    A direct optical method for the study of grain boundary melting

    The structure and evolution of grain boundaries underlies the nature of polycrystalline materials. Here we describe an experimental apparatus and light reflection technique for measuring disorder at grain boundaries in optically clear material, in thermodynamic equilibrium. The approach is demonstrated on ice bicrystals. Crystallographic orientation is measured for each ice sample. The type and concentration of impurity in the liquid can be controlled and the temperature can be continuously recorded and controlled over a range near the melting point.     Model studies of timber shell roofs.

    The effects of four types of fastening and of scaling on the properties of a timber shell membrane were studied. The membrane was a three layer type made of ex 4 x 1 in. T & G radiata pine boards, the centre layer running at right angles to the outer layers. The fastenings were three densities of nailing and one of nail-gluing. Timber properties were characterised by grading modulus (g.m.) which was the stiffness of prototype boards on the flat under centre point loading over a 3 ft span while the fastenings were characterised by the load-slip behaviour of representative joints. The joint behaviour showed little correlation with g.m. Prototype and 1/5 scale model elements were tested in compression shear, bending and torsion and elastic constants were computed. The various constants were affected to different degrees by the four types of fastening according to the level of stress carried by the fastening under the various element loadings. The model similitude obtained in the elements was good when this was based on g.m., the load-slip behaviour of representative joints and the weight of glue spread per unit area of glueline. Two 1/5 scale model cylindrical timber shells were built and tested, one nailed and the other nail-glued. Their deflections were about twice those calculated by a simplified analysis for orthotropic shells. Their relative behaviour was similar to the relative behaviour of the model elements in shear which suggests that the discrepancy between calculated and observed deflections arose from shear deformation which the analysis ignored

    The New York State Cardiac Registries History, Contributions, Limitations, and Lessons for Future Efforts to Assess and Publicly Report Healthcare Outcomes

    In 1988, the New York State Health Commissioner was confronted with hospital-level data demonstrating very large, multiple-year, interhospital variations in short-term mortality and complications for cardiac surgery. The concern with the extent to which these differences were due to variations in patients' pre-surgical severity of illness versus hospitals' quality of care led to the development of clinical registries for cardiac surgery in 1989 and for percutaneous coronary interventions in 1992 in New York. In 1990, the Department of Health released hospitals' risk-adjusted cardiac surgery mortality rates for the first time, and shortly thereafter, similar data were released for hospitals and physicians for percutaneous coronary interventions, cardiac valve surgery, and pediatric cardiac surgery (only hospital data). This practice is still ongoing. The purpose of this communication is to relate the history of this initiative, including changes or purported changes that have occurred since the public release of cardiac data. These changes include decreases in risk-adjusted mortality, cessation of cardiac surgery in New York by low-volume and high-mortality surgeons, out-of-state referral or avoidance of cardiac surgery/angioplasty for high-risk patients, alteration of contracting choices by insurance companies, and modifications in market share of cardiac hospitals. Evidence related to these impacts is reviewed and critiqued. This communication also includes a summary of numerous studies that used New York's cardiac registries to examine a variety of policy issues regarding the choice and use of cardiac procedures, the comparative effectiveness of competing treatment options, and the examination of the relationship among processes, structures, and outcomes of cardiac care

    Compensatory ingestion upon dietary restriction in Drosophila melanogaster

    Dietary restriction extends the lifespan of numerous, evolutionarily diverse species. In D. melanogaster, a prominent model for research on the interaction between nutrition and longevity, dietary restriction is typically based on medium dilution, with possible compensatory ingestion commonly being neglected. Possible problems with this approach are revealed by using a method for direct monitoring of D. melanogaster feeding behavior. This demonstrates that dietary restriction elicits robust compensatory changes in food consumption. As a result, the effect of medium dilution is overestimated and, in certain cases, even fully compensated for. Our results strongly indicate that feeding behavior and nutritional composition act concertedly to determine fly lifespan. Feeding behavior thus emerges as a central element in D. melanogaster aging

