    Metagenomic Analysis of Bacterial Community Structure and Dynamics of a Digestate and a More Stabilized Digestate-Derived Compost from Agricultural Waste

    Recycling of different products and waste materials plays a crucial role in circular economy, where the anaerobic digestion (AD) constitutes an important pillar since it reuses nutrients in the form of organic fertilizers. Knowledge about the digestate and compost microbial community structure and its variations over time is important. The aim of the current study was to investigate the microbiome of a slurry cow digestate produced on a farm (ADG) and of a more stabilized digestate-derived compost (DdC) in order to ascertain their potential uses as organic amendments in agriculture. The results from this study, based on a partial fragment of 16S bacterial rRNA NGS sequencing, showed that there is a greater microbial diversity in the DdC originated from agricultural waste compared to the ADG. Overall, the existence of a higher microbial diversity in the DdC was confirmed by an elevated number (1115) of OTUs identified, compared with the ADG (494 OTUs identified). In the DdC, 74 bacterial orders and 125 families were identified, whereas 27 bacterial orders and 54 families were identified in the ADG. Shannon diversity and Chao1 richness indexes were higher in DdC samples compared to ADG ones (Shannon: 3.014 and 1.573, Chao1: 68 and 24.75; p< 0.001 in both cases). A possible association between the microbiome composition at different stages of composting process and the role that these microorganisms may have on the quality of the compost-like substrate and its future uses is also discussed

    Osteochondroma as a cause of scapular winging in an adolescent: a case report and review of the literature

    INTRODUCTION: Winged scapula is defined as the prominence of the medial border of the scapula. The classic etiopathology of scapular winging are injuries to the spinal accessory or long thoracic nerves resulting respectively in trapezius and serratus anterior palsy. To the best of our knowledge, there are only few reports of scapular lesions being mistaken for winging of the scapula. We report a rare case of a large scapular osteochondroma arising from the medial border and causing a pseudowinging of the scapula. CASE PRESENTATION: A 17-year-old Caucasian boy came to us complaining about a winged left scapula. The patient had a complete painless range of motion, but a large hard bony swelling was palpable along the medial border of his left scapula. A grating sensation was felt when his arm was passively abducted and/or elevated causing discomfort. A lesion revealed on X-rays was diagnosed as an osteochondroma of the medial border of his scapula. After preoperative examinations, he underwent open surgery in order to remove the lesion. A histological examination confirmed the clinical diagnosis of osteochondroma. A clinical examination 3 months later showed a full and painless range of motion, the absence of the grating sensation during passive abduction and elevation and the complete disappearance of his left shoulder deformity. After 2 years of follow-up, there were no clinical or radiological signs of recurrence. CONCLUSIONS: Despite its rarity osteochondroma should be considered in the differential diagnosis for any adolescent presenting with a winging of the scapula

    High-level tolerance to triclosan may play a role in Pseudomonas aeruginosa antibiotic resistance in immunocompromised hosts: evidence from outbreak investigation

    <p>Abstract</p> <p>Background and methods</p> <p><it>Pseudomonas aeruginosa </it>is a major infectious threat to immunocompromised patients. We recently reported a fatal epidemic of multidrug-resistant <it>P. aeruginosa </it>in an onchoematology unit, linked to massive contamination of a triclosan-based disinfectant. The aim of this study is to evaluate the antimicrobial activity of triclosan and chlorhexidine digluconate against the epidemic strain of <it>P. aeruginosa</it>, to confirm the hypothesis that the soap dispenser acted as a continuous source of the infection during the outbreak, and to explore the potential role of triclosan in increasing the level of resistance to selected antibiotics.</p> <p>Susceptibility tests and time-kill assays for disinfectans were performed using two commercial formulations containing triclosan and chlorhexidine digluconate, respectively. Antibiotic susceptibility testing was performed by the broth microdilution method.</p> <p>Findings</p> <p>The <it>P. aeruginosa </it>epidemic strain exhibited an extremely high level of triclosan resistance (apparent MIC = 2,125 mg/L), while it was markedly susceptible to chlorhexidine digluconate (apparent MIC = 12.5 mg/L). Upon gradual adaptation to triclosan, the epidemic strain survived for a long period (> 120 h) in the presence of 3,400 mg/L (equivalent to 1.6 × MIC) of triclosan, concomitantly increasing the resistance to six antibiotics that are typical substrates of drug efflux pumps of the resistance nodulation division family. This effect was reversed by efflux pump inhibitors.</p> <p>Conclusions</p> <p>The epidemic <it>P. aeruginosa </it>strain was resistant to triclosan and its previous exposure to triclosan increases antibiotic resistance, likely through active efflux mechanisms. Since <it>P. aeruginosa </it>can become tolerant to elevated triclosan concentrations, the use of triclosan-based disinfectants should be avoided in those healthcare settings hosting patients at high risk for <it>P. aeruginosa </it>infection.</p

