    Entwicklung eines Anforderungsprofils fĂĽr Kontrolleure im Rahmen des Kontrollsystems nach der EU-Verordnung 2092/91

    Ziel der Studie ist es, das Anforderungsprofil für Kontrolleure auf der Grundlage der EU-Öko-Verordnung 2092/91 für die unterschiedlichen Kontrollbereiche zu ermitteln. Grundlage dafür bilden die Analyse der Tätigkeiten von Kontrolleuren aufgegliedert nach Branchen in die verschiedenen Kontrollbereiche Landwirtschaft, Verarbeitung und Import aus Drittländern. Ergänzend wurde erfasst, welche Anforderungen in anderen Kontroll- und Qualitätssicherungsnormen und -verordnungen an das Kontroll- bzw. Auditpersonal gestellt werden. Ableitend von diesen Analysen wurde das Anforderungsprofil für die drei Kontrollbereiche erstellt. Das Anforderungsprofil gliedert sich in vier Teile: die persönliche Qualifikation (in Anlehnung an ISO 10011), die Basisqualifikation (mündliche und schriftliche Kommunikation, Kalkulation, Berufserfahrung), die fachliche Qualifikation, die sich je nach Kontrollbereich unterscheidet sowie die Kenntnisse der gesetzlichen Regelungen. Für jeden Baustein im Anforderungsprofil wurde abgeleitet, mit welcher Berufsausbildung, Trainingsmaßnahme o.ä. diese Qualifikation erreicht und wie sie nachgewiesen werden kann. In diesem Zusammenhang wurde auch das in Deutschland übliche Zulassungsverfahren für Kontrollpersonal diskutiert. Die Verordnung (EWG) 2092/91 sowie der ergänzend geltenden EN 45011 enthält keine Vorgaben, welche die Zulassung von Kontrolleuren durch Aufsichtsbehörden vor dem Einsatz erforderlich machen. Es muss allerdings im Rahmen der Überwachung der Kontrollstellen überprüft werden, ob die Kontrollstellen über sachgerechte Personalkriterien, ein angemessenes dokumentiertes Verfahren für die Auswahl des Personals sowie individuelle Leistungsbeurteilungen verfügen und das Personal angemessen aus- und fortbilden. Eine Umfrage bei den Kontrollstellen im deutschsprachigen Raum erfasste die derzeitigen Aus- und Fortbildungsmaßnahmen für Kontrollpersonal. Auf Grundlage der Auswertung der Tätigkeitsanalysen und der Rückmeldungen der Kontrollstellen wurde festgestellt, dass Bedarf an einem kontrollstellenüber-greifenden Fortbildungsangebot besteht, das sich vor allem auf die Spezialbereiche im Kontrollsystem konzentriert (z.B. Umgang mit schwierigen Kontrollsituationen, Aktuelle Änderungen der VO 2092/91, Hofverarbeitung, Rückstandsanalytik und Probenahmeverfahren, Importkontrolle)

Organometallics. 1992;11(7):2351-2353.The diphosphenyl complex (eta-5-C5Me5)-(CO)2Fe-P=P-Mes* (Mes* = 2,4,6-tBu3C6H2) undergoes a [3 + 2] dipolar cycloaddition with hexafluoroacetone to give the metalla heterocycle (eta-5-C5Me5)(CO)-Fe-P(=PMes*)OC(CF3)2C(O) with a remarkably short Fe-P bond (2.084 (4) angstrom) and an exocyclic P=P bond. When stored in solution at -40-degrees-C, this complex partly rearranges to the metalated 1-oxa-2,3-diphosphetane (eta-5-C5Me5)(CO)2Fe-P-P(Mes*)OC(CF3)2.     Assessment of Confounding Factors Affecting the Tumor Markers SMRP, CA125, and CYFRA21-1 in Serum

    The purpose of this analysis was to evaluate if serum levels of potential tumor markers for the diagnosis of malignant mesothelioma and lung cancer are affected by confounding factors in a surveillance cohort of workers formerly exposed to asbestos. SMRP, CA125, and CYFRA21-1 concentrations were determined in about 1,700 serum samples from 627 workers formerly exposed to asbestos. The impact of factors that could modify the concentrations of the tumor markers was examined with linear mixed models. SMRP values increased with age 1.02-fold (95% CI 1.01–1.03) and serum creatinine concentration 1.32-fold (95% CI 1.20–1.45). Levels differed by study centers and were higher after 40 years of asbestos exposure. CA125 levels increased with longer storage of the samples. CYFRA21-1 values correlated with age 1.02-fold (95% CI 1.01–1.02), serum creatinine 1.21-fold (95% CI 1.14–1.30) and varied by study centers due to differences in sample handling. Tumor marker concentrations are influenced by subject-related factors, sample handling, and storage. These factors need to be taken into account in screening routine

