    Rat brain Na,K-ATPase partially purified by SDS from synaptic plasma membranes (SPM) was immobilized by adsorption on nitrocellulose (NC), polyvinylidene fluoride (PVDF) and glass fiber (GF) membranes, Partial SDS solubilization increased the enzyme activity by 40%. With regard to the presentation of the enzyme activity. nitrocellulose Was shown to be the optimal support for die immobilization, The enzyme showed the highest percentage activity (14%) after 30 min of SPM adsorption. at 20 degreesC under the vaccum. with 25 mug of proteins per NC disc filter. In addition, adsorption on NC stabilizes the Na,K-ATPase, since the activity was substantial 72 h after adsorption at 20 degreesC, After adsorption. the sensitivity of the enzyme to HgCl2 and CdCl2 inhibition was higher, The results show that immobilized Na,K-NTPase SPM can be used as a practical model for the detection of metal ions in different samples.Delimično prečišćena Na,K-ATPaza sinaptičkih plazma–membrana (SPM) mozga pacova imobilizovana je adsorpcijom na nitrocelulozne (NC) poliviniliden-fluorid (PVDF) membrane i membrane od staklenih vlakana (SV). Aktivnost enzima delimično prečišćenog solubilizacijom SDS-om povećana je oko 40%. Najveći procenat aktivnosti (14%) enzim zadržava posle 30 minuta adsorpcije SPM na 20ºC, pod vakuumom, sa 25 μg proteina po nitroceluloznom disku. Na,K-ATPaza imobilizovana na nitroceluloznoj membrani stabilna je 72 sata na 20ºC. Adsorpcijom, osetljivost enzima na inhibiciju Hg2+ i Cd2+ se povećava. Rezultati pokazuju da se imobilizovana Na,K-ATPaza SPM može koristiti za detekciju toksičnih metalnih jona u različitim uzorcima

    THE EFFECTS OF SOLAR ACTIVITY: ELECTRONS IN THE TERRESTRIAL LOWER IONOSPHERE

    Solar flare X-ray energy can cause strong enhancements of the electron density in the Earth’s atmosphere. This intense solar radiation and activity can cause sudden ionospheric disturbances (SIDs) and further create ground telecommunication interferences, blackouts as well as some natural disasters and caused considerable material damage. The focus of this contribution is on the study of these changes induced by solar X-ray flares using narrowband Very Low Frequency (VLF, 3–30 kHz) and Low Frequency (LF, 30–300 kHz) radio signal analysis. The model computation and simulation were applied to acquire the electron density enhancement induced by intense solar radiation. The obtained results confirmed the successful use of applied technique for detecting space weather phenomena such as solar explosive events as well for describing and modeling the ionospheric electron density which are important as the part of electric terrestrial-conductor environment through which external-solar wind (SW) electrons can pass and cause natural disasters on the ground like fires

    Use of high-energy ionizing radiation for microbiological decontamination of coastal soil in the Kolubara river basin, Serbia

    The Kolubara river pollutes the coastal land in the river basin and makes it unsuitable for agricultural activities in that area. Also, contaminated land poses a risk to the environment. Different methods can be used for soil decontamination. These methods include biological treatment/bioremediation, chemical oxidation, soil stabilization, physical methods, such as soil leaching, or treatment with high-energy ionizing radiation. Gamma irradiation of soil is a well-known method of inhibiting microbial activity. This paper investigated the influence of different doses and dose rates of gamma irradiation on microorganisms' decontamination of coastal soil, in the Kolubara river basin. The irradiation effects on reducing the total number of microorganisms and removing mold and pathogenic bacteria from soil samples were examined. Gamma radiation affects the soil's organic matter, causing the formation of free reactive radicals, which act as reducing and oxidizing agents, cleaving C-C bonds, and depolymerizing carbohydrates. It was found that a dose of 3 kGy of gamma radiation, neutralizes all pathogenic bacteria, a dose of 5 kGy deactivates mold in soil samples, and a dose of 10 kGy is optimal to kill all microorganisms in the samples and sterilize exposed soil. The research showed that the dose rate does not significantly affect microbiological decontamination of soil using gamma irradiation. The content of heavy metals in soil was determined, and the obtained values were compared with the remediation limit values prescribed by the regulations. It was concluded that the content of heavy metals in the analyzed soil samples is below the limit of remediation values. The only exception is the slightly increased copper content in one sample. The result of this research is the conclusion that the coastal land from the Kolubara basin can be decontaminated by gamma radiation treatment. This advanced soil treatment technology is available in Serbia because there is an industrial plant for gamma radiation treatment within the Vinča Institute

