    FORMULATION AND EVALUATION OF FLURBIPROFEN SUSTAINED RELEASE MATRIX TABLETS USING AN ALTERNATIVE TECHNIQUE AS POTENTIAL ECONOMIC APPROACH

    Objective: Development of sustained released tablets of flurbiprofen (FP) using an alternative technique to the traditional method of wet granulation process aiming to lower labor cost of the granulation process and formulating tablets with better characteristics. Methods: Eight matrix tablets formulae of FP were prepared by the alternative technique. The various characteristics of FP prepared tablets were investigated and comparatively evaluated by FP tablets prepared by the traditional method. The release data was analyzed according to various kinetic equations. The ulcerogenic effects of some FP tablets formulae were evaluated. Results: FP tablets prepared by the alternative technique displayed the best physical characteristics. All FP prepared tablets displayed good sustained-release patterns. FP tablets prepared by the traditional method showed a progress decrease in drug dissolution by increasing matrix concentration and hence, more matrix agent or multiple granulations was needed which makes granulation process to be difficult and cost. While, FP tablets prepared by the alternative technique displayed dissolution profiles with minimal differences in-between reflecting the low labor cost of granulation process where good sustained patterns could be obtained by a minor of the matrix agent. Histologically, the ulcerogenic effects of FP on the rats were highly reduced by FP tablets prepared by the alternative technique rather than others. The release kinetics of different prepared FP tablets displayed a coupled release pattern between diffusion and dissolution. Conclusion: This work proved the potential of the alternative technique as an effective economic approach for formulating FP sustained released tablets with better characteristics and low labor cost

    Predictors of Long Covid 19 Syndrome

    Background: Symptoms of covid persist in most cases with development of a multisystem syndrome called long covid syndrome.Objective: The aim of the work was to evaluate symptoms that persist after the acute stage of the disease in a cohort of patients with confirmed or suspected COVID 19 and to define the predictors for long COVID syndrome.Patients and method: This retrospective study involved 164 patients with previously confirmed or highly suspected COVID 19 and still attending to the post covid outpatient clinic in El-Minia University Chest Hospital complaining from persistence of symptoms or for regular follow up. Their data was obtained from their previous medical reports. Results: Post Covid 19 symptoms were found in about 86 % of the studied patients with 42.7% reported neuropsychiatric symptoms and 26.8% reported respiratory symptoms. The most common reported symptoms in nonsevere cases were the anxiety disorders (18.9%), followed by chronic fatigue and neuropathy (15.2%), while chronic dyspnea in 9.8% mostly in severe cases, while vertigo and headache in 9.1%, musculoskeletal symptoms in 6.1% and skin lesions in 3.7%.Conclusion: It could be concluded that persistence of symptoms is very common after acute Covid 19 infection. Chronic dyspnea was reported more frequently in severe cases while anxiety reported mostly by patients with mild disease

    Fermented Camel (Camelus dromedarius) and Bovine Milk Attenuate Azoxymethane-induced Colonic Aberrant Crypt Foci in Fischer 344 Rats

    Abstract: Background and Objective: Camel milk is a folk remedy that includes valuable nutrients and bioactive zoochemicals. However, the chemopreventive potential of camel milk against colon carcinogenesis is poorly understood. This study was conducted to investigate the chemopreventive potential of camel (Camelus dromedarius) and bovine milk as well as the impact of fermenting these milks with Lactobacillus acidophilus and Streptococcus thermophilus against early colon carcinogenesis as measured by the reduction of aberrant crypt foci (ACF) in azoxymethane (AOM)-treated Fischer 344 rats. Methodology: Each of 60 weanling male rats was assigned to one of 6 experimental diet groups: Fermented and unfermented camel milk with AOM, fermented and unfermented bovine milk with AOM and positive (PC, AOM only) and negative (NC, saline vehicle only) control groups. The animals were fed the corresponding diets for 3 weeks and then received two subcutaneous injections of AOM or vehicle for 2 consecutive weeks and they were then placed on the corresponding diets for 11 weeks. At termination, all rats were euthanized, colons were harvested and the ACF counts were determined for all tested groups. Immunohistochemical testing was then performed to examine cell proliferation and apoptosis in the camel milk groups. Results: Significant reductions (p<0.05) (48.4-62.1%) in the total ACF count were observed in the colons of the rats fed all milk diets compared with rats fed on PC. However, significant differences were not observed in the total ACF between the camel and bovine milk diets or between the fermented and unfermented milk diets. In addition, significant changes were not observed in the apoptotic index for the camel milk diet compared with the index values for PC and β-catenin was generally localized to the membrane in all examined specimens. Conclusion: By virtue of its bioactive components, camel milk exhibited a chemopreventive potential against early colon carcinogenesis, however, fermentation did not improve its chemopreventive potential

