    The 16-22 amino acid fragment of the beta-amyloid peptide associated with the Alzheimer's disease, Abeta, is capable of forming amyloid fibrils. Here we study the aggregation mechanism of Abeta(16-22) peptides by unbiased thermodynamic simulations at the atomic level for systems of one, three and six Abeta(16-22) peptides. We find that the isolated Abeta(16-22) peptide is mainly a random coil in the sense that both the alpha-helix and beta-strand contents are low, whereas the three- and six-chain systems form aggregated structures with a high beta-sheet content. Furthermore, in agreement with experiments on Abeta(16-22) fibrils, we find that large parallel beta-sheets are unlikely to form.     Genomic heterogeneity and prevalence of hepandensovirus in Penaeus esculentus from Western Australia, and P. merguiensis from the Gulf of Carpentaria, Australia

    Decapod Hepandensovirus 1 (HDV), formerly known as hepatopancreatic parvovirus, has been associated with stunting, lowered production and outright mortalities in prawns in aquaculture. Despite the fact that broodstock are sourced and aquaculture farms are planned in the regions of northern and Western Australia, data on these parvoviruses from this region are limited. The prevalence of HDV in Penaeus esculentus and Penaeus merguiensis is moderate (34–51%) in southern Western Australia, Exmouth Gulf and the Gulf of Carpentaria but statistically higher (P < 0.05) in Shark Bay (82%). We speculate this is due to the topography of Shark Bay combined with the currents of the Indian Ocean gyre (IOG). Despite an on average 8–12% genomic heterogeneity, the nucleotide sequences of HDV in WA most closely align with HDV in regions associated with the IOG; Thailand, India, Tanzania, Madagascar; eastern Asia, Korea and less commonly, with sequences from the eastern coast of Australia. This potentially changes the paradigm of a single strain of HDV being ubiquitous in Australia and there was little risk in moving broodstock from WA to the eastern states, so there was no testing of broodstock for HDV. There is no strong evidence to clarify whether the strain of HDV in WA P. esculentus came from either its’ nearest genetic relatives, P. monodon or P. semisulcatus or from P. merguiensis from the Solanderian province of Australia. P. esculentus HDV appears to be most related to strains within the IOG. The HDV nucleotide heterogeneity of wild prawns contrasts strongly to studies undertaken with prawns from aquaculture where genetic selection may have occurred

    This article explores a neglected period in the history of classical scholarship: the first decades of the eighteenth century. It focuses on the tension between an evolving idea of method, and the tradition of personal polemic which had been an important part of the culture of scholarship since the Renaissance. There are two case studies: the conflict between Jean Le Clerc and Pieter Burman, and the controversy that followed Richard Bentley's edition of Horace's Odes. Both demonstrate the need to revise current paradigms for writing the history of scholarship, and invite us to reconsider the role of methodology in producing of scholarly authority

    Background: Migraine is associated with many debilitating symptoms that affect daily functioning. My Migraine Voice is a large global cross-sectional study aimed at understanding the full burden and impact of migraine directly from patients suffering from ≥4 monthly migraine days (MMDs) with a history of prophylactic treatment failure. Methods: This study was conducted worldwide (31 countries across North and South Americas, Europe, the Middle East and Northern Africa, and the Asia-Pacific region) using an online survey administered to adults with migraine who reported ≥4 MMDs in the 3 months preceding survey administration, with pre-specified criteria of 90% having used preventive migraine treatment (80% with history of ≥1 treatment failure). Prophylactic treatment failure was defined as a reported change in preventive medication by individuals with migraine for any reason, at least once. Results: In total, 11,266 individuals participated in the survey. Seventy-four percent of the participants reported spending time in darkness/isolation due to migraine (average: 19 h/month). While 85% of all respondents reported negative aspects of living with migraine (feeling helpless, depressed, not understood), sleeping difficulties (83%), and fear of the next attack (55%), 57% shared ≥1 positive aspect (learning to cope, becoming a stronger person). Forty-nine percent reported feeling limited in daily activities throughout all migraine phases. Migraine impact on professional, private, or social domains was reported by 87% of respondents (51% in all domains). In the previous 12 months, 38% of respondents had visited the emergency department (average: 3.3 visits), whereas 23% stayed in hospital overnight (average: 3.2 nights) due to migraine. Conclusions: The burden of migraine is substantial among this cohort of individuals with at least 4 migraine days per month and for whom at least 1 preventive migraine treatment had failed. Interestingly, respondents reported some positive aspects in their migraine journey; the greater resilience and strength brought on by coping with migraine suggests that if future treatments could address existing unmet needs, these individuals with migraine will be able to maximize their contribution to society

