Quality of doctoral research supervision - design of a framework to evaluate and monitor the process
internacional. Ambos permitiram contextualizar o fenómeno, perceber as
puderam ser categorizadas segundo (i) o enquadramento do processo
estudantes e supervisores. As competências integradas nos questionários
qualidade integrado e sistematizado, considerando o enquadramento teórico e
doutoramento devem demonstrar e desenvolver.Due to a theoretical, conceptual and empirical emptiness within the Portuguese
research context on studies regarding doctoral research supervision, this study
aims to shed light on that phenomenon from an integrated and in-depth
perspective. Specifically, we have the main objective of designing a framework
on the quality of doctoral research supervision, in particular in what concerns
the quality profiles of supervisors and students in terms of the transferable and
supra-disciplinary competences they should possess and further enhance.
Consequently, at a medium-term, it will be possible not only to identify
institutional and practical recommendations, but also to design a suitable
evaluation and monitoring process on this practice.
Considering the Portuguese context and the main goal of this research, it has
been revisited (i) political discourses, mainly at European level, on Higher
Education and the 3rd cycle of Bologna, and (ii) international research-based
discourses. This in-depth analysis has allowed us to contextualise the
phenomenon under study, to understand current tendencies, to collate
contributions from different models and frameworks that exist worldwide, and to
plan our data collection, at a more specific level.
This research was carried out at the University of Aveiro. It can be described as
mainly qualitative in nature and it follows the method of case study. It has thus
been taken into consideration the ‘voices’ of doctoral students and supervisors
from this institution regarding what should integrate the quality of the doctoral
research supervision process.
The process of data collection started with exploratory semi-structured
interviews directed to experts on pedagogical supervision – the theme that is
semantically and epistemologically closer to doctoral research supervision. This
was the first empirical approach to this particular issue within the Portuguese
context. Simultaneously, it was important to better define the dimensions that
should be integrated within the instrument of data collection of the following
moment.
Afterwards, focus groups with doctoral students and supervisors have been
conducted. The conceptions that were shared by the participants were
categorised and semantically grouped considering (i) the broader context on
the doctoral research and supervision process, namely taking into account the
nature and value the participants attributed to the doctorate, and the changing
process they experienced; (ii) the quality of the doctoral research and
supervision process, taking into account intrinsic aspects, such as the
identification of the quality profiles of students and supervisors in terms of the
competences they should possess, as well as extrinsic aspects; (iii) problems
to the doctoral research and supervision process, also taking into consideration
intrinsic and extrinsic aspects.
Questionnaires directed to doctoral students and supervisors were applied in
the last phase of data collection, which was specifically focused on the quality
profiles of students and supervisors. As such, the questionnaires were
constituted by the competences that have emerged in the focus groups as
essential to integrate the quality profiles. In the questionnaires, the participants
had to attribute their level of agreement regarding each competence.
Therefore, the results shed light on the possibility of establishing a hierarchy of
competences with the main goal of better defining and characterising students
and supervisors’ quality profiles. It can be concluded that there are some
variables that may influence the order of the competences within the hierarchy.
This fact reinforces that the supervision process must be carefully
contextualised and understood when an evaluation and monitoring process is
designed and carried out. Nevertheless, when analysing the results it may also
be concluded the existence of competences that seem ‘free’ from influences.
However, more in-depth studies must be carried out to clarify this assumption.
Taking into consideration the overall research path, it is presented, at the end
of the study, an integrated systematised quality framework, considering the
inputs of the theoretical and empirical parts of our thesis. Moreover, it is
presented an articulated system of competences that refers to the quality
profiles doctoral students and supervisors should demonstrate and enhance

ABSTRACT: The main purpose of this work is to characterise, according to several points of view, the first group of the students over 23 years old, enrolled at the University of Aveiro in the academic year of 2006-07.
The theoretical framework of this study will be explored in an articulated manner, in order to better analyse and discuss the data, collected in a specific context: the University of Aveiro (Portugal).
Therefore, in the first part, we will start to design a more general and transversal context, based on the ideas of the complexity and supercomplexity
of today’s world, to delineate the demands of our society. Then, we will focus on certain topics, such as: Higher Education and the Bologna Process, Lifelong Learning and the Lisbon Strategy, taking into account their main principles which may be dialogued. When reflecting about certain documents, we will approach them accordingly to certain themes like the openness of Higher
Education to new publics, the emphasis put on equity, and the creation of new opportunities. Also, we will make a first approach to the characteristics of nontraditional adult students, our target population, accordingly to a theoretical
perspective, based on the literature review.
In the second part, we will describe our empirical study. After describing the research methodology, we will analyse and discuss the results collected
through questionnaires, motivation letters and semi-oriented interviews.
Quantitative and qualitative methods will be used. The data will be intersected with the main goal of discussing this subject through different but
interconnected perspectives, so that we can reach credible conclusions.
This may be a first contribute to characterise the group of non-traditional adult student over 23 years old of the University of Aveiro
Dissecting the role of lipocalin in Plasmodium falciparum
[Excerpt] Malaria, caused by Plasmodium parasites, is an infectious disease responsible for millions
of cases and deaths yearly. Despite the efforts, the disease control has been continuedly
threatened by the resilient capacity of the parasite to acquire resistance to antimalarial
drugs treatment, which target the symptomatic intraerythrocytic developmental cycle. This work was supported by NORTE-01-0145- FEDER-028178 funded by NORTE 2020 Portugal Regional Operational Program under PORTUGAL 2020 Partnership Agreement through the European Regional Development Fund and the by Fundação para a Ciência e Tecnologia (FCT). Optical spectrophotometry into hemozoin: towards sensitive malaria diagnosis
The lack of prompt and sensitive diagnosis hampers malaria control and elimination,
highlighting the need for next generation technology alternative for the established optical
microscopy and Rapid Diagnostic Tests (RDTs) as means of in situ point-of-care malaria
parasite detection. Hemozoin (Hz), a byproduct of hemoglobin (Hb) degradation inside
human infected red blood cells (RBCs), has been extensively explored as a malaria
biomarker. Along with malaria parasite maturation inside the RBC, Hb and Hz proportion
is inversely related, which originates specific optical spectra of healthy and infected
samples. Herein, we characterized the optical spectra of Plasmodium falciparum-infected
RBCs, aiming the development of an innovative diagnostic device, detecting malaria
without finger prick blood sampling, measuring directly in patients’ skin. Absorbance and
reflectance spectrophotometry demonstrate their potential by increasing the limit of
detection (LoD: 12 parasites/μL of RBC) when compared with microscopy or RDT (LoD:
50-200 parasites/μL of RBC). This sensitivity, coupled with the possible integration into
a low-cost, fast, and non-invasive diagnostic device meets the growing clinical demands
for malaria control and elimination. This work was supported by Fundação para a Ciência e Tecnologia (FCT) national funds,