    Schizophyllum commune: The main cause of dying trees of the Banja Luka arbored walks and parks

    In the frame of investigation of the main cause of dying trees of the main arbored walks (Mladena Stojanovića Aley and Park), the investigation of the presence and diversity of macrofungi in Banja Luka City were undertaken in the period 2006-2011. Relatively poor generic diversity of lignicolous (pathogenic or potentially pathogenic and saprotrophic) macrofungi with only 16 species representing this group (13 basidiomycets: Schizophyllum commune, Fomes fomentarius, Stereum hirsutum, Coriolus versicolor, Flammulina velutipes, Pseudotrametes gibbosa, Ganoderma applanatum, G. lucidum, G. adspersum, Polyporus squamosus, Meripilus giganteus, Laetiporus sulphureus, Auricu­laria auricula-judae, and 3 ascomycets: Nectria cinnabarina, Xylaria hypoxylon, X. poly­morpha) were recorded. Such a poor qualitative composition of this very important fungal group could be explained by the reduction in the number of plant species in arbored walks and alleys, as well as the reduction in the number of fungi resistant to heavy air pollution caused by nearby (1-5m) fuel combustion in engines. Although only preliminary, our results pointed to the necessity of conservation and protection of the most beautiful features of Banja Luka and its alleys and arbored walks, by undertaking the measures of curing damaged trees and treating them with fungicides in order to wipe out the epiphytia caused in more than 95% of cases (dated May 2011) by Split-gill (Schizophyllum commune), present on dead wood but also on damaged trees of Aesculus hyppocastaneum (127 trees), Tilia cordata (124 trees), Tilia platyphyllos (36 trees), Tilia argentea (40 trees), Acer negundo (20 trees), Platanus acerifolia (2 trees), Robinia pseudoacacia (3 trees), Fraxinus ornus (1 tree), Betula pendula (1 tree), Catalpa sp. (2 trees), etc. Altogether, during the last decade, around 200 trees collapsed or were sanitary cut in Banja Luka arbored walk from the Malta site to the Green bridge, a total length around 5 km. The reason for this was primarily due to Split-gill fungus and the restoration of arbored walks in the streets extremely polluted by engine fuel consumption in the zone of Mladena Stojanovića street. By analyzing the trees along the City of Banja Luka main street it could be concluded that, besides the appearance of suffocation of plants, due to wide asphalt surfaces that are located immediately next to the tree-trunks and heavy air pollution, fungi caused illnesses are the most important cause of the decline of trees. With its great adaptation to arid climate and ability to resist to the air pollution, Schizophyllum commune turned out to be the most aggressive and successful universal fungal invader of trees from old alleys, even threatening immuno­compromised human individuals.     Analysis of nuclear glucocorticoid receptor-DNA interaction in aged rat liver

