    A novel immunofluorescence flow cytometry technique detects the expansion of brown tides caused by Aureoumbra lagunensis to the Caribbean Sea

The definitive version was published in Applied and Environmental Microbiology 80 (2014): 4947-4957, doi:10.1128/AEM.00888-14.During the past 3 decades, brown tides caused by the pelagophytes Aureococcus anophagefferens and Aureoumbra lagunensis have caused ecological and economic damage to coastal ecosystems across the globe. While blooms of A. lagunensis had previously been confined to Texas, in 2012, an expansive brown tide occurred on Florida's East Coast, causing widespread disruption within the Indian River and Mosquito Lagoons and generating renewed interest in this organism. A major impediment to detailed investigations of A. lagunensis in an ecosystem setting has been the absence of a rapid and reliable method for cell quantification. The combination of their small size (3 to 5 ÎŒm) and nondescript extracellular features makes identification and enumeration of these cells with conventional methods a challenge. Here we report the development of an immunological-based flow cytometry method that uses a fluorescently labeled antibody developed against A. lagunensis. This method is species specific, sensitive (detection limit of 1.5 × 103 cells ml−1), precise (1% relative standard deviation of replicated samples), and accurate (108% ± 8% recovery of spiked samples) over a wide range of cell concentrations. Furthermore, this method effectively quantifies A. lagunensis in both glutaraldehyde- and formalin-preserved samples, yields a high throughput of samples (∌35 samples h−1), and is cost-effective, making it an ideal tool for managers and scientists. This method successfully documented the recurrence of a brown tide bloom in Florida in 2013. Bloom densities were highest in June (>2.0 × 106 cells ml−1) and spanned >60 km from the Ponce de Leon inlet in the northern Mosquito Lagoon south to Titusville in the Indian River Lagoon. Low levels of A. lagunensis cells were found >250 km south of this region. This method also quickly and accurately identified A. lagunensis as the causative agent of a 2013 brown tide bloom in Guantanamo Bay, Cuba, and thus should prove useful for both quantifying the dynamics of ongoing blooms of A. lagunensis as well as documenting new outbreaks of this harmful alga.This research was funded as part of an NOAA ECOHAB Event Response grant to C.J.G.     Children with grand Imaginaries: Bringing them closer to the world of science

    Although at present there are academic strategies for scientific dissemination, it is still a challenge for the general population to access quality information that is reliable, easy to understand and motivational towards science. This article proposes an image-based educational scientific dissemination tool with the goal of bringing children closer to science, through the recognition of relevant characters and their contributions. The study was developed along three stages. The first and second were conducted through a qualitative analytical approach with an interpretative perspective, using a documental method, with a review and analysis technique. During the first stage, a review and selection of studies related to scientific dissemination for children was conducted. The second one focused on the identification and selection of scientific characters, through the establishment of categories and criteria. The third stage relates to the design of outstanding characters’ biographical cards. The main contribution of this proposal is the assembly of a semantic network to portray a completed character profile, time context, place of origin, contributions, impacts, acknowledgements or prizes, as well as limitations or difficulties in context. Therefore, a mean of disclosure was devised through cards with a highly graphic and animated content regarding scientific characters sized in a gaming strategy called ‘Sapiencia’, a ludic and motivational learning tool

    DESIGN AND INTEGRATION OF A HUMAN-ROBOT PHYSICAL INTERACTION PLATFORM WITH PURPOSES OF MEDICAL DIAGNOSIS AND REHABILITATION OF UPPER LIMB

    In this paper a human-robot physical interaction system with purposes of diagnosis and rehabilitation of upper limb is proposed.Anunderactuatedhaptic device with six degrees of freedom is used, with low inertia and low joint friction.Adaptive control technique is used for passive haptic guidance and active exploration, in order to compensate the dynamic uncertainty of the human operator in the loop.To validate the experimental platform, a procedure is established with three steps: i) knowledge of the haptic interface (interaction with the kinematic virtual environment), ii) navigation in a virtual pipe with changes in the geometric characteristics (verification of position, velocity, collisions and runtime), and iii) haptic guidance in a structured path based on a clinical protocol (study of convergence and energy).Environmental conditions such as temperature, humidity, lighting and noise are characterizedwith purposes to define experimental conditions.In this work, we assess based on the NASA-TLX protocol,the workload perception of simple temporal-spatial tasks

    Safety of two-year caloric restriction in non-obese healthy individuals

    BACKGROUND: The extent to which sustained caloric restriction (CR) in healthy non-obese adults is safe has not been previously investigated. OBJECTIVE: Assess the safety and tolerability of sustained two-year CR intervention in healthy, non-obese adults. DESIGN: A multi-center, randomized controlled trial. Participants were randomized using a 2:1 allocation in favor of 25% CR vs. Ad-Libitum intake (AL). Adverse and serious adverse events (AE, SAE), safety laboratory tests, and other safety parameters were closely monitored. RESULTS: Three participants were withdrawn from the CR intervention because of the safety concerns. No deaths and one SAE was reported by participants in the CR group. Although the difference in AE between AL and CR groups was not significant, within the CR group, the incidence of nervous system (p = 0.02), musculoskeletal (p = 0.02) and reproductive system (p = 0.002) disorders was significantly higher in the normal-weight than in the overweight participants. At months 12 and 24, bone mineral densities at the lumbar spine, total hip, and femoral neck of participants in the CR group were significantly lower than in those in the AL group. CONCLUSIONS: Two-years of CR at levels achieved in CALERIE was safe and well tolerated. Close monitoring for excessive bone loss and anemia is important