    Association of coronary vessel characteristics with outcome in patients with percutaneous coronary interventions with incomplete revascularization

IMPORTANCE Many studies have compared outcomes for incomplete revascularization (IR) among patients undergoing percutaneous coronary interventions (PCI), but little is known about whether outcomes are related to the nature of the IR. OBJECTIVE To determine whether some coronary vessel characteristics are associated with worse outcomes in patients with PCI with IR. DESIGN, SETTING, AND PARTICIPANTS New York\u27s PCI registrywas used to examine mortality (median follow-up, 3.4 years) as a function of the number of vessels that were incompletely revascularized, the stenosis in those vessels, and whether the proximal left anterior descending artery was incompletely revascularized after controlling for other factors associated with mortality for patients with and without ST-elevationmyocardial infarction (STEMI). This was a multicenter study (all nonfederal PCI hospitals in New York State) that included 41 639 New York residents with multivessel coronary artery disease undergoing PCI in New York State between January 1, 2010, and December 31, 2012. EXPOSURES Percutaneous coronary interventions, with complete and incomplete revascularization. MAIN OUTCOMES AND MEASURES Medium-term mortality. RESULTS For patients with STEMI, the mean age was 62.8 years; 26.2%were women, 11.9% were Hispanic, and 81.5%were white. For other patients, the mean age was 66.6 years, 29.1%were women, 11.3%were Hispanic, and 79.1%were white. Incomplete revascularization was very common (78%among patients with STEMI and 71%among other patients). Patients with IR in a vessel with at least 90% stenosis were at higher risk than other patients with IR. This was not significant among patients with STEMI (17.18%vs 12.86%; adjusted hazard ratio [AHR], 1.16; 95%CI, 0.99-1.37) and significant among patients without STEMI (17.71% vs 12.96%; AHR, 1.15; 95%CI, 1.07-1.24). Similarly, patients with IR in 2 or more vessels had higher mortality than patients with completely revascularization and higher mortality than other patients with IR among patients with STEMI (20.37%vs 14.39%; AHR, 1.35; 95%CI, 1.15-1.59) and among patients without STEMI (20.10% vs 12.86%; AHR, 1.17; 95% CI, 1.09-1.59). Patients with proximal left anterior descending artery vessel IR had higher mortality than other patients with IR (20.09% vs 14.67%; AHR, 1.31; 95%CI, 1.04-1.64 for patients with STEMI and 20.78%vs 15.62%; AHR, 1.11; 95%CI, 1.01-1.23 for patients without STEMI). More than 20%of all PCI patients had IR of 2 or more vessels and more than 30% had IR with more than 90% stenosis. CONCLUSIONS AND RELEVANCE Patients with IR are at higher risk of mortality if they have IR with at least 90% stenosis, IR in 2 or more vessels, or proximal left anterior descending IR

    Leptin deficiency-induced obesity affects the density of mast cells in abdominal fat depots and lymph nodes in mice

