    Fluorescence quantum efficiency of CdSe/CdS magic-sized quantum dots functionalized with carboxyl or hydroxyl groups

    AbstractThe present letter reports the thermo-optical properties of functionalized CdSe/CdS magic-sized quantum dots (MSQDs) (sizes 1.9–2.3nm) with carboxyl (R–COOH) or hydroxyl (R–OH) groups in aqueous solutions. Atomic force microscopy and infrared transmittance measurements were used to determine the size of the QDs and to highlight the functionalized groups. Absolute nonradiative quantum efficiency (φ) and radiative quantum efficiency (η) values were determined by applying two techniques: thermal lens (TL) and an alternative method that analyzes the ring patterns generated in a laser beam due to thermally induced self-phase-modulation effects known as the conical diffraction.     Habitual exercise program protects murine intestinal, skeletal, and cardiac muscles against aging

    Aging and aerobic exercise are two conditions known to interfere with health and quality of life, most likely by inducing oxidative stress to the organism. We studied the effects of aging on the morphological and functional properties of skeletal, cardiac, and intestinal muscles and their corresponding oxidative status in C57BL/6 mice and investigated whether a lifelong moderate exercise program would exert a protective effect against some deleterious effects of aging. As expected, aged animals presented a significant reduction of physical performance, accompanied by a decrease of gastrocnemius cross-sectional area and cardiac hypertrophy. However, most interesting was that aging dramatically interfered with the intestinal structure, causing a significant thickening of the ileum muscular layer. Senescent intestinal myocytes displayed many mitochondria with disorganized cristae and the presence of cytosolic lamellar corpuscles. Lipid peroxidation of ileum and gastrocnemius muscle, but not of the heart, increased in aged mice, thus suggesting enhanced oxidative stress.     Dibucaine in Ionic-Gradient Liposomes: Biophysical, Toxicological, and Activity Characterization

    Administration of local anesthetics is one of the most effective pain control techniques for postoperative analgesia. However, anesthetic agents easily diffuse into the injection site, limiting the time of anesthesia. One approach to prolong analgesia is to entrap local anesthetic agents in nanostructured carriers (e.g., liposomes). Here, we report that using an ammonium sulphate gradient was the best strategy to improve the encapsulation (62.6%) of dibucaine (DBC) into liposomes. Light scattering and nanotracking analyses were used to characterize vesicle properties, such as, size, polydispersity, zeta potentials, and number. In vitro kinetic experiments revealed the sustained release of DBC (50% in 7 h) from the liposomes. In addition, in vitro (3T3 cells in culture) and in vivo (zebrafish) toxicity assays revealed that ionic-gradient liposomes were able to reduce DBC cyto/cardiotoxicity and morphological changes in zebrafish larvae. Moreover, the anesthesia time attained after infiltrative administration in mice was longer with encapsulated DBC (27 h) than that with free DBC (11 h), at 320 μM (0.012%), confirming it as a promising long-acting liposome formulation for parenteral drug administration of dibucaine.Fil: Couto, Verônica M..     Capacity for the management of kidney failure in the International Society of Nephrology Latin America region:Report from the 2023 ISN Global Kidney Health Atlas (ISN-GKHA)

    Successful management of chronic kidney disease (CKD) in Latin America (LA) continues to represent a challenge due to high disease burden and geographic disparities and difficulties in terms of capacity, accessibility, equity, and quality of kidney failure care. Although LA has experienced significant social and economic progress over the past decades, there are still important inequities in healthcare access. Through this third iteration of the International Society of Nephrology Global Kidney Health Atlas, the indicators regarding kidney failure care in LA are updated. Survey responses were received from 22 (71%) of 31 countries in LA representing 96.5% of its total population. Median CKD prevalence was 10.2% (IQR 8.4 - 12.3), median CKD disability-adjusted life year (DALYs) were 753.4 days (IQR 581.3 -1,072.5), and median CKD mortality was 5.5% (IQR 3.2 - 6.3). Regarding dialysis modality, hemodialysis (HD) continued to be the most utilized therapy, while peritoneal dialysis (PD) reached a plateau and kidney transplantation increased steadily over the past 10 years. In 20 (91%) countries, >50% of people with kidney failure could access dialysis; and in only 2 (9%) countries, people who had access to dialysis could initiate dialysis with PD. A mix of public and private systems collectively funded most aspects of kidney replacement therapy (KRT; dialysis and transplantation) with many people incurring up to 50% of out-of-pocket costs. Few LA countries had CKD/KRT registries and almost no acute kidney injury (AKI) registries were reported. There was large variability in the nature and extent of kidney failure care in LA mainly related to countries’ funding structures and limited surveillance and management initiatives

