    Defective DNA repair mechanisms in prostate cancer: impact of olaparib

    The field of prostate oncology has continued to change dramatically. It has truly become a field that is intensely linked to molecular genetic alterations, especially DNA-repair defects. Germline breast cancer 1 gene (BRCA1) and breast cancer 2 gene (BRCA2) mutations are implicated in the highest risk of prostate cancer (PC) predisposition and aggressiveness. Poly adenosine diphosphate ribose polymerase (PARP) proteins play a key role in DNA repair mechanisms and represent a valid target for new therapies. Olaparib is an oral PARP inhibitor that blocks DNA repair pathway and coupled with BRCA mutated-disease results in tumor cell death. In phase II clinical trials, including patients with advanced castration-resistant PC, olaparib seems to be efficacious and well tolerated. Waiting for randomized phase III trials, olaparib should be considered as a promising treatment option for PC

    Realization and characterization of a 2-photon 4-qubit linear cluster state

    We report on the experimental realization of a 4-qubit linear cluster state via two photons entangled both in polarization and linear momentum. This state was investigated by performing tomographic measurements and by evaluating an entanglement witness. By use of this state we carried out a novel nonlocality proof, the so-called ``stronger two observer all versus nothing'' test of quantum nonlocality.Comment: 4 pages, 4 figure

    Adjuvant radiation therapy in stage I seminoma: 20 years of oncologic results

    Aim: To report long term oncologic outcomes after adjuvant radiotherapy (RT) for stage I seminoma. Method: We reviewed the complete data set for all patients treated at our institute between 1988 and 2005 for stage I seminoma with adjuvant RT after radical orchiectomy. Results: A total of 85 patients were included. The median follow-3up was 15 years. The 20-3year overall survival (OS) and relapse free survival (RFS) were 92% and 96.3%, respectively. No severe acute and late complications were recorded. Overall 5.9% of patients had a second unrelated malignancy. Conclusion: Adjuvant RT is an efficacious and safe treatment in stage I seminom

    Il cognitive computing sta rivoluzionando le competenze e le conoscenze che devono essere acquisite dagli allievi universitari che si devono cimen-tare con la risoluzione dei problemi complessi, ciò è dovuto alla irruzione “dirompente” delle tecnologie quali: cloud, Big Data, IoT, dispositivi mobi-li e social network. Il cognitive computing, in particolare, rappresenta la tecnologia più dirompente che le integra tutte. In questo lavoro gli autori presentano lo studio di un caso, la rilevazione delle discariche abusive nelle zone urbane della città di Genova. Il lavoro è stato svolto con la collaborazione di IBM-Italia e l’assessorato all’ambiente del comune di Genova. L’algoritmo sviluppato consente la rilevazione e la segnalazione di ogni tipo di rifiuto in real time attraverso metodi di intelli-genza artificiale adoperando Watson-IBM. L’algoritmo è stato insignito come il miglior algoritmo italiano di IA mai sviluppato per questo dominio applicativo ed è stato premiato da IBM alla presenza dell’assessore all’ambiente del comune di Genova e del vicepresidente IBM Cloud Italia Alessandro la Volpe, durante la convention Party Cloud per Genova tenu-tasi a Milano nei giorni 11-12 Novembre 2018. Il premio è andato al giova-ne studente Vincenzo De Francesco il quale ha continuato su questa temati-ca nella sua tesi triennale. Questo lavoro, oltre a discutere lo studio di un caso, vuole riflettere su alcune implicazioni che questa tecnologia dirom-pente sta causando sulla didattica universitaria (e non solo) e su come alcu-ni skill, richiesti dalle aziende, ma non disponibili ancora nei nostri corsi di studio universitari, possono essere costruiti in corsi specifici come quel-lo di Cognitive Computing Systems impartito presso il Dipartimento di In-gegneria Elettrica e Tecnologie dell’Informazione dell’Università di Napoli Federico II

    A Mathematical Model for the Flow Resistance and the Related Hydrodynamic Dispersion Induced by River Dunes

