    A MODEL OF RETAIL OUTLET SELECTION FOR BEEF

    Multinomial logit models were used to explain consumer outlet selection when buying beef, specifically roasts, steaks, ground beef, and other types of beef. Outlets were grouped into supermarkets, butchers, warehouses, supercenters, and others, and the probability of selecting each outlet type over a range of demographic and other variables was tested. The models were estimated from household data, with 198,682 observations used in the estimation. Empirical results showed that the type of beef purchased and the size of the purchase played an importance role in the choice of outlet. Furthermore, the increase in mobility seen when consumers buy larger unit cuts could not be fully explained by price discounting.     Spatial intensity distribution analysis: studies of G Protein-coupled receptor oligomerization

    Spatial intensity distribution analysis (SpIDA) is a recently developed approach for determining quaternary structure information on fluorophore-labelled proteins of interest in situ. It can be applied to live or fixed cells and native tissue. Using confocal images, SpIDA generates fluorescence intensity histograms that are analysed by super-Poissonian distribution functions to obtain density and quantal brightness values of the fluorophore-labelled protein of interest. This allows both expression level and oligomerisation state of the protein to be determined. We describe the application of SpIDA to investigate the oligomeric state of G protein-coupled receptors (GPCRs) at steady state and following cellular challenge, and consider how SpIDA may be used to explore GPCR quaternary organisation in pathophysiology and to stratify medicines

    The Correlation Between Childhood Sexual Abuse, Intimate Partner Violence, and Sexual Revictimization on North America Emerging Adult Females\u27 romantic Relationship Satisfaction with Spiritual Well-Being and Post-Traumatic Growth as Mediators

    Problem Emerging adulthood is a time of great adjustment, in both personal and relational terms. One of emerging adulthood\u27s most important developmental tasks includes the excitement of intimate romantic relationships (Shulman & Connolly, 2013). These intimate romantic relationships can involve sexual as well as emotional intimacy (Fisher, 2006). When there has been sexual abuse in childhood, the perception of self and others is maladaptive and damaging, increasing one\u27s chance of experiencing sexual revictimization and intimate partner violence (Williams et al., 2010). This can greatly influence romantic relationship satisfaction. There are very few references to strength- based trauma mediators in the current literature. This study aims to determine the relationships between childhood sexual abuse (CSA) and romantic relationship satisfaction, with the additional power of post-traumatic growth and spiritual well-being as mediators. Method This was a quantitative non-experimental study that used descriptive statistics and structural equation modeling to answer the research questions. Data was collected by QuestionPro, with a total of 406 participants as the sample size. Participants completed a survey that measured CSA history, romantic relationship satisfaction, emerging adulthood sexual revictimization, intimate partner violence, post-traumatic growth, and spiritual well-being in emerging adult females. Results In general, most participants experienced moderate levels of relationship satisfaction, although those with a history of childhood sexual abuse reported statistically lesser satisfaction levels than those who do not have a CSA history. Regardless of CSA history, participants experienced high levels of love for their partner. Approximately half of this study\u27s participants have experienced some form of sexual revictimization since they were 14 years old, and another estimate of 25% have experienced sexual revictimization in the last 12 months. Those with a CSA history had statistically significant higher prevalence rates for sexual revictimization than those who did not have a childhood sexual abuse history. Having a history of CSA did not predict relationship satisfaction directly. Intimate partner violence was shown to be the strongest predictor of relationship satisfaction, with post-traumatic growth being the next strongest predictor. Revictimization experiences had a significant direct effect on spiritual well-being. Conclusions This study was a quantitative non-experimental design. Participants were United States female emerging adults. Results showed that CSA experiences did not predict relationship satisfaction directly in female emerging adults. Females with CSA experiences reported having a significantly higher rate of sexual revictimization than those who do not have a CSA history. Finally, the best predictors of relationship satisfaction levels were intimate partner violence and post-traumatic growth

    G protein-coupled receptors (GPCRs) have been classically described as monomeric entities that function by binding in a 1:1 stoichiometric ratio to both ligand and downstream signalling proteins. However, in recent years, a growing number of studies has supported the hypothesis that these receptors can interact to form dimers and higher order oligomers although the molecular basis for these interactions, the overall quaternary arrangements and the functional importance of GPCR oligomerization remain topics of intense speculation. Muscarinic acetylcholine receptors belong to class A of the GPCR family. Each muscarinic receptor subtype has its own particular distribution throughout the central and peripheral nervous systems. In the central nervous system, muscarinic receptors regulate several sensory, cognitive, and motor functions while, in the peripheral nervous system, they are involved in the regulation of heart rate, stimulation of glandular secretion and smooth muscle contraction. Muscarinic acetylcholine receptors have long been used as a model for the study of GPCR structure and function and to address aspects of GPCR dimerization using a broad range of approaches. In this review, the prevailing knowledge regarding the quaternary arrangement for the various muscarinic acetylcholine receptors has been summarized by discussing work ranging from initial results obtained using more traditional biochemical approaches to those generated with more modern biophysical techniques

    Dynamic regulation of quaternary organization of the M1 muscarinic receptor by subtype-selective antagonist drugs

