Estudo da estabilidade do éster metilanidroecgonina, marcador de uso do crack, benzoilecgonina e cocaína em urina humana
BACKGROUND: Typically, urine and other biological tissues have been analyzed for cocaine (COC) and/or metabolites to detect COC usage. COC undergoes numerous biotransformation and degradation reactions. Crack smokers are exposed to anhydroecgonine methyl ester (AME), which can be used as an analytical marker for crack smoking. The stability of this analyte in human urine has not been studied. In the body, COC is rapidly converted to metabolites by enzymatic and chemical processes, the major urinary metabolite being benzoylecgonine (BE). OBJECTIVES: This study was carried out in order to determine the effects of time and temperature on the stability of cocaine/crack metabolites in human urine. METHODS : The stability of AME, BE and COC in urine was investigated using samples of urine stored in freezers and refrigerators. The analytes were extracted from urine using a solid-phase extraction technique and analyzed by gas chromatography-flame ionization detection method. RESULTS: COC concentrations decreased while BE concentrations increased. AME concentrations remained stable. CONCLUSIONS: The temperature and the duration of storage are decisive in COC hydrolyzing. This study suggests that AME concentrations are not correlated to either storage duration or with storage temperature and AME is more stable than COC.CONTEXTO: Cocaína (COC) e/ou metabólitos tem sido analisados em urina e outros fluidos biológicos para se determinar o uso de COC. A COC está sujeita a numerosas reações de biotransformação e degradação. Indivíduos que fumam crack estão expostos ao éster metilanidroecgonina (AME), que pode ser empregado como marcador de uso desta droga. Não há referências na literatura a respeito da estabilidade deste analito em urina humana. No organismo a COC é rapidamente biotransformada em outros metabólitos por meio de processos químicos e enzimáticos e o principal metabólito urinário é a benzoilecgonina (BE). OBJETIVOS: O objetivo deste estudo foi determinar o efeito do tempo e da temperatura na estabilidade da COC, BE e do AME em urina humana. MÉTODOS: A estabilidade do AME, BE e COC em urina foi investigada por intermédio do armazenamento da urina em freezer e em geladeira. Os analitos foram extraídos pela técnica de extração em fase sólida e analisados por cromatografia gasosa acoplada ao detector por ionização em chama. RESULTADOS: As concentrações de COC decresceram enquanto as BE aumentaram. As concentrações de AME se mantiveram estáveis. CONCLUSÕES: A temperatura e o tempo de armazenamento são decisivos na hidrólise da COC. MELHORIA NA EFICIÊNCIA DA CLARIFICAÇÃO DE SUCO DE MARACUJÁ PELA COMBINAÇÃO DOS PROCESSOS DE MICROFILTRAÇÃO E ENZIMÁTICO
Este trabalho teve como objetivo a obtenção de suco
clarificado de maracujá, utilizando a técnica de
microfiltração. Introduziu-se a etapa de tratamento
enzimático antes da microfiltração, visando aumentar
a eficiência do processo pela redução da viscosidade
e do teor de polpa do suco. Foram testadas três
diferentes enzimas hidrolíticas em três níveis de
concentração. Os resultados não mostraram
diferenças significativas quanto ao Brix, pH e acidez
das amostras de suco hidrolisado em relação ao suco
integral. Observou-se efeito positivo da ação das
enzimas na redução da viscosidade e do teor de polpa.
A microfiltração foi realizada em sistema de membrana
tubular com tamanho médio de poro equivalente a
0,3 µm e pressão transmembrana de 1,5 bar. O suco
permeado apresentou-se límpido, isento de polpa e com
qualidade e sanidade microbiológica. Tais resultados
comprovaram a eficiência da microfiltração para
clarificação e estabilização de sucos de frutas.
IMPROVEMENT IN THE EFFICIENCY OF PASSION FRUIT JUICE
CLARIFICATION BY THE COMBINATION OF MICROFILTRATION AND
ENZYMATIC PROCESSES
Abstract
This work had as objective the obtention of clarified passion fruit juice by using the
microfiltration technique. A step of enzymatic treatment was introduced before
microfiltration, aiming to enhance process efficiency by reduction of viscosity and
pulp content of the juice. Three different hidrolytic enzymes were tested in three
concentration levels. The results didnt demonstrate significant differences in Brix,
pH and acidity of the hidrolyzed juice samples in relation to the concentrated juice.