Primary prevention of dementia -preventing risk factors leading to disease development - is an emerging global public health priority. Primary prevention can be achieved in two ways: individual-level or population-level. In this rapid review, we quantify the proportion of contributing interventional evidence to the dementia primary prevention literature that is concerned with either approach. We searched Medline, the National Institute for Health and Care Excellence, Cochrane, the World Health Organization, and Google to identify systematic reviews that described primary prevention interventions for dementia. We used search terms related to dementia risk reduction, intervention/policy, and review. We analysed reference lists of included dementia prevention reviews to identify contributing primary prevention evidence, and categorised these as either individual-level or population-level. Additionally, we examined search strategies to investigate the likelihood of reviews identifying available population-level interventions. We included twelve of the 527 articles retrieved.     Efficacy of a synthetic calcium phosphate with submicron surface topography as autograft extender in lapine posterolateral spinal fusion.

    Posterolateral spinal fusion (PLF) is a common procedure in orthopedic surgery that is performed to fuse adjacent vertebrae to reduce symptoms related to spinal conditions. In the current study, a novel synthetic calcium phosphate with submicron surface topography was evaluated as an autograft extender in a validated rabbit model of PLF. Fifty-nine skeletally mature New Zealand white rabbits were divided into three groups and underwent single-level intertransverse process PLF at L4-5 using (1) autologous bone graft (ABG) alone or in a 1:1 combination with (2) calcium phosphate granules (ABG/BCPgranules ), or (3) granules embedded in a fast-resorbing polymeric carrier (ABG/BCPputty ). After 6, 9, and 12 weeks, animals were sacrificed and spinal fusion was assessed by manual palpation, Radiographs, micro-CT, mechanical testing (12 weeks only), histology, and histomorphometry. Based on all endpoints, all groups showed a gradual progression in bone formation and maturation during time, leading to solid fusion masses between the transverse processes after 12 weeks. Fusion assessments by manual palpation, radiography and histology were consistent and demonstrated equivalent fusion rates between groups, with high bilateral fusion rates after 12 weeks. Mechanical tests after 12 weeks indicated substantially lower range of motion for all groups, compared to non-operated controls. By histology and histomorphometry, the gradual formation and maturation of bone in the fusion mass was confirmed for each graft type. With these results, we describe the equivalent performance between autograft and a novel calcium phosphate material as an autograft extender in a rabbit model of PLF using an extensive range of evaluation techniques. © 2019 Wiley Periodicals, Inc.     Dynamical Evolution of Young Embedded Clusters: A Parameter Space Survey

    This paper investigates the dynamical evolution of embedded stellar clusters from the protocluster stage, through the embedded star-forming phase, and out to ages of 10 Myr -- after the gas has been removed from the cluster. The relevant dynamical properties of young stellar clusters are explored over a wide range of possible star formation environments using N-body simulations. Many realizations of equivalent initial conditions are used to produce robust statistical descriptions of cluster evolution including the cluster bound fraction, radial probability distributions, as well as the distributions of close encounter distances and velocities. These cluster properties are presented as a function of parameters describing the initial configuration of the cluster, including the initial cluster membership N, initial stellar velocities, cluster radii, star formation efficiency, embedding gas dispersal time, and the degree of primordial mass segregation. The results of this parameter space survey, which includes about 25,000 simulations, provide a statistical description of cluster evolution as a function of the initial conditions. We also present a compilation of the FUV radiation fields provided by these same cluster environments. The output distributions from this study can be combined with other calculations, such as disk photoevaporation models and planetary scattering cross sections, to ascertain the effects of the cluster environment on the processes involved in planet formation.Comment: 65 pages including 20 figures, accepted to ApJ Supplemen
