Design of Electrophoresis Device for Optimation of Dna Visualization and Dna Concentration Using Software
Molecules of deoxyribo nucleic acid (DNA) show a strong polarization allowing for both motions of the dielectrophoresis induced by polarization and electrophoresis based on its negative charge. Considering high subjective and less quantifiable result of the visualization based qualitative test of DNA on gel electrophoresis, designing the tool using a combination of the principles of electrophoresis and dielectrophoresis completed with a software for optimization of DNA visualization and to measure the concentration of small and large–sized DNA fragment is very needed. Accuracy of measurement of DNA concentration using a spectrophotometer UV /VIS is depend on its availability in the laboratory. The aim of this study was to design device for optimization of DNA visualization and measuring the concentration in the gel electrophoresis using MatLab- based software. Experiment using this software measured the concentration of DNA based on its visualization and compared it with calculation obtained from spectrophotometer UV/VIS. The research results showed that the amount of DNA analysed using a spectrophotometer tend to similar with the measurement results using the MatLab-based software although there was differences in quantitative values

Analysis of Employee Perception About the Whistleblowing System Toward Intention to Conduct Whistleblowing
This research is entitled “Analysis of Employee Perception about the Whistleblowing System toward Intention to Conduct Whistleblowing (The Study on Employees of Purbalingga and Purwokerto Tax Office)”The objective of this research is to examined individual factors that can influence intention to conduct whistleblowing by applying the Theory of Planned Behavior, namely behavioral attitudes, subjective norms and perceived behavioral on the employees of the Purbalingga Tax Office and employees of Purwokerto Tax Office. This research was conducted at the Purbalingga Tax Office and Purwokerto Tax Office. Using variable that explain in Theory of Planned Behavior, the result of this study can be conclude that: (1) there is positive effect of behavioral attitude toward the intention of employee to conduct whistleblowing, (2) there is positive effect of subjective norm toward the intention of employee to conduct whistleblowing information, and (3) there is positive effect of perceived behavioral control toward the intention of employee to conduct whistleblowing
Substitusi Silase Secara Kimiawi Limbah Padat Surimi Ikan Swanggi (Priacanthus Macracanthus) Pada Tepung Ikan Terhadap Retensi Energi Dan Rasio Konversi Pakan Benih Ikan Nila (Oreochromis Niloticus) [Chemical Silage Substitution of Swanggi Surimi Solid Waste (Priacanthus Macracanthus) of Fish Meal Toward Energy Retention and Tiplapia Seeds (Oreochromis Niloticus) Feed Conversion Rate]
Swanggi surimi waste can be processed into economically valuable source of protein through chemical treatment processes produce crude protein content ranging from 51%. Results of high energy retention and reduced feed conversion ratio of solid waste as a substitute swanggi fish surimi fish meal is expected to optimize the growth of tilapia with lower feed prices, thereby reducing the high cost of feed in aquaculture. This study aims to determine the chemical silage substitution of solid waste swanggi fish surimi (P. macracanthus) in fish meal. The experimental design completely randomized design (CRD) with four treatments with five replications. The treatment used is the number of different proteins in each feed rations. The main parameters are observed energy retention (%) and feed conversion ratio. Analysis of data using analysis of variance (ANOVA) and followed Duncan's multiple range test. These results indicate that substitution is chemically silage solid waste swanggi surimi fish in fish meal provides highly significant effect (p <0.05) on the retention of energy and feed conversion ratio tilapia fish. The average retention of the highest energy on a few observations are in treatment P3 (67,90a ± 0.86%) and the lowest at P0 (64,72b ± 1.17%). Average feed conversion ratio was lowest for the treatment P3 (2,41c ± 0.028) and the highest pda P0 (2,50a ± 0.018)
A general secretion signal for the mycobacterial type VII secretion pathway
Mycobacterial pathogens use specialized type VII secretion (T7S) systems to transport crucial virulence factors across their unusual cell envelope into infected host cells. These virulence factors lack classical secretion signals and the mechanism of substrate recognition is not well understood. Here we demonstrate that the model T7S substrates PE25/PPE41, which form a heterodimer, are targeted to the T7S pathway ESX-5 by a signal located in the C terminus of PE25. Site-directed mutagenesis of residues within this C terminus resulted in the identification of a highly conserved motif, i.e., YxxxD/E, which is required for secretion. This motif was also essential for the secretion of LipY, another ESX-5 substrate. Pathogenic mycobacteria have several different T7S systems and we identified a PE protein that is secreted by the ESX-1 system, which allowed us to compare substrate recognition of these two T7S systems. Surprisingly, this ESX-1 substrate contained a C-terminal signal functionally equivalent to that of PE25. Exchange of these C-terminal secretion signals between the PE proteins restored secretion, but each PE protein remained secreted via its own ESX secretion system, indicating that an additional signal(s) provides system specificity. Remarkably, the YxxxD/E motif was also present in and required for efficient secretion of the ESX-1 substrates CFP-10 and EspB. Therefore, our data show that the YxxxD/E motif is a general secretion signal that is present in all known mycobacterial T7S substrates or substrate complexes
