    Design and evaluation of taste masked chewable dispersible tablet of lamotrigine by melt granulation

    Lamotrigine, an antiepileptic drug (AED) of the phenyltriazine class, is chemically unrelated to existing antiepileptic drugs. Lamotrigine is also used in the treatment of depression and bipolar disorder. But it is a bitter drug and slightly soluble in water. Thus, in the work under taken, an attempt was made to mask the taste and to formulate into a chewable dispersible tablet by complexation with Precirol ATO-05, which also acts as taste masking agent. Since, these tablets can be swallowed in the form of dispersion; it is suitable dosage form for paediatric and geriatric patients. Drug-Precirol ATO-05 was prepared in drug to Precirol ATO-05 ratio of 1:2, 1:1.5, 1:1, 1:0.5. The prepared tablets were evaluated for general appearance, content uniformity, hardness, friability, taste evaluation, mouth feel, in vitro disintegration time, and in vitro dissolution studies. Tablets with Precirol ATO-05 have shown good disintegrating features, also, the dispersion not showing any bitter taste, indicate the capability of Precirol ATO-05 used, both as taste masking agents. Almost more than 90 percent of drug was released from the formulation within 1 h. Further formulations were subjected to stability testing for 3 months at temperatures 25±5ÂșC/60±5%RH; 30±5ÂșC/65±5%RH and 40±5ÂșC/75±5%RH. Tablets have shown no appreciable changes with respect to taste, disintegration, and dissolution profiles.Keywords: Lamotrigine; Melt granulation; Precirol; Taste masking; Chewable dispersible tablets

    Scalar and Spinor Perturbation to the Kerr-NUT Spacetime

    We study the scalar and spinor perturbation, namely the Klein-Gordan and Dirac equations, in the Kerr-NUT space-time. The metric is invariant under the duality transformation involving the exchange of mass and NUT parameters on one hand and radial and angle coordinates on the other. We show that this invariance is also shared by the scalar and spinor perturbation equations. Further, by the duality transformation, one can go from the Kerr to the dual Kerr solution, and vice versa, and the same applies to the perturbation equations. In particular, it turns out that the potential barriers felt by the incoming scalar and spinor fields are higher for the dual Kerr than that for the Kerr.     Blind docking of 4-Amino-7-Chloroquinoline analogs as potential dengue virus protease inhibitor using CB Dock a web server

    Currently, there is no approved drug to combat dengue. Various quinoline derivatives are known for potential antimalarial, antiviral activities, etc. In the present work docking between 4-Amino-7-Chloroquinoline analogs was performed with dengue virus NS2B/NS3 protease using CB dock, a web server. Lys74, Ile165, Val147, Asn152, Asn167, Trp83 and Leu149 amino acid residues were found to be in contact with designed 4-Amino-7-Chloroquinoline analogs. Different modes of binding like hydrogen bonding, hydrophobic interactions, etc with designed compounds improve potential anti-dengue characteristics in silico. ADME results are in acceptable range

    MOLECULAR CHARACTERIZATION OF DIFFERENT ADVANCED WHEAT VARIETAL LINES USING SDS-PAGE ELECTROPHORESIS DATA OF SEED STORAGE PROTEINS

    ABSTRACT The present study was undertaken to evaluate the genetic diversity in gluten subunits and protein content in seed endosperm of wheat for nutritional grading of 17 advanced varietal lines of wheat using SDS-PAGE, which is useful for agricultural development regarding quality food stuffs and breeding purposes. After calculating the standard factor in spectrophotometer for unit absorbance at 595 nm and the amount of protein were calculated. The wheat lines PZ-TS-5, PZ-TS-4 and PZ-TS-9 contained maximum amount of HMW-Gs proteins as compared to rest of the lines. By comparing with the six bands of the standard marker, band numbers 4 were common in all 17 wheat lines, but the other bands showed variation. Most frequent HMW protein bands had mole. Wt. 29.03 KDa marker band, while, in LMW protein bands most frequent bands were among the 14 KDa marker band. In cluster analysis, the above mentioned 3 wheat lines grouped in the same cluster showed that they were originated from the same parental line and genetically less diverged

    Leptin receptor signaling regulates protein synthesis pathways and neuronal differentiation in pluripotent stem cells

    The role of leptin receptor (OB-R) signaling in linking pluripotency with growth and development and the consequences of dysfunctional leptin signaling on progression of metabolic disease is poorly understood. Using a global unbiased proteomics approach we report that embryonic fibroblasts (MEFs) carrying the db/db mutation exhibit metabolic abnormalities, while their reprogrammed induced pluripotent stem cells (iPSCs) show altered expression of proteins involved in embryonic development. An upregulation in expression of eukaryotic translation initiation factor 4e (Eif4e) and Stat3 binding to the Eif4e promoter was supported by enhanced protein synthesis in mutant iPSCs. Directed differentiation of db/db iPSCs toward the neuronal lineage showed defects. Gene editing to correct the point mutation in db/db iPSCs using CRISPR-Cas9, restored expression of neuronal markers and protein synthesis while reversing the metabolic defects. These data imply a direct role for OB-R in regulating metabolism in embryonic fibroblasts and key developmental pathways in iPSCs.publishedVersio

    Spherically symmetric dissipative anisotropic fluids: A general study

    The full set of equations governing the evolution of self--gravitating spherically symmetric dissipative fluids with anisotropic stresses is deployed and used to carry out a general study on the behaviour of such systems, in the context of general relativity. Emphasis is given to the link between the Weyl tensor, the shear tensor, the anisotropy of the pressure and the density inhomogeneity. In particular we provide the general, necessary and sufficient, condition for the vanishing of the spatial gradients of energy density, which in turn suggests a possible definition of a gravitational arrow of time.
