Designing democratic institutions for collaborative economic development: a European perspective
Collaborative approaches to local economic development have developed in a number of European countries. However collaborative working presents a new problem for policy makers and public management researchers. The problem is to design an institutional framework for the governance of economic development that provides for anchorage in the democratic system without loosing the benefits of flexible policy design and delivery. This is particularly important in a European context. The European Union has recognised the need for citizens to be more engaged in the governance of public policy at all scales - from local neighbourhoods to the transnational level. This chapter addresses the problem by examining the basic questions that any form of democratic governance design needs to address, and relating this to the case of economic development. The core democratic design questions are: How can legitimacy be secured? In what ways can relevant publics give consent to decisions? Through what means can the institution be held to account? The chapter discusses these three democratic imperatives and shows that different responses to these produce three archetypical governance designs - club, agency and polity. We then explore the way in which the problems of democratic governance have been solved empirically through longitudinal case studies of the expansion of Mainport Rotterdam and the management of economic, environmental, residential and transportation agendas in the Ghent canal area of the Flanders region of Belgium. The analysis shows that although different national and regional political contexts matter, the typology of archetypes offers a way of understanding the overall democratic orientation of a particular governance design as well as offering a basis from which policy makers can create their own solutions
The lethal response to Cdk1 inhibition depends on sister chromatid alignment errors generated by KIF4 and isoform 1 of PRC1
Cyclin-dependent kinase 1 (Cdk1) is absolutely essential for cell division. Complete ablation of Cdk1 precludes the entry of G2 phase cells into mitosis, and is early embryonic lethal in mice. Dampening Cdk1 activation, by reducing gene expression or upon treatment with cell-permeable Cdk1 inhibitors, is also detrimental for proliferating cells, but has been associated with defects in mitotic progression, and the formation of aneuploid daughter cells. Here, we used a large-scale RNAi screen to identify the human genes that critically determine the cellular toxicity of Cdk1 inhibition. We show that Cdk1 inhibition leads to fatal sister chromatid alignment errors and mitotic arrest in the spindle checkpoint. These problems start early in mitosis and are alleviated by depletion of isoform 1 of PRC1 (PRC1-1), by gene ablation of its binding partner KIF4, or by abrogation of KIF4 motor activity. Our results show that, normally, Cdk1 activity must rise above the level required for mitotic entry. This prevents KIF4-dependent PRC1-1 translocation to astral microtubule tips and safeguards proper chromosome congression. We conclude that cell death in response to Cdk1 inhibitors directly relates to chromosome alignment defects generated by insufficient repression of PRC1-1 and KIF4 during prometaphase
The effect of the EUâbrand on citizensâ trust in policies : replicating an experiment
Karens et al. (2016) conducted an experiment to measure the effect of the European Union (EU) brand on citizens' trust in policies. Experiments conducted with economics students in Belgium, Poland, and The Netherlands showed a consistently positive and significant effect of applying the EU brand, on trust in the policies. This study presents seven replications conducted several years after Karens et al.'s experiments. The replications show no significant effects of the EU brand on trust in policies. These findings demonstrate that brand effects may vary over time. To identify a population effect size across all experiments, a meta-analysis was conducted. The meta-analysis shows that-overall-the EU-brand has a small but significant positive effect on citizens' trust in policies. The article tests earlier findings, and discusses intricacies of conducting replications. It elaborates explanations for the results in the replications, and the replication problems with experiments based on evaluative conditioning

The white matter connectome as an individualized biomarker of language impairment in temporal lobe epilepsy.
ObjectiveThe distributed white matter network underlying language leads to difficulties in extracting clinically meaningful summaries of neural alterations leading to language impairment. Here we determine the predictive ability of the structural connectome (SC), compared with global measures of white matter tract microstructure and clinical data, to discriminate language impaired patients with temporal lobe epilepsy (TLE) from TLE patients without language impairment.MethodsT1- and diffusion-MRI, clinical variables (CVs), and neuropsychological measures of naming and verbal fluency were available for 82 TLE patients. Prediction of language impairment was performed using a robust tree-based classifier (XGBoost) for three models: (1) a CV-model which included demographic and epilepsy-related clinical features, (2) an atlas-based tract-model, including four frontotemporal white matter association tracts implicated in language (i.e., the bilateral arcuate fasciculus, inferior frontal occipital fasciculus, inferior longitudinal fasciculus, and uncinate fasciculus), and (3) a SC-model based on diffusion MRI. For the association tracts, mean fractional anisotropy was calculated as a measure of white matter microstructure for each tract using a diffusion tensor atlas (i.e., AtlasTrack). The SC-model used measurement of cortical-cortical connections arising from a temporal lobe subnetwork derived using probabilistic tractography. Dimensionality reduction of the SC was performed with principal components analysis (PCA). Each model was trained on 49 patients from one epilepsy center and tested on 33 patients from a different center (i.e., an independent dataset). Randomization was performed to test the stability of the results.ResultsThe SC-model yielded a greater area under the curve (AUC; .73) and accuracy (79%) compared to both the tract-model (AUC: .54, p < .001; accuracy: 70%, p < .001) and the CV-model (AUC: .59, p < .001; accuracy: 64%, p < .001). Within the SC-model, lateral temporal connections had the highest importance to model performance, including connections similar to language association tracts such as links between the superior temporal gyrus to pars opercularis. However, in addition to these connections many additional connections that were widely distributed, bilateral and interhemispheric in nature were identified as contributing to SC-model performance.ConclusionThe SC revealed a white matter network contributing to language impairment that was widely distributed, bilateral, and lateral temporal in nature. The distributed network underlying language may be why the SC-model has an advantage in identifying sub-components of the complex fiber networks most relevant for aspects of language performance
Spatial and temporal modulation of cell instructive cues in a filamentous supramolecular biomaterial
Supramolecular materials provide unique opportunities to mimic both the structure and mechanics of the biopolymer networks that compose the extracellular matrix. However, strategies to modify their filamentous structures in space and time in 3D cell culture to study cell behavior as encountered in development and disease are lacking. We herein disclose a multicomponent squaramide-based supramolecular material whose mechanics and bioactivity can be controlled by light through co-assembly of a 1,2-dithiolane (DT) monomer that forms disulfide cross-links. Remarkably, increases in storage modulus from âŒ200 Pa to >10 kPa after stepwise photo-cross-linking can be realized without an initiator while retaining colorlessness and clarity. Moreover, viscoelasticity and plasticity of the supramolecular networks decrease upon photo-irradiation, reducing cellular protrusion formation and motility when performed at the onset of cell culture. When applied during 3D cell culture, force-mediated manipulation is impeded and cells move primarily along earlier formed channels in the materials. Additionally, we show photopatterning of peptide cues in 3D using either a photomask or direct laser writing. We demonstrate that these squaramide-based filamentous materials can be applied to the development of synthetic and biomimetic 3D in vitro cell and disease models, where their secondary cross-linking enables mechanical heterogeneity and shaping at multiple length scales
The impact of public branding: an experimental study on the effects of branding policy on citizen trust
Branding has become common in the public sector as brands are increasingly used to influence citizensâ
associations with public organizations and public services. Using experimental research replicated in three European
countries, this article investigates the effect of using the European Union (EU) brand on trust in polices. Experiments
were conducted among economics students in Belgium, Poland, and the Netherlands to test the hypothesis that adding
EU brand elements to policies positively affects trust in those policies. The results show a consistent positive and
significant effect of applying the EU brand to trust in the policies in all countries and for both policies included in the
experimentâeven in the Netherlands, a country characterized by a negative overall EU sentiment. These findings