    Garlic (Allium spp.) viruses: detection, distribution and remediation attempts in a European garlic collection

    Garlic is an important vegetable crop in numerous countries used as food and natural based medicine. Similar to the majority of vegetatively propagated plants, garlic may be affected by several viruses that can cause severe crop losses. The present study aimed to screen 105 garlic accessions (mother plants) from 5 European countries (Germany, Czech Republic, Poland, Italy, and France) for possible presence of Onion yellow dwarf virus (OYDV), Leek yellow stripe virus (LYSV), Garlic common latent virus (GCLV) and Shallot latent virus (SLV). The occurrence of three Allexiviruses (GarV-A, GarV-B and GarV-C) in mixed assays was also investigated. Meristem-tip culture assays were performed in order to attempt eradication of the studied viruses. Garlic viruses identification was made by ELISA and RT-PCR. ELISA outcomes showed that all 105 garlic accessions were infected by different virus combinations. The OYDV and LYSV were identified, by ELISA, in all countries at 96% and 88,6% respectively and by RT-PCR at 99% and 96%. Furthermore, GCLV and SLV were detected by ELISA in about 88% and by RT-PCR at 89% and 90%, respectively with the exception of the studied Allexiviruses which were not amplified by RT-PCR with ALLEX1/ALLEX2 primers. Smaller meristem size (0,3-1,5 mm) led to better virus elimination efficiency (29%) compared to 8% obtained for the larger size (2-2,5 mm). The outcomes were opposite (16% vs. 90%) for plants regeneration. Virus elimination efficiency was linked to the virus type, e.g., OYDV and LYSV were eradicated at 90% while GCLV and Allexiviruses were difficult to eliminate (57,4% and 55,6% of eradication). Given the economic relevance of garlic crops worldwide and the frequently reported incidence of viral infections, it is important to make virus-free germplasm available. Therefore, investigating the garlic germplasm sanitary status and constantly improving it is of crucial importance aiming to increase the overall garlic production

    Effect of irrigation regimes and artificial mycorrhization on insect pest infestations and yield in tomato crop

    A 2-year field experiment was carried out to test the effect of root symbionts treatments in combination with different watering levels on tomato yield and pest infestation rates. A split-plot experimental design was followed, where the main treatments were three irrigation regimes, and the subplot factor was the mycorrhizal treatment (two mycorrhizal treatments [M1 and M2] and the control). The M1 treatment consisted in the use of a commercial preparation (Micosat F; CCS Aosta, Italy) containing a mixture of vesicular-arbuscular mycorrhizae (VAM), bacteria of the rhizosphere and saprophytic fungi, while M2 treatment used only arbuscular mycorrhizal fungi. Insect pests sampled in the field during the 2 years of experiment were all sap-feeders (Trialeurodes vaporariorum, Macrosiphum euphorbiae, Frankliniella occidentalis and an unidentified species of leafhopper). Results did not show any impact of root symbionts on pests, whereas water deficit significantly reduced plant infestation rates. Both mycorrhization treatments and water supply resulted in a significantly positive effect on crop yield

    Evaluation of Resistance to TSWV and Agronomic Behaviour of Some TSWV-Resistant Tomato Genotypes in Southern Italy

    A two-year research was carried out in Ofanto valley (Basilicata region, southern Italy) in 2006-2007, for evaluating agronomic behaviour of new processing tomato genotypes resistant to tomato spotted wilt virus (TSWV) and ascertaining their effective resistance to the virus. Five TSWV-resistant genotypes (cultivars and lines) were compared in each year the TSWV susceptible cultivar ‘Perfectpeel’ was used as a control. Two transplant dates were adopted in both years: 1) mean-early and 2) mean-late. A split-plot design with 3 reps was followed in the two years. Yield and fruit quality were considered as agronomic traits. About virological aspects, field observations and laboratory analysis (ELISA, IME) were carried out during crop cycles. Phytoplasma infection frequency was also assessed for the same plants. The control cultivar ‘Perfectpeel’ resulted highly productive in both years that were however characterized by low incidence of virus infections; beside, among new genotypes ‘Vespro’ and ‘Suerte’ gave a good yield, while ‘Candia’ and ‘Isi 23259’ exhibited high fruit quality. Virological studies showed that the new processing tomato genotypes were indeed TSWV-resistant. Finally, phytoplasma infections had a dissimilar incidence among plants of the tested genotypes

    Growth and yield promoting effect of artificial mycorrhization combined with different fertiliser rates on field-grown tomato

