    Cooperation Between Photosynthetic and Antioxidant Systems: An Important Factor in the Adaptation of Ulva prolifera to Abiotic Factors on the Sea Surface

    Large-scale green tides have occurred continuously in the Yellow Sea of China from 2007 to 2018, and the causative species of the Yellow Sea green tide (YSGT) is Ulva prolifera. The thalli form floated thallus mats, and the thalli from different layers of the thallus mat suffer significantly different environmental conditions. In the present study, the environmental conditions of the surface layer (SL), middle layer (ML), and lower layer (LL) of the thallus mat from mid-June (Stage I) to mid-July (Stage II) were simulated. Photosynthetic traits and antioxidant systems were measured. The results showed that (1) photoprotective [non-photochemical quenching (NPQ) and cyclic electron transport (CEF)] and antioxidant systems both play important roles in protecting against abiotic factors in U. prolifera. (2) Cooperation between NPQ and CEF was observed in the ML group; CEF and the antioxidant system in the SL group work synergistically to protect the thalli. Furthermore, an inferred spatiotemporal attribute regarding the YSGT is presented: the significant changes in abiotic factors on the sea surface can easily affect the thalli of SL and ML from mid-June to mid-July, and those of LL can be affected in mid-July.     The use of selected bacteria and yeasts to control vibrio spp. in live food

    Vibrio species are a significant causative of mass mortality in mariculture worldwide, which can quickly accumulate in live food and transmit into the larval gut. With restrictions on the use of antibiotics in aquaculture, finding a proper solution to reduce the risk of Vibriosis is vital. This study aimed to evaluate the susceptibility of Vibrio harveyi, V. campbellii, V. anguillarum, and V. parahaemolyticus to twenty-six bacterial and yeast strains and use the beneficial ones to enrich live food (Branchiopod, Artemia franciscana, rotifer, Brachionus plicatilis and copepod, Tigriopus japonicus). Thus, a modified disk diffusion method was applied. After a susceptibility assay, the bacteria and yeast beneficial in suppressing the Vibrio species were labeled by fluorescent stain and used to measure the accumulation potential in different live foods. Also, the beneficial bacteria and yeast were used to enrich live foods, and then the count of loaded Vibrio was estimated after 5, 10, 15, and 20 h by the serial dilution method. From the total bacteria and yeast strains that were used, Candida parapsilosis, Pseudoalteromonas flavipulchra, Lactobacillus sakei, Bacillus natto, and B. amyloliquefaciens inhibited all four Vibrio species. The results of microbial labeling showed that L. sakei in Artemia, C. parapsilosis in rotifers, and V. harveyi in copepods had the highest accumulation rate. The results of the estimation of loaded Vibrio in different live foods also showed that the use of beneficial bacteria and yeast each significantly reduced the count of Vibrio. Application of bacteria and yeast to suppress pathogenic Vibrio maybe a sustainable method for preventing this pathogen from harmfully invading aquaculture and may also aid in reducing the chances of antibiotic resistance in pathogenic Vibrio

    Sex plays a role in the construction of epiphytic bacterial communities on the algal bodies and receptacles of Sargassum thunbergii

    The community structures of epiphytic bacteria on the surface of macroalgae are closely related to their host algae, but there is a lack of research on the differences between the epiphytic bacterial communities of male and female algae and their reproductive tissues. In this study, high-throughput sequencing was used to compare epiphytic bacterial community structures on the intertidal macroalgae Sargassum thunbergii and their receptacles between male and female samples. The epiphytic bacteria on the male and female algal bodies and receptacles had similar community structures with a large number of shared bacteria, but the samples clearly clustered separately, and the abundances of dominant taxa, specific bacteria, and indicator species differed, indicating that epiphytic bacterial communities differed significantly between the male and female S. thunbergii and their receptacles. In addition, the abundance of many predicted functional genes was significantly different between epiphytic bacteria on male and female algal bodies and receptacles, especially metabolism-related genes, and the abundances of predicted functional genes of epiphytic bacteria were significantly higher on both types of male samples than on female samples. Our study confirmed that the sex of the host algae influenced the epiphytic bacterial community structures on algae and algal reproductive tissues, and this role was mainly related to the host metabolism. The results reveal the role of host plant sex in the formation of epiphytic bacterial communities. These findings are helpful for obtaining an in-depth understanding of the construction mechanism of algae-associated bacteria

    Influence of dietary zinc on growth, zinc bioaccumulation and expression of genes involved in antioxidant and innate immune in juvenile mud crabs (Scylla paramamosain)

    The aim of present study was to investigate the effects of dietary Zn level on growth performance, Zn bioaccumulation, antioxidant capacity and innate immunity in juvenile mud crab (Scylla paramamosain). Six semi-purified diets were formulated to contain dietary Zn levels of 44.5, 56.9, 68.5, 97.3, 155.6 or 254.7 mg·kg-1, respectively. Dietary Zn level significantly influenced percent weight gain (PWG), with highest observed in crab fed the diet containing 97.3 mg·kg-1 Zn. Tissue Zn concentrations significantly increased as dietary Zn levels increased from 44.5 to 254.7 mg·kg-1. Retention of Zn in hepatopancreas increased with dietary Zn levels up to 68.5 mg·kg-1 and then significantly decreased. Moreover, inadequate dietary Zn (44.5 and 56.9 mg·kg-1) reduced anti-oxidation markers including total superoxide dismutase and copper/zinc superoxide dismutase activities and total anti-oxidant level. Crabs fed the diet with 44.5 mg·kg-1 Zn also showed significantly lower expression of genes involved in antioxidant status, such as Cu/Zn sod, glutathione peroxidase, catalase and thioredoxin than those fed diets containing 68.5 and 97.3 mg·kg-1 Zn. Highest activities of phenoloxidase and alkaline phosphatase were recorded in crab fed the diets containing 68.5 and 97.3 mg·kg-1 Zn. Expression levels of prophenoloxidase and toll-like receptor 2 were higher in crab fed the 97.3 mg·kg-1 Zn diet compared to crab fed the other diets. Based on PWG alone, the optimal dietary Zn level was estimated to be 82.9 mg·kg-1, with 68.5 to 97.3 mg·kg-1 recommended for maintaining optimal Zn bioaccumulation, oxidation resistance and innate immune response of juvenile mud crab