    Quality Of Life Assessment In Children With Cerebral Palsy

    Objectives: Evaluate the QoL of children with CP followed at Núcleo de Tratamento e Estimulação Precoce, in Fortaleza, Ceará, Brazil; identify the most affected domains of the Pediatric Quality of Life Inventory. Methods: This was a cross-sectional, of which target population consisted of parents of children with CP (N = 62). Data collection was carried out by applying the socioeconomic questionnaire and the Health-related Quality of Life tool – HRQoL-PedsQL version 4.0, module applied to parents. Data analysis was performed with SPSS statistical software, version 23.0. Results: The results after transformation of scores into a scale of 0 to 100 in PedsQL, the data showed statistical difference between QOL domains except for physical capacity and social aspect and in descending order of impairment, the domains were related to school activity, the emotional aspect, physical capacity and the social aspect. Conclusions: Given the above, to attain improvement in QoL indicators, it is reasonable to propose interdisciplinary intervention among healthcare professionals in order to develop improved strategies aiming to promote early integration of children with CP in schools, to obtain significant results

    Antioxidant dietary deficiency induces caspase activation in chick skeletal muscle cells

    Apoptosis and necrosis are two distinct forms of cell death that can occur in response to different agents and stress conditions. In order to verify if the oxidative stress induced by dietary selenium and vitamin E deficiencies can lead muscle cells to apoptosis, one-day-old chicks were reared using diets differing in their vitamin E (0 or 10 IU/kg) and selenium (0 or 0.15 ppm) supplementation. Chick skeletal muscle tissue was obtained from 28-day-old animals and used to verify apoptosis occurrence based on caspase activity detection and DNA fragmentation. Antioxidant deficiency significantly increased caspase-like activity assessed by the hydrolysis of fluorogenic peptide substrates (Abz-peptidyl-EDDnp) at lambdaexc = 320 nm and lambdaem = 420 nm. Proteolytic activation was not accompanied by typical internucleosomal DNA fragmentation detected by field inversion gel electrophoresis. Although the general caspase inhibitor N-benzyloxycarbonyl-Val-Ala-Asp(O-Me) fluoromethyl ketone (Z-VAD-fmk) (0 to 80 muM) did not block caspase-like activity when preincubated for 30 min with muscle homogenates, the hydrolyzed substrates presented the same cleavage profile in HPLC (at the aspartic acid residue) when incubated with the purified recombinant enzyme caspase-3.     Intestine of dystrophic mice presents enhanced contractile resistance to stretching despite morphological impairment

    Protein dystrophin is a component of the dystrophin-associated protein complex, which links the contractile machinery to the plasma membrane and to the extra-cellular matrix. Its absence leads to a condition known as Duchenne muscular dystrophy (DMD), a disease characterized by progressive skeletal muscle degeneration, motor disability, and early death. in mdx mice, the most common DMD animal model, loss of muscle cells is observed, but the overall disease alterations are less intense than in DMD patients. Alterations in gastrointestinal tissues from DMD patients and mdx mice are not yet completely understood. Thus, we investigated the possible relationships between morphological (light and electron microscopy) and contractile function (by recording the isometric contractile response) with alterations in Ca2+ handling in the ileum of mdx mice. We evidenced a 27% reduction in the ileal muscular layer thickness, a partial damage to the mucosal layer, and a partial damage to mitochondria of the intestinal myocytes. Functionally, the ileum from mdx presented an enhanced responsiveness during stretch, a mild impairment in both the electromechanical and pharmacomechanical signaling associated with altered calcium influxinduced contraction, with no alterations in the sarcoplasmic reticulum Ca2+ storage (maintenance of the caffeine and thapsigargin-induced contraction) compared with control animals.
