Economical genotyping of little blue penguin (Eudyptula minor) clades from feather-based DNA

    Get PDF
Determination of clade membership is a crucial requirement for many research questions addressing phylogeography, population structure, mating patterns, speciation, and hybridisation. The little blue penguin (Eudyptula minor) can be separated into two deeply divergent clades. However, assigning clade membership in little blue penguins requires molecular methods. Genetic sequencing can be used to identify clade membership but is expensive. Here, we present an economical alternative to the use of sequencing to determine little blue penguin clade membership. We extracted DNA from feathers using a method that produced reasonable quantities of DNA. We then amplified the D-loop section of the mitochondrial control region from total genomic DNA extracts, using the primers 'C L-tRNAglu' and 'D H-Dbox' followed by digestion with the restriction enzyme AluI. When visualised on a gel, distinctive banding patterns clearly indicated clade membership. We sequenced a subset of our samples and verified the accuracy of this method. The methods we present should facilitate little blue penguin research through a cost-effective approach to clade analysis as well as a successful technique to extract DNA from feathers when blood or tissue samples are not available

K−K^- - nucleus relativistic mean field potentials consistent with kaonic atoms

K−K^- atomic data are used to test several models of the K−K^- nucleus interaction. The t(ρ\rho)ρ\rho optical potential, due to coupled channel models incorporating the Λ\Lambda(1405) dynamics, fails to reproduce these data. A standard relativistic mean field (RMF) potential, disregarding the Λ\Lambda(1405) dynamics at low densities, also fails. The only successful model is a hybrid of a theoretically motivated RMF approach in the nuclear interior and a completely phenomenological density dependent potential, which respects the low density theorem in the nuclear surface region. This best-fit K−K^- optical potential is found to be strongly attractive, with a depth of 180 \pm 20 MeV at the nuclear interior, in agreement with previous phenomenological analyses.Comment: revised, Phys. Incremental expansions for Hubbard-Peierls systems

The ground state energies of infinite half-filled Hubbard-Peierls chains are investigated combining incremental expansion with exact diagonalization of finite chain segments. The ground state energy of equidistant infinite Hubbard (Heisenberg) chains is calculated with a relative error of less than 3⋅10−33 \cdot 10^{-3} for all values of UU using diagonalizations of 12-site (20-site) chain segm ents. For dimerized chains the dimerization order parameter dd as a function of the onsite repulsion interaction UU has a maximum at nonzero values of UU, if the electron-phonon coupling gg is lower than a critical value gcg_c. The critical value gcg_c is found with high accuracy to be gc=0.69g_c=0.69. For smaller values of gg the position of the maximum of d(U)d(U) is approximately 3t3t, and rapidly tends to zero as gg approaches gcg_c from below. We show how our method can be applied to calculate breathers for the problem of phonon dynamics in Hubbard-Peierls systems.Comment: 4 Pages, 3 Figures, REVTE

Kaon effective mass and energy from a novel chiral SU(3)-symmetric Lagrangian

    Get PDF
A new chiral SU(3) Lagrangian is proposed to describe the properties of kaons and antikaons in the nuclear medium, the ground state of dense matter and the kaon-nuclear interactions consistently. The saturation properties of nuclear matter are reproduced as well as the results of the Dirac-Br\"{u}ckner theory. Our numerical results show that the kaon effective mass might be changed only moderately in the nuclear medium due to the highly non-linear density effects. After taking into account the coupling between the omega meson and the kaon, we obtain similar results for the effective kaon and antikaon energies as calculated in the one-boson-exchange model while in our model the parameters of the kaon-nuclear interactions are constrained by the SU(3) chiral symmetry.Comment: 13 pages, Latex, 3 PostScript figures included; replaced by the revised version, to appear in Phys. On the exact conservation laws in thermal models and the analysis of AGS and SIS experimental results

The production of hadrons in relativistic heavy ion collisions is studied using a statistical ensemble with thermal and chemical equilibrium. Special attention is given to exact conservation laws, i.e. certain charges are treated canonically instead of using the usual grand canonical approach. For small systems, the exact conservation of baryon number, strangeness and electric charge is to be taken into account. We have derived compact, analytical expressions for particle abundances in such ensemble. As an application, the change in K/πK/\pi ratios in AGS experiments with different interaction system sizes is well reproduced. The canonical treatment of three charges becomes impractical very quickly with increasing system size. Thus, we draw our attention to exact conservation of strangeness, and treat baryon number and electric charge grand canonically. We present expressions for particle abundances in such ensemble as well, and apply them to reproduce the large variety of particle ratios in GSI SIS 2 A GeV Ni-Ni experiments. At the energies considered here, the exact strangeness conservation fully accounts for strange particle suppression, and no extra chemical factor is needed.Comment: Talk given at Strangeness in Quark Matter '98, Padova, Italy (1998). Influence of Impact Parameter on Thermal Description of Relativistic Heavy Ion Collisions at GSI/SIS

Attention is drawn to the role played by the size of the system in the thermodynamic analysis of particle yields in relativistic heavy ion collisions at SIS energies. This manifests itself in the non-linear dependence of K+ and K- yields in AAAA collisions at 1 -- 2 A.GeV on the number of participants. It is shown that this dependence can be quantitatively well described in terms of a thermal model with a canonical strangeness conservation. The measured particle multiplicity ratios (pi+/p, pi-/pi+, d/p, K+/pi+ and K+/K- but not eta/pi0) in central Au-Au and Ni-Ni collisions at 0.8 -- 2.0 A.GeV are also explained in the context of a thermal model with a common freeze-out temperature and chemical potential. Including the concept of collective flow a consistent picture of particle energy distributions is derived with the flow velocity being strongly impact-parameter dependent.Comment: revtex, 20 figure

Comparing plasma and faecal measures of steroid hormones in Adelie penguins Pygoscelis adeliae

    Get PDF
Physiological measurements of both stress and sex hormones are often used to estimate the consequences of natural or human-induced change in ecological studies of various animals. Different methods of hormone measurement exist, potentially explaining variation in results across studies; methods should be cross-validated to ensure that they correlate. We directly compared faecal and plasma hormone measurements for the first time in a wild free-living species, the Adelie penguin (Pygoscelis adeliae). Blood and faecal samples were simultaneously collected from individual penguins for comparison and assayed for testosterone and corticosterone (or their metabolites). Sex differences and variability within each measure, and correlation of values across measures were compared. For both hormones, plasma samples showed greater variation than faecal samples. Males had higher mean corticosterone concentrations than females, but the difference was only statistically significant in faecal samples. Plasma testosterone, but not faecal testosterone, was significantly higher in males than females. Correlation between sample types was poor overall, and weaker in females than in males, perhaps because measures from plasma represent hormones that are both free and bound to globulins, whereas measures from faeces represent only the free portion. Faecal samples also represent a cumulative measure of hormones over time, as opposed to a plasma ‘snapshot’ concentration. Our data indicate that faecal sampling appears more suitable for assessing baseline hormone concentrations, whilst plasma sampling may best define immediate responses to environmental events. Consequently, future studies should ensure that they select the most appropriate matrix and method of hormone measurement to answer their research questions
