Ceramic Consumption in a Boston Immigrant Tenement
In the late nineteenth and early twentieth centuries, Boston’s North End became home to thousands of European immigrants, mostly from Ireland and Italy. The majority of these immigrant families lived in crowded tenement apartments and earned their wages from low-paying jobs such as manual laborers or store clerks. The Ebenezer Clough House at 21 Unity Street was originally built as a single-family colonial home in the early eighteenth century but was later repurposed as a tenement in the nineteenth century. In 2013, the City of Boston Archaeology Program excavated the rear lot of the Clough House, recovering 36,465 artifacts, including 4,298 ceramic sherds, across 14 site-wide contexts. One context, the main midden, has been interpreted as a multi-use household trash deposit dating from the 1870s to the 1910s, during which the tenement was home to a rotation of over 100 working-class families, most of them immigrants. This project couples ceramic analysis with in-depth archival research to illuminate the consumption strategies of Boston’s immigrant working class. I conclude that tenants primarily used decorated but mismatched and older ceramic ware types, valuing thrift and prioritizing family needs while consuming differently than their middle-class counterparts
Methanogen activity and microbial diversity in Gulf of Cádiz mud volcano sediments
The Gulf of Cádiz is a tectonically active continental margin with over sixty mud volcanoes (MV) documented, some associated with active methane (CH4) seepage. However, the role of prokaryotes in influencing this CH4 release is largely unknown. In two expeditions (MSM1-3 and JC10) seven Gulf of Cádiz MVs (Porto, Bonjardim, Carlos Ribeiro, Captain Arutyunov, Darwin, Meknes, and Mercator) were analyzed for microbial diversity, geochemistry, and methanogenic activity, plus substrate amended slurries also measured potential methanogenesis and anaerobic oxidation of methane (AOM). Prokaryotic populations and activities were variable in these MV sediments reflecting the geochemical heterogeneity within and between them. There were also marked differences between many MV and their reference sites. Overall direct cell numbers below the SMTZ (0.2–0.5 mbsf) were much lower than the general global depth distribution and equivalent to cell numbers from below 100 mbsf. Methanogenesis from methyl compounds, especially methylamine, were much higher than the usually dominant substrates H2/CO2 or acetate. Also, CH4 production occurred in 50% of methylated substrate slurries and only methylotrophic CH4 production occurred at all seven MV sites. These slurries were dominated by Methanococcoides methanogens (resulting in pure cultures), and prokaryotes found in other MV sediments. AOM occurred in some slurries, particularly, those from Captain Arutyunov, Mercator and Carlos Ribeiro MVs. Archaeal diversity at MV sites showed the presence of both methanogens and ANME (Methanosarcinales, Methanococcoides, and ANME-1) related sequences, and bacterial diversity was higher than archaeal diversity, dominated by members of the Atribacterota, Chloroflexota, Pseudomonadota, Planctomycetota, Bacillota, and Ca. “Aminicenantes.” Further work is essential to determine the full contribution of Gulf of Cádiz mud volcanoes to the global methane and carbon cycles
Development of High-Purity Optical Grade Single-Crystal CVD Diamond for Intracavity Cooling
Microwave assisted chemical vapour deposited bulk diamond products have been used in a range of high power laser systems, due to low absorption across a range of wavelengths and exceptional thermal properties. However the application of polycrystalline products has frequently been limited to applications at longer wavelengths or thermal uses outside of the optical path due to the birefringence and scatter that are intrinsic properties of the polycrystalline materials. However, there are some solid state structures, including thin disc gain modules and amplifiers, that will gain significantly in terms of potential output powers if diamond could be used as a heat spreader in the optical path as well as a heat spreader on the rear surface of the disk. Therefore single crystal grades of diamond have been developed that overcome the limitations of the polycrystalline material, with low absorption, low scatter and low birefringence grades for demanding optical applications. We will present new data, characterising the performance of these materials across infra-red and visible wavelengths with absorption coefficient measured by laser calorimetry at a range of wavelengths from 1064 nm to 452 nm
Fine Structure in the Circumstellar Environment of a Young, Solar-like Star: the Unique Eclipses of KH 15D
Results of an international campaign to photometrically monitor the unique
pre-main sequence eclipsing object KH 15D are reported. An updated ephemeris
for the eclipse is derived that incorporates a slightly revised period of 48.36
d. There is some evidence that the orbital period is actually twice that value,
with two eclipses occurring per cycle. The extraordinary depth (~3.5 mag) and
duration (~18 days) of the eclipse indicate that it is caused by circumstellar
matter, presumably the inner portion of a disk. The eclipse has continued to
lengthen with time and the central brightness reversals are not as extreme as
they once were. V-R and V-I colors indicate that the system is slightly bluer
near minimum light. Ingress and egress are remarkably well modeled by the
passage of a knife-edge across a limb-darkened star. Possible models for the

Rescue of the MERTK phagocytic defect in a human iPSC disease model using translational read-through inducing drugs.