    Abstract: In order to contribute to the understanding of mechanisms by which regulatory proteins recognize genetic information stored in DNA, analyses of their interaction with specific nucleotides are usually performed. In this study, the electrophoretic mobility shift assay (EMSA) was applied to analyze the interaction of nuclear proteins from the liver of rats of different age i.e., young (3-month-old), middle- aged (12-month-old) and aged (24-month-old), with radioactively labelled synthetic oligonucleotide analogues, corresponding to GRE. The levels of GRE binding activity were assessed by quantitative densitometric scanning of the autoradiograms. The results showed statistically significant decreasing values of up to 78% and 49% in middle aged and old animals, respectively, compared to young animals (p < 0.05). The specificity of the nuclear proteins-GRE interaction was demonstrated by competition experiments with unlabelled GRE. In a supershift assay, using the antibody BuGR2, it was shown that the GR proteins present in nuclear extracts have a high affinity for the GRE probe. The stabilities of the protein-DNA complexes were analysed and it was concluded that they changed during ageing.U cilju doprinosa razumevanju mehanizama pomoću kojih regulatorni proteini prepoznaju genetičku informaciju koju nosi DNK, analiziraju se njihove interakcije sa specifičnim nukleotidima. U ovom radu je metodom EMSA analizirana interakcija jedarnih proteina iz jetri pacova iz tri starosne grupe (mladi - 3 meseca, srednje doba – 12 meseci i stari – 24 meseca) sa sintetičkim, radioaktivno obeleženim, oligonukleotidnim analogom GRE. Nivo vezujuće aktivnosti GRE je određivan kvantitativno denzitometrijskom autoradiografijom. Rezultati su pokazali da postoji statistički značajan pad vrednosti GRE-vezujuće aktivnosti do 78% kod životinja srednjeg starosnog doba i do 49 % kod starih životinja, u poređenju sa vrednostima dobijenim za mlade životinje (p < 0.05). Specifičnost interakcije jedarnih proteina i GRE je određena eksperimentima kompeticije sa neobeleženim GRE. Korišćenjem antitela BuGR2 pokazano je da je glukokortikoidni receptor protein koji u jedarnom ekstraktu ima najveći afinitet za GRE probu. Analizirana je stabilnost kompleksa protein-DNK i zaključeno je da se menja tokom starenja.nul

    The avarone/avarol quinone/hydroquinone couple shows considerable antitumor activity. In this work, covalent modification of beta-lactoglobulin by avarone and its derivatives as well as by the synthetic steroidal quinone 2,5(10)-estradiene-1,4,17-trione and its derivatives were studied. The techniques for studying chemical modification of beta-lactoglobulin by quinones were: UV/Vis spectrophotometry, SDS PAGE and isoelectrofocusing. SDS PAGE results suggest that polymerization of the protein Occurs. It Could be seen that the protein of 18 kD gives the bands of 20 kD, 36 kD, 40 kD, 45 kD, 64 kD and 128 kD depending on modification agent. The shift of the pl of the protein (5.4) upon modification toward lower values (from pl 5.0 to 5.3) indicated that lysine amino groups are the principal site of the reaction of beta-lactoglobulin with the quinones.Hinonsko/hidrohinonski par avaron/avarol pokazuje značajnu antitumorsku aktivnost. U ovom radu proučavane su kovalentne modifikacije β-laktoglobulina avaronom, sintetičkim steroidnim hinonom 2,5(10)-estradien-1,4,17-trionom i njihovim derivatima. Tehnike za praćenje hemijske modifikacije bile su: UV/Vis spektrofotometrija, SDS PAGE i izoelektrofokusiranje. Rezultati SDS PAGE ukazuju da se dešava polimerizacija proteina.Može se videti da protein od 18 kD daje trake od 20 kD, 36 kD, 40 kD, 45 kD, 64 kD i 128 kD u zavisnosti od agensa za modifikaciju. Pomeranje pI vrednosti proteina (5,4) nakon modifikacije ka nižim vrednostima (od pI 5,0 do 5,3) pokazuje da su amino-grupe lizina glavna mesta reakcije β-laktoglobulina sa hinonima

    Abdominal aortic aneurysm volume and relative intraluminal thrombus volume might be auxiliary predictors of rupture—an observational cross-sectional study