In this study we evaluate the use of bicinchoninic acid (BCA) as a ligand for Cu(I) under physiological reaction conditions. We demonstrate that the BCA–Cu(I)-complex represents an efficient catalyst for the conjugation of fluorophores or biotin to alkyne- or azide-functionalized proteins resulting in increased or at least equal reaction yields compared to commonly used catalysts like Cu(I) in complex with TBTA (tris[(1-benzyl-1H-1,2,3-triazol-4-yl)methyl]amine) or BPAA (bathophenanthroline disulfonic acid).     Cyanine Dye Coupling Mediates Self-assembly of a pH Sensitive Peptide into Novel 3D Architectures

    Synthetic multichromophore systems are of great importance in artificial light harvesting devices, organic optoelectronics, tumor imaging and therapy. Here, we introduce a promising strategy for the construction of self-assembled peptide templated dye stacks based on coupling of a de novo designed pH sensitive peptide with a cyanine dye Cy5 at its N-terminus. Microscopic techniques, in particular cryogenic TEM (cryo-TEM) and cryo-electron tomography technique (cryo-ET), reveal two types of highly ordered three-dimensional assembly structures on the micrometer scale. Unbranched compact layered rods are observed at pH 7.4 and two-dimensional membrane-like assemblies at pH 3.4, both species displaying spectral features of H-aggregates. Molecular dynamics simulations reveal that the coupling of Cy5 moieties promotes the formation of both ultrastructures, whereas the protonation states of acidic and basic amino acid side chains dictates their ultimate three-dimensional organization

    Regulatory T Cells and IL-10 Independently Counterregulate Cytotoxic T Lymphocyte Responses Induced by Transcutaneous Immunization

    The imidazoquinoline derivate imiquimod induces inflammatory responses and protection against transplanted tumors when applied to the skin in combination with a cognate peptide epitope (transcutaneous immunization, TCI). Here we investigated the role of regulatory T cells (T(reg)) and the suppressive cytokine IL-10 in restricting TCI-induced cytotoxic T lymphocyte (CTL) responses.TCI was performed with an ointment containing the TLR7 agonist imiquimod and a CTL epitope was applied to the depilated back skin of C57BL/6 mice. Using specific antibodies and FoxP3-diphteria toxin receptor transgenic (DEREG) mice, we interrogated inhibiting factors after TCI: by depleting FoxP3(+) regulatory T cells we found that specific CTL-responses were greatly enhanced. Beyond this, in IL-10 deficient (IL-10(-/-)) mice or after blocking of IL-10 signalling with an IL-10 receptor specific antibody, the TCI induced CTL response is greatly enhanced indicating an important role for this cytokine in TCI. However, by transfer of T(reg) in IL-10(-/-) mice and the use of B cell deficient JHT(-/-) mice, we can exclude T(reg) and B cells as source of IL-10 in the setting of TCI.We identify T(reg) and IL-10 as two important and independently acting suppressors of CTL-responses induced by transcutaneous immunization. Advanced vaccination strategies inhibiting T(reg) function and IL-10 release may lead the development of effective vaccination protocols aiming at the induction of T cell responses suitable for the prophylaxis or treatment of persistent infections or tumors

    Quality-of-life evaluations in children and adolescents with Ewing sarcoma treated with pencil-beam-scanning proton therapy.

    BACKGROUND With improved survival rates for children with cancer, quality-of-life (QoL) issues have increasingly become the focus of attention. We report the QoL of children with Ewing sarcoma (EWS) treated with pencil-beam-scanning proton therapy (PT). METHODS A PEDQOL (QoL questionnaire for children 4-18 years) self/proxy questionnaire was used to prospectively assess the QoL of 23 children <18 years with EWS treated with PT. This questionnaire evaluates eight different domains. Children (self-rating) and parents (proxy-rating) filled out the questionnaire at the start of PT (E1), 2 months after treatment (E2), and thereafter once yearly (E≥3). RESULTS Compared with healthy controls, parents rated the QoL of their children at E1 significantly worse in all but two (cognition and social functioning-family) domains. At E4, significant differences between the two groups only remained in three of eight domains. At E1, children self-rated their QoL significantly worse in the domain Physical functioning (p = .004) and significantly better in the domain Body image (p = .044) compared to healthy controls, whereas no significant differences were observed at E4. For the longitudinal comparison E1 versus E4, according to parents, Emotional functioning, Cognition and Social functioning-peers were slightly decreased 2 years after PT. The children rated Emotional functioning and Body image poorly 2 years after PT. CONCLUSIONS Children with EWS usually recovered seemingly well to normal QoL levels 2 years after the end of PT. They tended to rate their QoL substantially higher than their parents. However, in the longitudinal analysis at 2 years, children rated their Emotional functioning and Body image scores poorly