    <p>Abstract</p> <p>Background</p> <p>Mast cells are implicated in the pathogenesis of obesity and insulin resistance. Here, we explored the effects of leptin deficiency-induced obesity on the density of mast cells in metabolic (abdominal fat depots, skeletal muscle, and liver) and lymphatic (abdominal lymph nodes, spleen, and thymus) organs. Fourteen-week-old male leptin-deficient <it>ob/ob </it>mice and their controls fed a standard chow were studied. Tissue sections were stained with toluidine blue to determine the density of mast cells. CD117/c-kit protein expression analysis was also carried out. Furthermore, mast cells containing immunoreactive tumor necrosis factor-α (TNF-α), a proinflammatory cytokine involved in obesity-linked insulin resistance, were identified by immunostaining.</p> <p>Results</p> <p><it>ob/ob </it>mice demonstrated adiposity and insulin resistance. In abdominal fat depots, mast cells were distributed differentially. While most prevalent in subcutaneous fat in controls, mast cells were most abundant in epididymal fat in <it>ob/ob </it>mice. Leptin deficiency-induced obesity was accompanied by a 20-fold increase in the density of mast cells in epididymal fat, but a 13-fold decrease in subcutaneous fat. This finding was confirmed by CD117/c-kit protein expression analysis. Furthermore, we found that a subset of mast cells in epididymal and subcutaneous fat were immunoreactive for TNF-α. The proportion of mast cells immunoreactive for TNF-α was higher in epididymal than in subcutaneous fat in both <it>ob/ob </it>and control mice. Mast cells were also distributed differentially in retroperitoneal, mesenteric, and inguinal lymph nodes. In both <it>ob/ob </it>mice and lean controls, mast cells were more prevalent in retroperitoneal than in mesenteric and inguinal lymph nodes. Leptin deficiency-induced obesity was accompanied by increased mast cell density in all lymph node stations examined. No significant difference in the density of mast cells in skeletal muscle, liver, spleen, and thymus was noted between <it>ob/ob </it>and control mice.</p> <p>Conclusions</p> <p>This study demonstrates that leptin deficiency-induced obesity is accompanied by alterations in the density of mast cells in abdominal fat depots. The divergent distribution of mast cells in subcutaneous versus visceral fat might partially account for their differential biological behavior. Mast cells might also play a role in adaptive immune response occurring in regional lymph nodes in obesity.</p

    Water- and nutrient-dependent effects of dietary restriction on Drosophila lifespan

    Dietary restriction (DR) is a widely conserved intervention leading to lifespan extension. Despite considerable effort, the mechanisms underlying DR remain poorly understood. In particular, it remains unclear whether DR prolongs life through conserved mechanisms in different species. Here, we show that, in the most common experimental conditions, lifespan extension by DR is abolished by providing Drosophila with ad libitum water, without altering food intake, indicating that DR, as conventionally studied in flies, is fundamentally different from the phenomenon studied in mammals. We characterize an alternative dietary paradigm that elicits robust lifespan extension irrespective of water availability, and thus likely represents a more relevant model for mammalian DR. Our results support the view that protein:carbohydrate ratio is the main dietary determinant of fly lifespan. These findings have broad implications for the study of lifespan and nutrition

    Photoproduction of π0\pi^0-pairs off protons and off neutrons

    Total cross sections, angular distributions, and invariant-mass distributions have been measured for the photoproduction of π0π0\pi^0\pi^0 pairs off free protons and off nucleons bound in the deuteron. The experiments were performed at the MAMI accelerator facility in Mainz using the Glasgow photon tagging spectrometer and the Crystal Ball/TAPS detector. The accelerator delivered electron beams of 1508 and 1557~MeV, which produced bremsstrahlung in thin radiator foils. The tagged photon beam covered energies up to 1400~MeV. The data from the free proton target are in good agreement with previous measurements and were only used to test the analysis procedures. The results for differential cross sections (angular distributions and invariant-mass distributions) for free and quasi-free protons are almost identical in shape, but differ in absolute magnitude up to 15\%. Thus, moderate final-state interaction effects are present. The data for quasi-free neutrons are similar to the proton data in the second resonance region (final state invariant masses up to \approx1550~MeV), where both reactions are dominated by the N(1520)3/2Δ(1232)3/2+πN(1520)3/2^-\rightarrow \Delta(1232)3/2^+\pi decay. At higher energies, angular and invariant-mass distributions are different. A simple analysis of the shapes of the invariant-mass distributions in the third resonance region is consistent with strong contributions of an NNσN^{\star}\rightarrow N\sigma decay for the proton, while the reaction is dominated by a sequential decay via a Δπ\Delta\pi intermediate state for the neutron. The data are compared to predictions from the Two-Pion-MAID model and the Bonn-Gatchina coupled channel analysis.Comment: accepted for publication in Eur. Phys. J.
