Effects of single mutations on the stability of horseradish peroxidase to hydrogen peroxide
Horseradish peroxidase (HRP) is a commonly used enzyme in many biotechnological fields. Improvement of HRP stability would further increase its potential application range. In the present study, 13 single- and three double-mutants of solvent exposed, proximal lysine and glutamic acid residues were analysed for enhanced H2O2 stability. Additionally, five single- and one pentuple-consensus mutants were investigated. Most mutants displayed little or no alteration in H2O2 stability; however, three (K232N, K241F and T110V) exhibited significantly increased H2O2 tolerances of 25- (T110V), 18- (K232N), and 12-fold (K241F). This improved stability may be due to an altered enzyme-H2O2 catalysis pathway or to removal of potentially oxidisable residues
Algorithms for optimizing drug therapy
BACKGROUND: Drug therapy has become increasingly efficient, with more drugs available for treatment of an ever-growing number of conditions. Yet, drug use is reported to be sub optimal in several aspects, such as dosage, patient's adherence and outcome of therapy. The aim of the current study was to investigate the possibility to optimize drug therapy using computer programs, available on the Internet. METHODS: One hundred and ten officially endorsed text documents, published between 1996 and 2004, containing guidelines for drug therapy in 246 disorders, were analyzed with regard to information about patient-, disease- and drug-related factors and relationships between these factors. This information was used to construct algorithms for identifying optimum treatment in each of the studied disorders. These algorithms were categorized in order to define as few models as possible that still could accommodate the identified factors and the relationships between them. The resulting program prototypes were implemented in HTML (user interface) and JavaScript (program logic). RESULTS: Three types of algorithms were sufficient for the intended purpose. The simplest type is a list of factors, each of which implies that the particular patient should or should not receive treatment. This is adequate in situations where only one treatment exists. The second type, a more elaborate model, is required when treatment can by provided using drugs from different pharmacological classes and the selection of drug class is dependent on patient characteristics. An easily implemented set of if-then statements was able to manage the identified information in such instances. The third type was needed in the few situations where the selection and dosage of drugs were depending on the degree to which one or more patient-specific factors were present. In these cases the implementation of an established decision model based on fuzzy sets was required. Computer programs based on one of these three models could be constructed regarding all but one of the studied disorders. The single exception was depression, where reliable relationships between patient characteristics, drug classes and outcome of therapy remain to be defined. CONCLUSION: Algorithms for optimizing drug therapy can, with presumably rare exceptions, be developed for any disorder, using standard Internet programming methods
Section on Prospects for Dark Matter Detection of the White Paper on the Status and Future of Ground-Based TeV Gamma-Ray Astronomy
This is a report on the findings of the dark matter science working group for
the white paper on the status and future of TeV gamma-ray astronomy. The white
paper was commissioned by the American Physical Society, and the full white
paper can be found on astro-ph (arXiv:0810.0444). This detailed section
discusses the prospects for dark matter detection with future gamma-ray
experiments, and the complementarity of gamma-ray measurements with other
indirect, direct or accelerator-based searches. We conclude that any
comprehensive search for dark matter should include gamma-ray observations,
both to identify the dark matter particle (through the charac- teristics of the
gamma-ray spectrum) and to measure the distribution of dark matter in galactic