Deeper into hemozoin formation pathway of Plasmodium falciparum
Plasmodium falciparum malaria remains a public health problem, accounting for half
millions of lives yearly. Artemisinin-combination therapies are the front-line treatment
against malaria, which efficacy is now threatened by parasite’s-acquired resistance. Most
antimalarial drugs target the symptomatic intra-erythrocytic stage of infection. During
parasite development inside the human erythrocytes, parasites digest hemoglobin as a
source of nutrients. This causes the release of toxic free heme, which the parasite
detoxifies into inert crystals, called hemozoin. It is known that parasites with
abnormalities in this pathway are committed to death. However, despite its importance,
the orchestration of hemoglobin digestion into hemozoin formation has not been entirely
resolved, and its players have not been identified. Through reverse genetics, we explored
the role of P. falciparum aspartic proteases and other genes thought to be involved in the
formation of hemozoin and their capacity to modulate drug response. We believe that a
deeper understanding of this pathway is of great interest and relevant for the identification
of new targets, both for diagnosis and for the development of antimalarial drugs.
Furthermore, it will also provide insights into drug resistance, overall contributing for
better malaria control and management. This work was supported by Fundação para a Ciência e Tecnologia (FCT) national funds, under the national support to R&D units grant, through the reference project UIDB/04436/2020 and UIDP/04436/2020. Multilayer thin-film optical filters for reflectance-based malaria diagnostics
Malaria diagnosis relies on optical microscopy and/or rapid diagnostic tests based on detecting specific malaria antigens. The clinical sensitivity of these methods is highly dependent on parasite density, with low levels of detection at low parasite density, challenging the worldwide malaria elimination efforts. Therefore, there is a need for diagnostic methods with higher sensitivity, demanding innovative diagnostics devices able to detect malaria at low parasite density and at early stages of the disease. We propose an innovative optical device for malaria diagnosis, based on optical reflectance spectrophotometry, for the detection of parasites through the quantification of haemozoin. For this purpose, a set of eight thin-film optical filters, based on multilayer stacks of MgO/TiO2 and SiO2/TiO2 thin-films, with high transmittance and low full width at half maximum (FWHM) at specific wavelengths, was designed and fully characterized (both numerically and experimentally). A preliminary assessment of its potential to reconstruct the original spectra of red blood cells was performed, both in uninfected and Plasmodium falciparum-infected samples. The obtained results show that, although the experimental filters have a non-ideal performance characteristic, they allow us to distinguish, based on only 8 discrete points in the optical spectrum, between healthy and malaria infected samples, up to a detection limit of 12 parasites/μL of red blood cells. Those results enhance the potential of using such a device for malaria diagnostics, aiming for non-invasiveness.Project NORTE-01-0145-FEDER-028178 funded by NORTE

Characterization of the malaria parasite optical features for development of a non-invasive diagnostic device
[Excerpt] Early and accurate malaria diagnosis is critical for the disease control and elimination [1].
Microscopy and/or immuno-rapid tests remain the standard diagnosis [2, 3], nevertheless it
requires a skin puncture for blood sampling and not sensitive enough for reliable detect lowdensity parasitemias, urging the need to develop more sensitive and non-invasive tools.
Symptoms of the disease starts when parasites infect the red blood cell (RBC), suffering
biochemical and morphological changes [4]. Parasite survival is dependent on hemozoin (Hz)
formation as a by-product of heme detoxification process of the parasite upon haemoglobin
(Hb) degradation and therefore a good unique feature to identify parasites presence in
patients’.
Taking advantage of the fact that the Hz and Hb molar extinction coefficients differ significantly,
especially at certain wavelengths [4, 5], and their proportion is inversely related upon parasite
maturation inside the RBC, each stage of malaria is characterized by specific absorbance and
reflectance spectra, according to the Hb/Hz concentrations on the iRBC.