    Although rhodopsin-like G protein-coupled receptors can exist as both monomers and non-covalently associated dimers/oligomers, the steady-state proportion of each form and whether this is regulated by receptor ligands is unknown. Herein we address these topics for the M1 muscarinic acetylcholine receptor, a key molecular target for novel cognition enhancers, by employing Spatial Intensity Distribution Analysis. This method can measure fluorescent particle concentration and assess oligomerization states of proteins within defined regions of living cells. Imaging and analysis of the basolateral surface of cells expressing some 50 molecules.microm-2 of the human muscarinic M1 receptor identified an ~75/25 mixture of receptor monomers and dimers/oligomers. Both sustained and shorter-term treatment with the selective M1 antagonist pirenzepine resulted in a large shift in the distribution of receptor species to favor the dimeric/oligomeric state. Although sustained treatment with pirenzepine also resulted in marked upregulation of the receptor, simple mass-action effects were not the basis for ligand-induced stabilization of receptor dimers/oligomers. The related antagonist telenzepine also produced stabilization and enrichment of the M1 receptor dimer population but the receptor subtype non-selective antagonists atropine and N-methylscopolamine did not. In contrast, neither pirenzepine nor telenzepine altered the quaternary organization of the related M3 muscarinic receptor. These data provide unique insights into the selective capacity of receptor ligands to promote and/or stabilize receptor dimers/oligomers and demonstrate that the dynamics of ligand regulation of the quaternary organization of G protein-coupled receptors is markedly more complex than previously appreciated. This may have major implications for receptor function and behavior

    Large-Scale Goodness Polarity Lexicons for Community Question Answering

    We transfer a key idea from the field of sentiment analysis to a new domain: community question answering (cQA). The cQA task we are interested in is the following: given a question and a thread of comments, we want to re-rank the comments so that the ones that are good answers to the question would be ranked higher than the bad ones. We notice that good vs. bad comments use specific vocabulary and that one can often predict the goodness/badness of a comment even ignoring the question, based on the comment contents only. This leads us to the idea to build a good/bad polarity lexicon as an analogy to the positive/negative sentiment polarity lexicons, commonly used in sentiment analysis. In particular, we use pointwise mutual information in order to build large-scale goodness polarity lexicons in a semi-supervised manner starting with a small number of initial seeds. Despite a general perception that GPCRs exist as monomers an extensive literature has examined whether GPCRs can also form dimers and even higher-order oligomers, and if such organization influences various aspects of GPCR function, including cellular trafficking, ligand binding, G protein coupling and signalling. Here we focus on recent studies that employ approaches ranging from computational methods to single molecule tracking and both quantal brightness and fluorescence fluctuation measurements to assess the organization, stability and potential functional significance of dimers and oligomers within the class A, rhodopsin-like GPCR family

    ‘Being there’ is what matters:Methodological and ethical challenges when undertaking research on the outdoor environment with older people during and beyond the COVID-19 pandemic

    This paper reflects on adapting research methods and processes during the COVID-19 pandemic, drawing on our experiences of conducting research on the outdoor environment with older people (aged 50+) living in Scotland. First, we discuss the challenges to the organisation of research experienced in the context of changing government and university guidelines and managing delays to planned research timelines. The shift toward remote methods stimulated by the pandemic transformed traditional notions of the research field. We consider some of the implications of this for outdoor environment research, grounded as it is on exploring the interaction between people and the places they are embedded within. Further, despite a growth of literature highlighting the benefits of remote research, we found uses for digital and online approaches limited when working with older people. Second, we reflect on whether research with older people in the context of a pandemic can be conducted ethically. Drawing on our research we describe how developing an ‘ethics of care’ included negotiating with formal ethics processes but also the relational, situated ethics of qualitative health research that, because of the pandemic, had begun to shift in new ways. We describe the often intangible impacts of COVID-19 such as social isolation and bereavement that we uncovered as researchers entering into the lives of older people. In closing, we outline some of the key lessons learnt from conducting research on outdoor environments with older people to enable future qualitative health research during and beyond the pandemic

    A molecular basis for selective antagonist destabilization of dopamine D3 receptor quaternary organization

    The dopamine D3 receptor (D3R) is a molecular target for both first-generation and several recently-developed antipsychotic agents. Following stable expression of this mEGFP-tagged receptor, Spatial Intensity Distribution Analysis indicated that a substantial proportion of the receptor was present within dimeric/oligomeric complexes and that increased expression levels of the receptor favored a greater dimer to monomer ratio. Addition of the antipsychotics, spiperone or haloperidol, resulted in re-organization of D3R quaternary structure to promote monomerization. This action was dependent on ligand concentration and reversed upon drug washout. By contrast, a number of other antagonists with high affinity at the D3R, did not alter the dimer/monomer ratio. Molecular dynamics simulations following docking of each of the ligands into a model of the D3R derived from the available atomic level structure, and comparisons to the receptor in the absence of ligand, were undertaken. They showed that, in contrast to the other antagonists, spiperone and haloperidol respectively increased the atomic distance between reference α carbon atoms of transmembrane domains IV and V and I and II, both of which provide key interfaces for D3R dimerization. These results offer a molecular explanation for the distinctive ability of spiperone and haloperidol to disrupt D3R dimerization
