    Medlinella amphoroidea gen. et sp. nov. (Bacillariophyta) from the neck skin of Loggerhead sea turtles (Caretta caretta)

    Medlinella amphoroidea gen. et sp. nov. is described from the dorsal neck skin of loggerhead sea turtles (Caretta caretta). The presence of girdle septa, multiple copulae, and the marine epizoic habitat of Medlinella amphoroidea are characteristic features shared with many species in the similar Tripterion, Chelonicola, and Poulinea genera. The semi-lanceolate valve shape, the asymmetric valve face with distinct dorsal and ventral striae, and the volate pore occlusions distinguish Medlinella from these genera. Medlinella amphoroidea accounted for up to 50% of all diatom valves on the skin of examined loggerhead turtles. Examination of the type slides of Tripterion kalamensis and T. philoderma for comparative purposes revealed morphological features that were either insufficiently or incorrectly described in the original publications. Our observations confirm that T. philoderma lacks septa and therefore does not conform to the genus description of Tripterion. The description of cingulum structure in Tripterion kalamensis is amended to identify multiple porose copulae that are open at one end. While the description of Medlinella creates another monotypic genus within a group of similar marine epizoic genera, we feel the novel character state (volate occlusions) present in this taxon is significant. Clearly, however, further phylogenetic analysis of morphological, or the development of molecular characters in the group of similar genera is required

    Drosophila TNF modulates tissue tension in the embryo to facilitate macrophage invasive migration

    Migrating cells penetrate tissue barriers during development, inflammatory responses, and tumor metastasis. We study if migration in vivo in such three-dimensionally confined environments requires changes in the mechanical properties of the surrounding cells using embryonic Drosophila melanogaster hemocytes, also called macrophages, as a model. We find that macrophage invasion into the germband through transient separation of the apposing ectoderm and mesoderm requires cell deformations and reductions in apical tension in the ectoderm. Interestingly, the genetic pathway governing these mechanical shifts acts downstream of the only known tumor necrosis factor superfamily member in Drosophila, Eiger, and its receptor, Grindelwald. Eiger-Grindelwald signaling reduces levels of active Myosin in the germband ectodermal cortex through the localization of a Crumbs complex component, Patj (Pals-1-associated tight junction protein). We therefore elucidate a distinct molecular pathway that controls tissue tension and demonstrate the importance of such regulation for invasive migration in vivo

    A randomized comparison of HBP versus RVP: Effect on left ventricular function and biomarkers of collagen metabolism

    Background: Right ventricular pacing (RVP) can result in pacing-induced cardiomyopathy (PICM). It is unknown whether specific biomarkers reflect differences between His bundle pacing (HBP) and RVP and predict a decrease in left ventricular function during RVP. Aims: To compare the effect of HBP and RVP on the LV ejection fraction (LVEF) and to study how they affect serum markers of collagen metabolism. Methods: Ninety-two high-risk PICM patients were randomized to HBP or RVP. Their clinical characteristics, echocardiography, and serum levels of TGF-β1, MMP-9, ST2-IL, TIMP-1, and Gal-3 were studied before and six months after pacemaker implantation. Results: Fifty-three patients were randomized to HBP and 39 patients to RVP. HBP failed in 10 patients, which crossed over to the RVP group. Patients with RVP had significantly lower LVEF compared to HBP after six months of pacing (−5% and −4% in as-treated and intention-to-treat analysis, respectively). Levels of TGF-β1 after 6 months were lower in HBP than RVP (mean difference −6 ng/ml; P = 0.009) and preimplant Gal-3 and ST2-IL levels were higher in RVP patients with a decline in the LVEF ≥ 5% compared to those with a decline of < 5% (mean difference 3 ng/ml and 8 ng/ml; P = 0.02 for both). Conclusion: In high-risk PICM patients, HBP was superior to RVP in providing more physiological ventricular function, as reflected by higher LVEF and lower levels of TGF-β1. Among RVP patients, LVEF declined more in those with higher baseline Gal-3 and ST2-IL levels than those with lower levels

    Identification and biochemical analysis of a novel APOB mutation that causes autosomal dominant hypercholesterolemia

