen Atoms on the Properties of Lanthanide Hydrides by DMFT

    Lanthanide hydrogen-rich materials have long been considered as one of the candidates with high-temperature superconducting properties in condensed matter physics, and have attracted great interest. Attempts to investigate the effects of different compositions of lanthanide hydrogen-rich materials are ongoing, with predictions and experimental studies in recent years having shown that substances such as LaH 10 , CeH 9 , and LaH 16 exhibit extremely high superconducting temperatures between 150-250 GPa. In particular, researchers have noted that in those materials an increase in the f character at the Fermi level leads to an increase in the superconducting temperature. Here, we further elaborate on the effect of the ratios of lanthanide to hydrogen in these substances with the aim to bring more clarity to the study of superhydrides in these extreme cases by comparing a variety of lanthanide hydrogen-rich materials with different ratios using the DMFT method, and provide ideas for later structural predictions and material property studies.Comment: 7pages, 5figure

    Hexameric procyanidins inhibit colorectal cancer cell growth through both redox and non-redox regulation of the epidermal growth factor signaling pathway

    Dietary proanthocyanidins (PAC) consumption is associated with a decreased risk for colorectal cancer (CRC). Dysregulation of the epidermal growth factor (EGF) receptor (EGFR) signaling pathway is frequent in human cancers, including CRC. We previously showed that hexameric PAC (Hex) exert anti-proliferative and pro-apoptotic actions in human CRC cells. This work investigated if Hex could exert anti-CRC effects through its capacity to regulate the EGFR pathway. In proliferating Caco-2 cells, Hex acted attenuating EGF-induced EGFR dimerization and NADPH oxidase-dependent phosphorylation at Tyr 1068, decreasing EGFR location at lipid rafts, and inhibiting the downstream activation of pro-proliferative and anti-apoptotic pathways, i.e. Raf/MEK/ERK1/2 and PI3K/Akt. Hex also promoted EGFR internalization both in the absence and presence of EGF. While Hex decreased EGFR phosphorylation at Tyr 1068, it increased EGFR Tyr 1045 phosphorylation. The latter provides a docking site for the ubiquitin ligase c-Cbl and promotes EGFR degradation by lysosomes. Importantly, Hex acted synergistically with the EGFR-targeted chemotherapeutic drug Erlotinib, both in their capacity to decrease EGFR phosphorylation and inhibit cell growth. Thus, dietary PAC could exert anti-CRC actions by modulating, through both redox- and non-redox-regulated mechanisms, the EGFR pro-oncogenic signaling pathway.     Emergence of Symmetry in Complex Networks

    Many real networks have been found to have a rich degree of symmetry, which is a very important structural property of complex network, yet has been rarely studied so far. And where does symmetry comes from has not been explained. To explore the mechanism underlying symmetry of the networks, we studied statistics of certain local symmetric motifs, such as symmetric bicliques and generalized symmetric bicliques, which contribute to local symmetry of networks. We found that symmetry of complex networks is a consequence of similar linkage pattern, which means that nodes with similar degree tend to share similar linkage targets. A improved version of BA model integrating similar linkage pattern successfully reproduces the symmetry of real networks, indicating that similar linkage pattern is the underlying ingredient that responsible for the emergence of the symmetry in complex networks.Comment: 7 pages, 7 figure