Inherited retinal dystrophies are an important cause of blindness, for which currently there are no effective treatments. In order to study this heterogeneous group of diseases, adequate disease models are required in order to better understand pathology and to test potential therapies. Induced pluripotent stem cells offer a new way to recapitulate patient specific diseases in vitro, providing an almost limitless amount of material to study. We used fibroblast-derived induced pluripotent stem cells to generate retinal pigment epithelium (RPE) from an individual suffering from retinitis pigmentosa associated with biallelic variants in MERTK. MERTK has an essential role in phagocytosis, one of the major functions of the RPE. The MERTK deficiency in this individual results from a nonsense variant and so the MERTK-RPE cells were subsequently treated with two translational readthrough inducing drugs (G418 & PTC124) to investigate potential restoration of expression of the affected gene and production of a full-length protein. The data show that PTC124 was able to reinstate phagocytosis of labeled photoreceptor outer segments at a reduced, but significant level. These findings represent a confirmation of the usefulness of iPSC derived disease specific models in investigating the pathogenesis and screening potential treatments for these rare blinding disorders
Anthropogenic supply of nutrients in a wildlife reserve may compromise conservation success
In nutrient-poor wildlife reserves it has become common-practice to provide supplemental mineral resources for wildlife. Yet, the impacts of anthropogenic mineral supplementation on large herbivore nutrition, behaviour, and subsequent impact on ecosystem processes have received little attention. Here, we examine the contribution of anthropogenic mineral lick provision to wildlife nutrient intake across a community of mammalian herbivores (>10 kg) in the southern Kalahari Desert. Based on predicted daily nutrient intake and a faecal nutrient assessment, many large herbivore species appear deficient in phosphorus (P), sodium (Na), or zinc (Zn). For these nutrients, anthropogenic salt and mineral licks constitute an important source of nutrient intake helping to reduce or overcome requirement deficits. Larger-bodied species disproportionately consumed licks, acquiring more nutritional benefits. A comprehensive assessment of animal body condition indicated that, in general, large herbivores display good health. However, bulk grazers, non-ruminants and females displayed poorer body condition. We discuss how provisioning of anthropogenic mineral licks may inflate large herbivore populations beyond the long-term carrying capacity of the reserve by decoupling wildlife fecundity from nutrient-related feedbacks on population growth. Over time, this could compromise ecosystem integrity through habitat degradation, modified species interactions and trophic cascades. Based on results presented here, it is clear that anthropogenic provisioning of mineral licks should be considered cautiously by wildlife managers aiming to conserve natural processes in landscapes
Biostimulation of jarosite and iron oxide-bearing mine waste enhances subsequent metal recovery
Novel resource recovery technologies are required for metals-bearing hazardous wastes in order to achieve circular economy outcomes and industrial symbiosis. Iron oxide and co-occurring hydroxysulphate-bearing wastes are globally abundant and often contain other elements of value. This work addresses the biostimulation of indigenous microbial communities within an iron oxide/ hydroxysulphate-bearing waste and its effect on the subsequent recoverability of metals by hydrochloric, sulphuric, citric acids, and EDTA. Laboratory-scale flow-through column reactors were used to examine the effect of using glycerol (10% w/w) to stimulate the in situ microbial community in an iron oxide/ hydroxysulphate-bearing mine waste. The effects on the evolution of leachate chemistry, changes in microbiological community, and subsequent hydrometallurgical extractability of metals were studied. Results demonstrated increased leachability and selectivity of Pb, Cu, and Zn relative to iron after biostimulation with a total of 0.027kg of glycerol per kg of waste. Biostimulation, which can be readily applied in situ, potentially opens new routes to metal recovery from globally abundant waste streams that contain jarosite and iron oxides
CRLX101 nanoparticles localize in human tumors and not in adjacent, nonneoplastic tissue after intravenous dosing
Nanoparticle-based therapeutics are being used to treat patients with solid tumors. Whereas nanoparticles have been shown to preferentially accumulate in solid tumors of animal models, there is little evidence to prove that intact nanoparticles localize to solid tumors of humans when systemically administered. Here, tumor and adjacent, nonneoplastic tissue biopsies are obtained through endoscopic capture from patients with gastric, gastroesophageal, or esophageal cancer who are administered the nanoparticle CRLX101. Both the pre- and postdosing tissue samples adjacent to tumors show no definitive evidence of either the nanoparticle or its drug payload (camptothecin, CPT) contained within the nanoparticle. Similar results are obtained from the predosing tumor samples. However, in nine of nine patients that were evaluated, CPT is detected in the tumor tissue collected 24–48 h after CRLX101 administration. For five of these patients, evidence of the intact deposition of CRLX101 nanoparticles in the tumor tissue is obtained. Indications of CPT pharmacodynamics from tumor biomarkers such as carbonic anhydrase IX and topoisomerase I by immunohistochemistry show clear evidence of biological activity from the delivered CPT in the posttreatment tumors
Pyrotechnic Actuator for Retracting Tubes Between MSL Subsystems
An apparatus, denoted the "retractuator" (a contraction of "retracting actuator"), was designed to help ensure clean separation between the cruise stage and the entry-vehicle subsystem of the Mars Science Laboratory (MSL) mission. The retractuator or an equivalent mechanism is needed because of tubes that (1) transport a heat-transfer fluid between the stages during flight and (2) are cut immediately prior to separation of the stages retractuator. The role of the retractuator is to retract the tubes, after they are cut and before separation of the subsystem, so that cut ends of the tubes do not damage thermal-protection coats on the entry vehicle and do not contribute to uncertainty of drag and consequent uncertainty in separation velocity