    The effects of rearing water depths and feed types on the growth performance of African catfish (Clarias gariepinus)

    African catfish (Clarias gariepinus) has been growing in popularity as a culture species globally. Although considerable research has been carried out on this species’ feeding preferences, no research has been carried out on optimum water level for growth. This is particularly surprising given this species' benthic nature. This study assessed the effect of different combinations of water levels and types of feed on the growth performance of African catfish. The study employed a 3 × 2 factorial design with three ponds of different water levels (0.5, 1 and 1.5 m) and two types of feeds (floating and sinking). Twelve earthen ponds (1 × 2 m) were each stocked with 16 catfish fingerlings (mean weight ~100 g), and their growth was monitored for 12 weeks. The fish cultured in the shallowest water grew significantly faster than those cultured in the deepest ponds. At the same time, fish cultured in the shallowest ponds had the lowest feed intake rates and consequently the lowest feed conversion ratios. Catfish fed sinking pellets grew faster than those fed floating pellets although the difference was not significant. There was no significant interaction between pond water depth and feed type. The results of this study suggest that the growth and feeding efficiency of C. gariepinus can be optimized by culturing in fairly shallow ponds (0.5 m)

    Green Synthesis of Silver Nanoparticles Using Hypericum perforatum L. Aqueous Extract with the Evaluation of Its Antibacterial Activity against Clinical and Food Pathogens

    The rapid development of nanotechnology and its applications in medicine has provided the perfect solution against a wide range of different microbes, especially antibiotic-resistant ones. In this study, a one-step approach was used in preparing silver nanoparticles (AgNPs) by mixing silver nitrate with hot Hypericum perforatum (St. John’s wort) aqueous extract under high stirring to prevent agglomeration. The formation of silver nanoparticles was monitored by continuous measurement of the surface plasma resonance spectra (UV-VIS). The effect of St. John’s wort aqueous extract on the formation of silver nanoparticles was evaluated and fully characterized by using different physicochemical techniques. The obtained silver nanoparticles were spherical, monodisperse, face-centered cubic (fcc) crystal structures, and the size ranges between 20 to 40 nm. They were covered with a capping layer of organic compounds considered as a nano dimension protective layer that prevents agglomeration and sedimentation. AgNPs revealed antibacterial activity against both tested Gram-positive and Gram-negative bacterial strains causing the formation of 13–32 mm inhibition zones with MIC 6.25–12.5 µg/mL; Escherichia coli strains were resistant to tested AgNPs. The specific growth rate of S. aureus was significantly reduced due to tested AgNPs at concentrations ≥½ MIC. AgNPs did not affect wound migration in fibroblast cell lines compared to control. Our results highlighted the potential use of AgNPs capped with plant extracts in the pharmaceutical and food industries to control bacterial pathogens’ growth; however, further studies are required to confirm their wound healing capability and their health impact must be critically evaluate

    miRNome of Child A hepatocellular carcinoma in Egyptian patients

    IntroductionHepatocellular carcinoma (HCC) has different etiologies that contribute to its heterogeneity. In regards to the number of HCC patients, Egypt ranks third in Africa and fifteenth worldwide. Despite significant advancements in HCC diagnosis and treatment, the precise biology of the tumor is still not fully understood, which has a negative impact on patient outcomes.MethodsAdvances in next-generation sequencing (NGS) have increased our knowledge of the molecular complexity of HCC.Results &amp; discussionIn this research, 16 HCC and 6 tumor adjacent tissues (control) of Child A Egyptian patients were successfully profiled for the expression profile of miRNAs by NGS. Forty-one differentially expressed miRNAs (DEMs) were found by differential expression analysis, with 31 being upregulated and 10 being downregulated. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis was then conducted on these differentially expressed miRNAs revealing that Sensitivity and specificity analysis showed that hsa-miR-4488, hsa-miR-3178, and hsa-miR-3182 were unique miRNAs as they are expressed in HCC tissues only. These miRNAs were all highly involved in AMPK signaling pathways. However, hsa-miR-214-3p was expressed in control tissues about eight times higher than in cancer tissues and was most abundant in “pathways in cancer and PI3K-Akt signaling pathway” KEGG terms. As promising HCC diagnostic markers, we here suggest hsa-miR-4488, hsa-miR-3178, hsa-miR-3182, and hsa-miR-214-3p. We further urge future research to confirm these markers' diagnostic and prognostic potential as well as their roles in the pathophysiology of HCC

