    Assessment of crash fire hazard of LH sub 2 fueled aircraft

    The relative safety of passengers in LH2 - fueled aircraft, as well as the safety of people in areas surrounding a crash scene, has been evaluated in an analytical study. Four representative circumstances were postulated involving a transport aircraft in which varying degrees of severity of damage were sustained. Potential hazard to the passengers and to the surroundings posed by the spilled fuel was evaluated for each circumstance. Corresponding aircraft fueled with liquid methane, Jet A, and JP-4 were also studied in order to make comparisons of the relative safety. The four scenarios which were used to provide a basis for the evaluation included: (1) a small fuel leak internal to the aircraft, (2) a survivable crash in which a significant quantity of fuel is spilled in a radial pattern as a result of impact with a stationary object while taxiing at fairly low speed, (3) a survivable crash in which a significant quantity of fuel is spilled in an axial pattern as a result of impact during landing, and (4) a non-survivable crash in which a massive fuel spill occurs instantaneously

    Study of methane fuel for subsonic transport aircraft

    The cost and performance were defined for commercial transport using liquid methane including its fuel system and the ground facility complex required for the processing and storage of methane. A cost and performance comparison was made with Jet A and hydrogen powered aircraft of the same payload and range capability. Extensive design work was done on cryogenic fuel tanks, insulation systems as well as the fuel system itself. Three candidate fuel tank locations were evaluated, i.e., fuselage tanks, wing tanks or external pylon tanks

    Study of advanced fuel system concepts for commercial aircraft and engines

    The impact on a commercial transport aircraft of using fuels which have relaxed property limits relative to current commercial jet fuel was assessed. The methodology of the study is outlined, fuel properties are discussed, and the effect of the relaxation of fuel properties analyzed. Advanced fuel system component designs that permit the satisfactory use of fuel with the candidate relaxed properties in the subject aircraft are described. The two fuel properties considered in detail are freezing point and thermal stability. Three candidate fuel system concepts were selected and evaluated in terms of performance, cost, weight, safety, and maintainability. A fuel system that incorporates insulation and electrical heating elements on fuel tank lower surfaces was found to be most cost effective for the long term

    Study of fuel systems for LH2-fueled subsonic transport aircraft, volume 1

    Several engine concepts examined to determine a preferred design which most effectively exploits the characteristics of hydrogen fuel in aircraft tanks received major emphasis. Many candidate designs of tank structure and cryogenic insulation systems were evaluated. Designs of all major elements of the aircraft fuel system including pumps, lines, valves, regulators, and heat exchangers received attention. Selected designs of boost pumps to be mounted in the LH2 tanks, and of a high pressure pump to be mounted on the engine were defined. A final design of LH2-fueled transport aircraft was established which incorporates a preferred design of fuel system. That aircraft was then compared with a conventionally fueled counterpart designed to equivalent technology standards

    Molecular Cloning, Characterization and Expression Analysis of Two Members of the Pht1 Family of Phosphate Transporters in Glycine max

    BACKGROUND: Phosphorus is one of the macronutrients essential for plant growth and development. The acquisition and translocation of phosphate are pivotal processes of plant growth. In a large number of plants, phosphate uptake by roots and translocation within the plant are presumed to occur via a phosphate/proton cotransport mechanism. PRINCIPAL FINDINGS: We cloned two cDNAs from soybean (Glycine max), GmPT1 and GmPT2, which show homology to the phosphate/proton cotransporter PHO84 from the budding yeast Saccharomyces cerevisiae. The amino acid sequence of the products predicted from GmPT1 and GmPT2 share 61% and 63% identity, respectively, with the PHO84 in amino acid sequence. The deduced structure of the encoded proteins revealed 12 membrane-spanning domains with a central hydrophilic region. The molecular mass values are ∼58.7 kDa for GmPT1 and ∼58.6 kDa for GmPT2. Transiently expressed GFP-protein fusions provide direct evidence that the two Pi transporters are located in the plasma membrane. Uptake of radioactive orthophosphate by the yeast mutant MB192 showed that GmPT1 and GmPT2 are dependent on pH and uptake is reduced by the addition of uncouplers of oxidative phosphorylation. The K(m) for phosphate uptake by GmPT1 and GmPT2 is 6.65 mM and 6.63 mM, respectively. A quantitative real time RT-PCR assay indicated that these two genes are expressed in the roots and shoots of seedlings whether they are phosphate-deficient or not. Deficiency of phosphorus caused a slight change of the expression levels of GmPT1 and GmPT2. CONCLUSIONS: The results of our experiments show that the two phosphate transporters have low affinity and the corresponding genes are constitutively expressed. Thereby, the two phosphate transporters can perform translocation of phosphate within the plant