    Present work is aimed at the derivation of a simply usable equation for the total flow resistance associated with river bedforms, by a unifying approach allowing for bypassing some of the limiting restrictions usually adopted in similar types of studies. Specifically, we focused on the effect induced by the out-of-phase free surface undulations appearing in presence of sand dunes. The proposed expression, obtained by combining the balance of momentum referred to the control volume whose longitudinal dimension coincides with the dune wavelength and the energy balance integrated between its extreme sections, was tested by comparison with some laboratory experimental measurements available in the literature and referred to steady flow past fixed, variably rough bedforms. In terms of shear stress or friction factor, the proposed theory provides estimates in good agreement with the real data, especially if evaluated against the performances provided by other classical similar approaches. Moreover, when analyzed in terms of hydrodynamic dispersive properties as a function of the skin roughness on the basis of a previously derived analytical solution, the dune-covered beds seem to behave like meandering channels, responsible for a globally enhanced fluid particles longitudinal spreading, with a relatively reduced effect in the presence of less pronounced riverbed modelling

    Effective electrothermal analysis of electronic devices and systems with parameterized macromodeling

    We propose a parameterized macromodeling methodology to effectively and accurately carry out dynamic electrothermal (ET) simulations of electronic components and systems, while taking into account the influence of key design parameters on the system behavior. In order to improve the accuracy and to reduce the number of computationally expensive thermal simulations needed for the macromodel generation, a decomposition of the frequency-domain data samples of the thermal impedance matrix is proposed. The approach is applied to study the impact of layout variations on the dynamic ET behavior of a state-of-the-art 8-finger AlGaN/GaN high-electron mobility transistor grown on a SiC substrate. The simulation results confirm the high accuracy and computational gain obtained using parameterized macromodels instead of a standard method based on iterative complete numerical analysis

    In search of the seismogenic fault of the March 23rd 2018 earthquake (Mw 3.7) near Brindisi (Puglia, Southern Italy)

    T The portion of the southern Adriatic Sea, adjacent to the Murge area (Apulian Foreland, Southern Italy), is considered mainly aseismic. The recent March 23rd 2018 earthquake (Mw 3.7), occurred near Brindisi (Puglia Region), giving us occasion to reconsider data coming from different sources (instrumentally-recorded, historically-documented and palaeoseismologically-interpreted earthquakes), that suggest some moderate seismic shocks in this portion of the Adriatic Sea. The present study, based on the re-interpretation of public domain seismic profiles and exploration well logs available in the ViDEPI Project, has been focused on the recognition of faults with Quaternary activity in the epicentral area. A S-dipping fault, with a main dip-slip component of movement, is suggested to be the seismogenic source. Its geometry is consistent with the fault plane solution and the depth of the hypocenter calculated by the INGV (Istituto Nazionale di Geofisica e Vulcanologia).  </div

    Role of Scaffold Protein Proline-, Glutamic Acid-, and Leucine-Rich Protein 1 (PELP1) in the Modulation of Adrenocortical Cancer Cell Growth

    PELP1 acts as an estrogen receptor (ER) coactivator that exerts an essential role in the ER's functions. ER coregulators have a critical role in the progression and response to hormonal treatment of estrogen-dependent tumors. We previously demonstrated that, in adrenocortical carcinoma (ACC), ER\u3b1 is upregulated and that estradiol activates the IGF-II/IGF1R signaling pathways defining the role of this functional cross-talk in H295R ACC cell proliferation. The aim of this study was to determine if PELP1 is expressed in ACC and may play a role in promoting the interaction between ER\u3b1 and IGF1R allowing the activation of pathways important for ACC cell growth. The expression of PELP1 was detected by Western blot analysis in ACC tissues and in H295R cells. H295R cell proliferation decrease was assessed by A3-(4,5-Dimethylthiaoly)-2,5-diphenyltetrazolium bromide (MTT) assay and [3H] thymidine incorporation. PELP1 is expressed in ACC tissues and in H295R cells. Moreover, treatment of H295R with E2 or IGF-II induced a multiprotein complex formation consisting of PELP1, IGF1R, ER\u3b1, and Src that is involved in ERK1/2 rapid activation. PELP1/ER/IGF1R/c-Src complex identification as part of E2- and IGF-II-dependent signaling in ACC suggests PELP1 is a novel and more efficient potential target to reduce ACC growth