    Patients with autosomal dominant hypercholesterolemia (ADH) have a high risk of developing cardiovascular disease that can be effectively treated using statin drugs. Molecular diagnosis and family cascade screening is recommended for early identification of individuals at risk, but up to 40% of families have no mutation detected in known genes. This study combined linkage analysis and exome sequencing to identify a novel variant in exon 3 of APOB (Arg50Trp). Mass spectrometry established that low-density lipoprotein (LDL) containing Arg50Trp APOB accumulates in the circulation of affected individuals, suggesting defective hepatic uptake. Previously reported mutations in APOB causing ADH have been located in exon 26. This is the first report of a mutation outside this region causing this phenotype, therefore, more extensive screening of this large and highly polymorphic gene may be necessary in ADH families. This is now feasible due to the high capacity of recently available sequencing platforms

    Abdominal aortic aneurysm is associated with a variant in low-density lipoprotein receptor-related protein 1

    Abdominal aortic aneurysm (AAA) is a common cause of morbidity and mortality and has a significant heritability. We carried out a genome-wide association discovery study of 1866 patients with AAA and 5435 controls and replication of promising signals (lead SNP with a p value < 1 × 10-5) in 2871 additional cases and 32,687 controls and performed further follow-up in 1491 AAA and 11,060 controls. In the discovery study, nine loci demonstrated association with AAA (p < 1 × 10-5). In the replication sample, the lead SNP at one of these loci, rs1466535, located within intron 1 of low-density-lipoprotein receptor-related protein 1 (LRP1) demonstrated significant association (p = 0.0042). We confirmed the association of rs1466535 and AAA in our follow-up study (p = 0.035). In a combined analysis (6228 AAA and 49182 controls), rs1466535 had a consistent effect size and direction in all sample sets (combined p = 4.52 × 10-10, odds ratio 1.15 [1.10-1.21]). No associations were seen for either rs1466535 or the 12q13.3 locus in independent association studies of coronary artery disease, blood pressure, diabetes, or hyperlipidaemia, suggesting that this locus is specific to AAA. Gene-expression studies demonstrated a trend toward increased LRP1 expression for the rs1466535 CC genotype in arterial tissues; there was a significant (p = 0.029) 1.19-fold (1.04-1.36) increase in LRP1 expression in CC homozygotes compared to TT homozygotes in aortic adventitia. Functional studies demonstrated that rs1466535 might alter a SREBP-1 binding site and influence enhancer activity at the locus. In conclusion, this study has identified a biologically plausible genetic variant associated specifically with AAA, and we suggest that this variant has a possible functional role in LRP1 expression

    Adverse socio-professional events and couple's vulnerability: explora-tory analyses of the role of conjugal configurations

    To what extent do external factors make couples vulnerable? The concern of this thesis is the role of conjugal configurations in the impact of adverse events on conjugal problems, conjugal dissatisfaction or thoughts of separation. The topic is addressed within the lives vulnerability framework. The empirical analyses are done on 721 heterosexual lasting couples from the Swiss Social Survey “Social Stratification, Cohesion and Conflict in Contemporary Families”. The analysis shows that, when controlled for the previous level of conjugal satisfaction, the bicentric configurations reinforce the negative impact of adverse events on a couple's satisfaction, while the sparse configurations reduce it. These results provide some new insight into family sociology, which has until now considered dense and supportive bicentric networks to be mainly positive regarding the quality and stability of conjugal relationships, especially compared to the sparse or interfering types of networks

    The dissertation contributes to the knowledge about non-destructive testing of adhesion of a coating to the steel substrate. Two technologies have been used for the deposition of the coating: the technology of conventional casting for the tin-base coating "Stanit" and the technology of thermal spray coating for the tin coating "Babbitt", copper-base coating CuSn8 and for an NiAl interlayer. The above coating types have been compared using metallography, non-destructive ultrasonic and dye penetrant testing, X-ray diffraction, GD-OES depth profiling and energy-dispersive electron microanalysis. The following information has been collected that has to be sorted acording to the deposition technology used. For the evaluation of adhesion between the steel substrate and the coatings prepared by conventional casting, suitability of the ultrasound testing was proved, with the dye penetrant test as a supplementary method. Measurement parameters were defined for the ultrasonic testing. For te WAS technology (wire arc spraying), the non-destructive methods did not prove to be reasonably applicable in practice. The reason is that significant changes in the morphology of the structure occur in this technology. Layered structure with interfaces between the splatters of the coating, non-deformed particles and single layers of the coating results in a strong attenuation of the ultrasound waves. The coating exhibit a poor acoustic transmittion or no acoustic transmittion at all. results obtained by the non-destructive methods and other experimental methods used have been compared and a good agreement was found.Available from STL, Prague, CZ / NTK - National Technical LibrarySIGLECZCzech Republi