    Ion homeostasis in the Chloroplast

    peer reviewedThe chloroplast is an organelle of high demand for macro- and micro-nutrient ions, which are required for the maintenance of the photosynthetic process. To avoid deficiency while preventing excess, homeostasis mechanisms must be tightly regulated. Here, we describe the needs for nutrient ions in the chloroplast and briefly highlight their functions in the chloroplastidial metabolism. We further discuss the impact of nutrient deficiency on chloroplasts and the acclimation mechanisms that evolved to preserve the photosynthetic apparatus. We finally present what is known about import and export mechanisms for these ions. Whenever possible, a comparison between cyanobacteria, algae and plants is provided to add an evolutionary perspective to the description of ion homeostasis mechanisms in photosynthesis

    Chloroplast envelope membranes: a dynamic interface between plastids and the cytosol.

    Chloroplasts are bounded by a pair of outer membranes, the envelope, that is the only permanent membrane structure of the different types of plastids. Chloroplasts have had a long and complex evolutionary past and integration of the envelope membranes in cellular functions is the result of this evolution. Plastid envelope membranes contain a wide diversity of lipids and terpenoid compounds serving numerous biochemical functions and the flexibility of their biosynthetic pathways allow plants to adapt to fluctuating environmental conditions (for instance phosphate deprivation). A large body of knowledge has been generated by proteomic studies targeted to envelope membranes, thus revealing an unexpected complexity of this membrane system. For instance, new transport systems for metabolites and ions have been identified in envelope membranes and new routes for the import of chloroplast-specific proteins have been identified. The picture emerging from our present understanding of plastid envelope membranes is that of a key player in plastid biogenesis and the co-ordinated gene expression of plastid-specific protein (owing to chlorophyll precursors), of a major hub for integration of metabolic and ionic networks in cell metabolism, of a flexible system that can divide, produce dynamic extensions and interact with other cell constituents. Envelope membranes are indeed one of the most complex and dynamic system within a plant cell. In this review, we present an overview of envelope constituents together with recent insights into the major functions fulfilled by envelope membranes and their dynamics within plant cells

    Characterization of PitA and PitB from Escherichia coli

    Escherichia coli contains two major systems for transporting inorganic phosphate (P(i)). The low-affinity P(i) transporter (pitA) is expressed constitutively and is dependent on the proton motive force, while the high-affinity Pst system (pstSCAB) is induced at low external P(i) concentrations by the pho regulon and is an ABC transporter. We isolated a third putative P(i) transport gene, pitB, from E. coli K-12 and present evidence that pitB encodes a functional P(i) transporter that may be repressed at low P(i) levels by the pho regulon. While a pitB(+) cosmid clone allowed growth on medium containing 500 μM P(i), E. coli with wild-type genomic pitB (pitA ΔpstC345 double mutant) was unable to grow under these conditions, making it indistinguishable from a pitA pitB ΔpstC345 triple mutant. The mutation ΔpstC345 constitutively activates the pho regulon, which is normally induced by phosphate starvation. Removal of pho regulation by deleting the phoB-phoR operon allowed the pitB(+) pitA ΔpstC345 strain to utilize P(i), with P(i) uptake rates significantly higher than background levels. In addition, the apparent K(m) of PitB decreased with increased levels of protein expression, suggesting that there is also regulation of the PitB protein. Strain K-10 contains a nonfunctional pitA gene and lacks Pit activity when the Pst system is mutated. The pitA mutation was identified as a single base change, causing an aspartic acid to replace glycine 220. This mutation greatly decreased the amount of PitA protein present in cell membranes, indicating that the aspartic acid substitution disrupts protein structure