Positive effect was observed in the action of the enzymes in the reduction of
viscosity and pulp content. The microfiltration was realized in a tubular membrane
system with average size of the pore equivalent to 0.3 m and transmembrane
pressure of 1.5 bar. The permeate juice was clear, free from pulp and with good
micribiological quality. Such results proved the efficiency of microfiltration for
clarification and stabilization of fruit juices
UTILIZAÇÃO DO RETENTADO DA ULTRAFILTRAÇÃO DO SUCO DE ACEROLA NA ELABORAÇÃO DE LICOR
Estudou-se a preparação de licor mediante maceração
alcoólica da fração retida no processo de ultrafiltração do
suco de acerola, em sistema de membranas tubulares com
peso molecular de corte de 30-80 kDa, área filtrante de
0,05 m2 e pressão de 200 kPa. As características sensoriais
e nutricionais do licor foram avaliadas tendo-se como
referência o licor preparado a partir da polpa de acerola
fresca pelo mesmo processo de fabricação. Os licores foram
preparados partindo-se de mistura 1:1 de polpa de acerola
ou do retentado da ultrafiltração e álcool de milho. Após 15
dias de maceração adicionou-se açúcar até concentração
de 250 g/L e o teor alcoólico foi ajustado para 18 oGL. Os
licores apresentaram teores de vitamina C semelhantes, na
faixa de 0,400 g/100 g, cerca de seis vezes mais que a dose
diária recomendada para um adulto. As análises
microbiológicas da polpa de acerola, do retentado e dos
licores atenderam aos padrões estabelecidos pela legislação
vigente. O aroma alcoólico tornou-se mais evidenciado no
licor de retentado enquanto que o aroma e o sabor frutal
mostraram-se mais pronunciados no licor de polpa de acerola.
A percepção do aroma doce também foi bastante
pronunciada no licor de polpa de acerola. A pungência do
licor de retentado mostrou-se menor que a do licor de polpa
de acerola. Os resultados obtidos demonstraram que o licor
elaborado a partir do retentado da ultrafiltração do suco de
acerola apresenta características similares às do licor
preparado com a própria polpa.
UTILIZATION OF ACEROLA JUICE ULTRAFILTRATION RETENTATE IN A LIQUEUR
ELABORATION
Abstract
The preparation of a liqueur by alcoholic maceration of the fraction retained in the
acerola juice ultrafiltration process, in tubular membrane system with molecular weight
of 30-80 kDa cut-off, filtration area of 0.05 m2 and 200 kPa of transmembrane pressure
was studied. The nutritional and sensory characteristics of the liqueur were evaluated
having as standard the liqueur prepared from fresh acerola pulp by the same fabrication
process. Liqueurs were prepared from a 1:1 mixture of acerola pulp or retentate and
corn alcohol. After 15 days of maceration, sugar was added until a concentration of
250 g/L and alcohol grade was adjusted to 18 oGL. The vitamin C content of both liqueurs were similar, about 0,400 g/100 g, corresponding to almost six times the vitamin C
recommended dietary allowance for an adult. Microbiological analyses of acerola pulp,
retentate and liqueurs attended the standards established by legislation. Alcoholic aroma
was more pronounced in the retentate liqueur while the fruit aroma and flavor were
more pronounced in the pulp liqueur. The perception of the sweet aroma was also well
pronounced in the acerola pulp liqueur. The pungency of the retentate liqueur was lower
than the acerola pulp liqueur. The results obtained showed that the liqueur elaborated
from the acerola juice ultrafiltration retentate present similar characteristics to the liqueur
prepared from the pulp
Physical, chemical and microbiological characteristics of cocoa pulp
O objetivo deste trabalho foi caracterizar físico-química e microbiologicamente a polpa de cacau (Theobroma cacao L.), comercializada na região sudeste da Bahia, visando contribuir para a melhoria da qualidade do produto. A polpa foi extraída após diferentes tratamentos de limpeza dos frutos, os quais pouco influíram na qualidade microbiológica da polpa. Todas as amostras apresentaram baixa contagem de coliformes totais e fecais. Na contagem total em placas foram determinadas até 104 UFC/g, e a contagem de bolores e leveduras também esteve acima de 3x103 UFC/g. A polpa de cacau in natura apresentou teor de sólidos solúveis na faixa de 17o a 19o