Patterns of in situ Mineral Colonization by Microorganisms in a ~60°C Deep Continental Subsurface Aquifer
The microbial ecology of the deep biosphere is difficult to characterize, owing in part to sampling challenges and poorly understood response mechanisms to environmental change. Pre-drilled wells, including oil wells or boreholes, offer convenient access, but sampling is frequently limited to the water alone, which may provide only a partial view of the native diversity. Mineral heterogeneity demonstrably affects colonization by deep biosphere microorganisms, but the connections between the mineral-associated and planktonic communities remain unclear. To understand the substrate effects on microbial colonization and the community response to changes in organic carbon, we conducted an 18-month series of in situ experiments in a warm (57°C), anoxic, fractured carbonate aquifer at 752 m depth using replicate open, screened cartridges containing different solid substrates, with a proteinaceous organic matter perturbation halfway through this series. Samples from these cartridges were analyzed microscopically and by Illumina (iTag) 16S rRNA gene libraries to characterize changes in mineralogy and the diversity of the colonizing microbial community. The substrate-attached and planktonic communities were significantly different in our data, with some taxa (e.g., Candidate Division KB-1) rare or undetectable in the first fraction and abundant in the other. The substrate-attached community composition also varied significantly with mineralogy, such as with two Rhodocyclaceae OTUs, one of which was abundant on carbonate minerals and the other on silicic substrates. Secondary sulfide mineral formation, including iron sulfide framboids, was observed on two sets of incubated carbonates. Notably, microorganisms were attached to the framboids, which were correlated with abundant Sulfurovum and Desulfotomaculum sp. sequences in our analysis. Upon organic matter perturbation, mineral-associated microbial diversity differences were temporarily masked by the dominance of putative heterotrophic taxa in all samples, including OTUs identified as Caulobacter, Methyloversatilis, and Pseudomonas. Subsequent experimental deployments included a methanogen-dominated stage (Methanobacteriales and Methanomicrobiales) 6 months after the perturbation and a return to an assemblage similar to the pre-perturbation community after 9 months. Substrate-associated community differences were again significant within these subsequent phases, however, demonstrating the value of in situ time course experiments to capture a fraction of the microbial assemblage that is frequently difficult to observe in pre-drilled wells
Differential regulation of TROP2 release by PKC isoforms through vesicles and ADAM17
TROP2, a cancer cell surface protein with both pro-oncogenic and anti-oncogenic properties is cleaved by ADAM17. ADAM17 dependent cleavage requires novel PKC activity which is blocked by the ADAM10/ADAM17 inhibitor GW64 as well as by the PKC inhibitor Bim-1. Full length TROP2 release is induced by classical PKC activation and blocked by Gö6979, without affecting ADAM17 dependent TROP2 cleavage. Full length TROP2 is released in ectosomes, as inhibition of endocytosis did not prevent release. Inhibition of the atypical PKC isoform PKCζ stimulated metalloproteinase dependent N-terminal alternative TROP2 cleavage. The resulting alternative TROP2 cleavage product remains membrane associated via a disulphide bond, but is released in microvesicles with an average size of 107 nm. Inhibition of endocytosis following PKCζ inhibition prevented alternative cleavage and release of TROP2, suggesting that these events require endocytic uptake and exosomal release of the corresponding microvesicles. The alternative TROP2 cleavage product was also found in PC3 cell lysates following deglycosylation, and may represent a novel biomarker in prostate cancer
From Quantum Systems to L-Functions: Pair Correlation Statistics and Beyond
The discovery of connections between the distribution of energy levels of
heavy nuclei and spacings between prime numbers has been one of the most
surprising and fruitful observations in the twentieth century. The connection
between the two areas was first observed through Montgomery's work on the pair
correlation of zeros of the Riemann zeta function. As its generalizations and
consequences have motivated much of the following work, and to this day remains
one of the most important outstanding conjectures in the field, it occupies a
central role in our discussion below. We describe some of the many techniques
and results from the past sixty years, especially the important roles played by
numerical and experimental investigations, that led to the discovery of the
connections and progress towards understanding the behaviors. In our survey of
these two areas, we describe the common mathematics that explains the
remarkable universality. We conclude with some thoughts on what might lie ahead
in the pair correlation of zeros of the zeta function, and other similar

Community-associated Methicillin-resistant Staphylococcus aureus in Pediatric Patients
Community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) infections increased from 2000 to 2003 in hospitalized pediatric patients in Houston. CA-MRSA was associated with greater illness than was infection with methicillin-susceptible strains. Children with CA-MRSA were younger and mostly African American. Of MRSA isolates, 4.5% had the inducible macrolide-lincosamide-streptogramin B phenotype

Wivesâ part-time employment and marital stability in Great Britain, West Germany and the United States
Many hail wivesâ part-time employment as a workâfamily balance strategy, but theories offer competing predictions as to the effects of wivesâ employment on relationship stability. We use panel data to test these competing hypotheses among recent cohorts of first-married couples in Great Britain, West Germany 1 and the United States. We find effects of wivesâ employment on marital stability var y across the countries. In West Germany with its high-quality part-time employment, couples where the wife works part time are significantly more stable. In the more liberal British and US labour markets, neither wivesâ part- nor full-time employment significantly alters divorce risk. In the United States, however, mothers working part time have significantly lower divorce risk. West German and British husbandsâ unemployment proves more detrimental to marital stability than wivesâ employment. These results highlight the importance of the socioeconomic context in structuring the optimal employment participation of both partners