    Objectives: The study aimed to identify differences and compare anatomical and biomechanical features between elective and ruptured abdominal aortic aneurysms (AAAs). Methods: Data (clinical, anatomical, and biomechanical) of 98 patients with AAA, 75 (76.53%) asymptomatic (Group aAAA) and 23 (23.46%) ruptured AAA (Group rAAA), were prospectively collected and analyzed. Anatomical, morphological, and biomechanical imaging markers like peak wall stress (PWS) and rupture risk equivalent diameter (RRED), comorbid conditions, and demographics were compared between the groups. Biomechanical features were assessed by analysis of Digital Imaging and Communication in Medicine images by A4clinics (Vascops), and anatomical features were assessed by 3Surgery (Trimensio). Binary and multiple logistic regression analysis were used and adjusted for confounders. Accuracy was assessed using receiving operative characteristic (ROC) curve analysis. Results: In a multivariable model, including gender and age as confounder variables, maximal aneurysm diameter [MAD, odds ratio (OR) = 1.063], relative intraluminal thrombus (rILT, OR = 1.039), and total aneurysm volume (TAV, OR = 1.006) continued to be significant predictors of AAA rupture with PWS (OR = 1.010) and RRED (OR = 1.031). Area under the ROC curve values and correct classification (cc) for the same parameters and the model that combines MAD, TAV, and rILT were measured: MAD (0.790, cc = 75%), PWS (0.713, cc = 73%), RRED (0.717, cc = 55%), TAV (0.756, cc = 79%), rILT (0.656, cc = 60%), and MAD + TAV + rILT (0.797, cc = 82%). Conclusion: Based on our results, in addition to MAD, other important predictors of rupture that might be used during aneurysm surveillance are TAV and rILT. Biomechanical parameters (PWS, RRED) as valuable predictors should be assessed in prospective clinical trials.     Creating a new academic governance in Poland : assumptions to Law HEI 2.0 (Ustawa 2.0)

    W Polsce trwają zaawansowane prace nad przebudową systemu nauki i szkolnictwa wyższego. Powszechne jest przekonanie środowiska akademickiego, ale również opinii społecznej, że istniejące rozwiązania nie wykorzystują w pełni możliwości kadry naukowo-dydaktycznej, studentów i potencjału polskich uczelni. Powszechnie znane są miary słabości, związane chociażby z pozycją uczelni polskich w rankingach międzynarodowych [Wróblewski 2013]. Również efekty edukacyjne, umasowienie kształcenia i niekorzystne zmiany w etosie akademickim napawają niepokojem [Sułkowski 2016]. Naprawy wymaga zatem zarówno poziom ładu systemowego szkolnictwa i nauki (governance), jak i zarządzanie uczelniami wyższymi w Polsce (management) [Antonowicz, Brdulak, Hulicka i in. 2016]. Strategicznym elementem tej zmiany ma być przyjęcie nowej ustawy regulującej system nauki i szkolnictwa wyższego. Na początku 2017 roku odbyły się prezentacje projektów "Założeń do ustawy 2.0", przygotowanych przez zespoły wyłonione w trybie konkursowym w 2016 roku. Rezultaty pracy to trzy koncepcje zmiany ładu legislacyjnego w polskiej nauce i szkolnictwie wyższym. Każdy z trzech projektów ma swoją dominującą logikę, jednak wszystkie próbują odpowiadać na podobne problemy polskiego systemu nauki i kształcenia wyższego, trafnie zidentyfikowane przez zespoły konkursowe na etapie diagnozy. Posługując się metodą porównawczą, pozwolę sobie poddać analizie propozycje Kolegów, zwracając uwagę przede wszystkim na rozwiązania mające poprawić sytuację polskich uczelni i całego systemu szkolnictwa.In Poland we are working on a modification of higher education system because the current solutions are not satisfactory for Polish education potential, as well as for people engaged in this field and, last but not least, students. Law HEI 2.0 (Ustawa 2.0) is supposed to be the strategic element of change. Polish academics prepared projects to the new regulation and three of them have been chosen. The article is a critical analysis of these proposals and it brings attention to solutions that might improve a situation of Polish higher education and the whole system of education