Brix, acidez em torno de 1,5 (solução 0,01 N) e pH entre 3,1 e 3,6. A viscosidade aparente foi de 96 cP, à temperatura de 25oC, para uma taxa de deformação de 200 s-1. As provas qualitativas de peroxidase e polifenoloxidase apresentaram resultado positivo.The objective of this research was to characterize the cocoa (Theobroma cacao L.) pulp traded in Southeast Bahia from the physical, chemical and microbiological standpoints, aiming to improve its quality. The pulp was extracted after different cleaning treatments of the fruit, which did not show any significant influence on the microbiological quality of the pulp. All samples showed low values for total and fecal coliforms; the total plate count determinations reached up to 104 CFU/g and the mold and yeast count was above 3x103CFU/g. The characteristics of the natural cocoa pulp were: soluble solids between 17oand 19o Brix, acidity around 1.5 (0,01 N solution), pH between 3.1 and 3.6, apparent viscosity of 96 cP, at 25°C and deformation rate of 200 s-1. The results were also positive for qualitative determinations of peroxidase and polyphenoloxidase activities
Suco de acerola microfiltrado: avaliação da vida-de-prateleira Microfiltered acerola juice: evaluation of shelf life
Este trabalho teve como objetivo estudar a estabilidade física, química e microbiológica do suco clarificado de acerola, obtido por microfiltração. O suco microfiltrado foi recolhido em um sistema fechado, previamente esterilizado, sendo, em seguida, transferido para garrafas de polietileno tereftalato (PET) ou de vidro, também previamente sanificadas, dentro de uma capela de fluxo laminar. O suco clarificado foi mantido em geladeira (4ºC) e à temperatura ambiente (30ºC) por um período de 90 dias. A avaliação da vida-de-prateleira do suco de acerola clarificado mostrou que o armazenamento em geladeira, tanto em garrafas de vidro quanto de PET, possibilita a preservação das características do suco, não tendo havido diferenças significativas entre os dois tipos de embalagem.<br>This work had as objective to study the physical, chemical and microbiological stability of clarified acerola juice, obtained by membrane process. Microfiltered juice was collected in a sterilized closed system and transferred to glass or PET bottles, also previously sterilized, in a laminar cabinet. Products were maintained under refrigeration (4ºC) and room (30ºC) temperatures during 90 days. Changes in phenolic composition, ascorbic acid and antioxidant capacity in cashew apple (
Introduction. Cashew apple
is a rich source of sugars, vitamin C and polyphenols. In spite
of its nutritional value, this pseudo-fruit has been left unexploited
to a large extent in the crop-growing areas. Some reports of the
chemical characteristics of cashew apple have been published. However,
nothing is known about the changes in the composition of its bioactive compounds
in the course of ripening. Materials and methods. Cashew
apples at three different maturity stages were examined with respect
to their ascorbic acid content, phenolic compounds and antioxidant
capacity. Ascorbic acid was quantified by HPLC. Phenolic compounds
were identified and quantified by using HPLC-ESI-MS/MS by comparison
with authentic standard compounds. The antioxidant capacity was
measured by TOSC assay against peroxyl radicals and peroxynitrite.
Results. Amounts of identified phenolic compounds were
the highest in unripe cashew apple and decreased in the course of
ripening. Myricetin 3-O-rhamnoside, quercetin 3-O-galactoside and quercetin
3-O-rhamnoside turned out to be the main flavonoids in all maturity
stages. The antioxidant capacity and the concentration of ascorbic
acid increased in the course of ripening. The antioxidant activity
was considerably influenced by ascorbic acid, more than by the content of
phenolic compounds. Conclusion. This study provides, for
the first time, information on changes in bioactive compounds and
the antioxidant capacity in cashew apple during ripening. A dietary
or technological exploitation of ascorbic acid is useful in the
ripe condition. The unripe pseudo-fruits are a good source for the extraction
of polyphenols with regard to possible food technological purposes
or the preparation of food supplements