    Effects of dietary lipid level on growth, fatty acid profiles, antioxidant capacity and expression of genes involved in lipid metabolism in juvenile swimming crab, Portunus trituberculatus

    The regulation of lipogenesis and lipolysis mechanisms related to consumption of lipid has not been studied in swimming crab. The aims of present study were to evaluate the effects of dietary lipid levels on growth, enzymes activities, and expression of genes of lipid metabolism in hepatopancreas of juvenile swimming crab. Three isonitrogenous diets were formulated to contain crude lipid levels at 5.8 %, 9.9 % and 15.1 %, respectively. Crabs fed the diet containing 15.1 % lipid had significantly lower weight gain, specific growth rate and survival, and higher feed conversion ratio than those fed the 5.8 % and 9.9 % lipid diets. Crabs fed 5.8 % lipid had lower malondialdehyde concentrations in the hemolymph and hepatopancreas than those fed the other diets. Highest glutathione peroxidase in hemolymph and superoxide dismutase in hepatopancreas were observed in crabs fed 5.8 % lipid. The lowest fatty acid synthase and glucose 6-phosphate dehydrogenase activities in hepatopancreas were observed in crabs fed 15.1 % lipid, whereas crabs fed 5.8 % lipid had lower carnitine palmitoyltransferase-1 activity than those fed the other diets. Crabs fed 15.1 % lipid showed lower hepatopancreas expression of genes involved in LC-PUFA biosynthesis, lipoprotein clearance, fatty acid uptake, fatty acid oxidation, lipid anabolism and lipid catabolism than those fed the other diets, whereas expression of some genes of lipoprotein assembly and fatty acid oxidation were up-regulated compared with crabs fed 5.8 % lipid. Overall, high dietary lipid level can inhibit growth, reduce feed utilization and reduce antioxidant enzyme activities.     A possible pathway for rapid growth of sulfate during haze days in China

    Rapid industrialization and urbanization have caused frequent occurrence of haze in China during wintertime in recent years. The sulfate aerosol is one of the most important components of fine particles (PM[subscript 2. 5]) in the atmosphere, contributing significantly to the haze formation. However, the heterogeneous formation mechanism of sulfate remains poorly characterized. The relationships of the observed sulfate with PM[subscript 2. 5], iron, and relative humidity in Xi'an, China have been employed to evaluate the mechanism and to develop a parameterization of the sulfate heterogeneous formation involving aerosol water for incorporation into atmospheric chemical transport models. Model simulations with the proposed parameterization can successfully reproduce the observed sulfate rapid growth and diurnal variations in Xi'an and Beijing, China. Reasonable representation of sulfate heterogeneous formation in chemical transport models considerably improves the PM2. 5 simulations, providing the underlying basis for better understanding the haze formation and supporting the design and implementation of emission control strategies

    Characterization and health risk assessment of airborne pollutants in commercial restaurants in northwestern China: Under a low ventilation condition in wintertime

    Impacts on indoor air quality of dining areas from cooking activities were investigated in eight categories of commercial restaurants including Szechwan Hotpot, Hunan, Shaanxi Noodle, Chinese Barbecue, Chinese Vegetarian, Korean Barbecue, Italian, and Indian, in Northwestern China during December 2011 to January 2012. Chemical characterization and health risk assessment for airborne carbonyls, and particulate-bound polycyclic aromatic hydrocarbons (PAHs) and heavy metals were conducted under low ventilation conditions in wintertime. The highest total quantified carbonyls (Sigma(carbonyls)) concentration of 313.6 mu g m(-3) was found in the Chinese Barbecue, followed by the Szechwan Hotpot (222.6 mu g m(-3)) and Indian (221.9 mu g m(-3)) restaurants. However, the highest Sigma(carbonyls) per capita was found at the Indian restaurant (4500 mu g capita(-1)), suggesting that cooking methods such as stir-fly and bake for spices ingredients released more carbonyls from thermal cooking processes. Formaldehyde, acetaldehyde, and acetone were the three most abundant species, totally accounting for >60% of mass concentrations of the Sigma(carbonyls). Phenanthrene, chrysene, and benzo[a]anthracene were the three most abundant PAHs. Low molecular weight fraction (Sigma PAHs(<= 178)) had the highest contributions accounting for 40.6%-65.7%, much greater than their heaver counterparts. Diagnostic PAHs ratios suggest that cooking fuel and environmental tobacco smoke (ETS) contribute to the indoor PAHs profiles. Lead was the most abundant heavy metal in all sampled restaurants. High quantity of nickel was also found in samples due to the emissions from stainless-steel made kitchen utensils and cookware and ETS. Cancer risk assessments on the toxic substances demonstrate that the working environment of dining areas were hazard to health. Formation of reactive organic species (ROS) from the cooking activities was evidenced by measurement of hydroxyl radical (center dot OH) formed from simulating particulate matter (PM) react with surrogate lung fluid. The highest center dot OH concentration of 294.4 ng m(-3) was detected in Chinese Barbecue. In addition, the elevation of the concentrations of PM and center dot OH after non-dining periods implies that the significance of formation of oxidizing-active species indoor at poor ventilation environments.