    Combination of plant inoculation with a commercial mycorrhizal formulation with half or full fertiliser application rates was evaluated for the effects on plant growth and yield and mycorrhization occurrence throughout two consecutive field tomato crops in southern Italy. Mycorrhizal formulation was inoculated on tomato seedling roots both in the nursery and after transplant. Inoculated tomato seedlings were significantly larger than non-inoculated seedlings less than 30 days after the first inoculation in the nursery. Above ground dry biomass and stem number of inoculated plants were found to be higher also at the end of each crop. Positive effects of mycorrhizal inoculation were extended also to marketable yield of both crops, mainly due to an increased number and weight of clusters and fruits. Mycorrhizal treatment also improved crop earliness, seen in terms of anticipating plant flowering, increasing first harvest yield, and reducing average harvesting time compared to non-inoculated plants. Both rates of mineral fertilisers positively affected tomato growth and marketable yield, but did not influence fruit quality parameters. No significant interaction was found between mineral fertilisation and plant mycorrhization. Crop inoculation with mycorrhizal formulations could reduce the amounts of fertilisers and pesticides being used, and could represent a sustainable technique to improve crop yield and profitability

    Improvement of Tuberculosis Laboratory Capacity on Pemba Island, Zanzibar: A Health Cooperation Project.

    Low-income countries with high Tuberculosis burden have few reference laboratories able to perform TB culture. In 2006, the Zanzibar National TB Control Programme planned to decentralize TB diagnostics. The Italian Cooperation Agency with the scientific support of the "L. Spallanzani" National Institute for Infectious Diseases sustained the project through the implementation of a TB reference laboratory in a low-income country with a high prevalence of TB. The implementation steps were: 1) TB laboratory design according to the WHO standards; 2) laboratory equipment and reagent supplies for microscopy, cultures, and identification; 3) on-the-job training of the local staff; 4) web- and telemedicine-based supervision. From April 2007 to December 2010, 921 sputum samples were received from 40 peripheral laboratories: 120 TB cases were diagnosed. Of all the smear-positive cases, 74.2% were culture-positive. During the year 2010, the smear positive to culture positive rate increased up to 100%. In March 20, 2010 the Ministry of Health and Social Welfare of Zanzibar officially recognized the Public Health Laboratory- Ivo de Carneri as the National TB Reference Laboratory for the Zanzibar Archipelago. An advanced TB laboratory can represent a low cost solution to strengthen the TB diagnosis, to provide capacity building and mid-term sustainability

    Comparison of Bioactive Substances Content between Commercial and Wild-Type Isolates of Pleurotus eryngii

    Mushrooms belonging to Pleurotus genus have been demonstrated to have important nutritional and medicinal value and their regular intake prevent many diseases, reduce the infection probability and increase immunity. In order to investigate the bioactive compounds produced by seven commercial (‘142 F’, ‘142 E’, ‘D+’, ‘V turbo’, ‘V 142’, ‘A 12’, ‘V 160’) and five wild-type (‘Albino 1107’, ‘Altamura 1603’, ‘Muro Lucano 139’, ‘Conversano 1250’, ‘Albino beige chiaro 1094’) P. eryngii isolates, the following qualitative analyses were performed: Total polyphenol content, antioxidant activity (EC50 of ABTS) and antiradical power (ARP) in fresh lyophilized and dry basidioma, and water content, -glucans and phenolic compounds in fresh samples. Standard methods were employed for each of the above mentioned aims. Total polyphenol content was diverse among the P. eryngii isolates. In particular, an elevated polyphenolic content was found in fresh lyophilized P. eryngii samples of the commercial isolates ‘V 142’ followed by ‘A 12’. The highest antiradical activity (ARP) was obtained by ‘Muro Lucano 139’ isolate. Wild P. eryngii isolates were characterized by higher water and -glucans contents compared to the commercial ones, and the highest values were registered for the ‘Albino beige chiaro 1094’ isolate. In conclusion, the present study allowed us to identify the commercial and wild-type P. eryngii isolates from the Basilicata region, with high nutritional and medicinal value based on their bioactive compounds

    Clinical Relevance of Bone Density Values from CT Related to Dental Implant Stability: A Retrospective Study

    PURPOSE: The majority of the techniques used to assess the primary implant stability are subjective and empirical and can be used during or after the surgery. The aim of this study is to evaluate the bone density prior to surgery, in order to give recommendations to the clinician about the best surgical technique and the type of implant which is needed. MATERIALS AND METHODS: A surgeon operated on 75 patients for 269 implants over the period 2010-2014. He required a CT to plan the surgery and he documented the type, the diameters, and the lengths of the implants, the insertion torque, and the ISQ values. At a later stage another clinician measured bone density and cortical thickness. We endeavoured to get the most accurate superimposition between the implants placed by the surgeon and those placed by the clinician. RESULTS: In maxilla ISQ showed a significant positive correlation with HU values detected for coronal-buccal (r = 0.302; p = 0.020) and middle-lingual (r = 0.295; p = 0.023). Torque showed a positive correlation with cortical bone thickness at the middle of the ridge (\u3c1 = 0.196; p = 0.032). CONCLUSION: It is important to take into consideration the Hounsfield Units and the cortical thickness as predictive parameters during the preoperative assessment, with regard to the choice of the implant type as well as the surgical technique