    Programmed Chemotherapy for Patients with Metastatic Unresectable Gastric Cancer

    BACKGROUND: Recent advances in the treatment of metastatic unresectable gastric cancers (MGC) include the development of new antitumor drugs and new regimens for their use. However, the selection of individually designed regimens by gastric cancer (GC) subtype remains problematic. Here, we investigated the clinical usefulness of programmed chemotherapy. METHODOLOGY/PRINCIPAL FINDINGS: MGC patients were classified into three groups by clinical condition. We implemented a chemotherapy program consisting of S-1 combination regimens. Median survival time (MST) of level 1 patients was 416 days (95% CI: 313-506 days), with an overall response rate of 47%. MSTs of level 2 and 3 patients were 208 (95% CI: 153-287 days) and 95 days (95% CI: 28-136 days), respectively. Grade 3-4 toxicities were neutropenia in 12% and anorexia in 6%. All treatment- related toxicities were resolved, and no treatment-related deaths occurred. CONCLUSIONS/SIGNIFICANCE: This program provided reasonable selection of case-matching regimens and may improve the survival of patients with MGC. Further, it may represent the first clinical tool to provide efficient chemotherapy course selection for MGC. Ongoing analysis of newly developed drugs and regimens will allow the efficacy of this chemotherapy program to be improved

    Soil application of effective microorganisms and nitrogen alleviates salt stress in hot pepper (Capsicum annum L.) plants

    The application of effective microorganisms (EMs) and/or nitrogen (N) have a stimulating effect on plants against abiotic stress conditions. The aim of the present study was to determine the impact of the co-application of EMs and N on growth, physio-biochemical attributes, anatomical structures, nutrients acquisition, capsaicin, protein, and osmoprotectant contents, as well as the antioxidative defense system of hot pepper (Capsicum annum L.) plants. In the field trials, EMs were not applied (EMs-) or applied (EMs+) along with three N rates of 120, 150, and 180 kg unit N ha-1 (designated as N120, N150, and N180, respectively) to hot pepper plants grown in saline soils (9.6 dS m-1). The application of EMs and/or high N levels attenuated the salt-induced damages to hot pepper growth and yield. The application of EMs+ with either N150 or N180 increased the number, average weight and yield of fruits by 14.4 or 17.0%, 20.8 or 20.8% and 28.4 or 27.5%, respectively, compared to hot pepper plants treated with the recommended dose (EMs- × N150). When EMs+ was individually applied or combined with either N150 or N180, increased accumulation of capsaicin were observed by 16.7 or 20.8%, protein by 12.5 or 16.7%, proline by 19.0 or 14.3%, and total soluble sugars by 3.7 or 7.4%, respectively, in comparison with those treated with the integrative EMs- × N150. In addition, the non-enzymatic contents (ascorbate, and glutathione) and enzymatic activities (catalase, superoxide dismutase, and glutathione reductase) of the antioxidant defense systems significantly increased in hot pepper plants treated with EMs+ alone or combined with N150 or N180 under salt stress conditions. Higher accumulation of nutrients (N, P, K+, and Ca2+) along with reduced Na+ acquisition was also evidenced in response to EMs+ or/and high N levels. Most anatomical features of stems and leaves recovered in hot pepper plants grown in saline soils and supplied with EMs+ and N. The application of EMs and N is undoubtedly opening new sustainable approaches toward enhancing abiotic stress tolerance in crops (e.g. hot pepper)

    Ferritin level : a predictor of severity and mortality in hospitalized COVID-19 patients

    Introduction: This study aims to investigate in-hоsрitаl mоrtаlity in severe асute resрirаtоry syndrоme соrоnаvirus 2 раtients strаtified by serum ferritin levels. Methods: Patients were stratified based on ferritin levels (ferritin levels ≤ 1000 or >1000). Results: Approximately 89% (118) of the patients with ferritin levels > 1000 had pneumonia, and 51% (67) had hypertension. Fever (97, 73.5%) and shortness of breath (80, 61%) were two major symptoms among the patients in this group. Logistic regression analysis indicated that ferritin level (odds ratio [OR] = 0.36, 95% confidence interval [CI] = 0.21–0.62; p 1000. Conclusion: In this study, higher levels of serum ferritin were found to be an independent predictor of in-hоsрitаl mоrtаlity
