    El objetivo de este trabajo es realizar el modelamiento y calcular la fuerza total, tangencial, radial y axial, porque, velocidad angular de desplazamiento lateral, rigidez del resorte lineal y del amortiguador viscoso en el dominio del tiempo debido a las vibraciones generadas en el sistema de transmisión de un aerogenerador de inducción doblemente alimentado con velocidad fija por la acción de las cargas mecánicas. Para realizar el modelamiento y simulación se utilizará un modelo multi masa del sistema de transmisión, donde cada componente tendrá un grado de libertad y las fuerzas de contacto se modelan con un resorte lineal y su respectiva rigidez. Además, el modelo de masa única no es lo suficiente preciso para un análisis vibracional de esta magnitud.Abstract : The objective of this work is modeling and calculating the total, tangential, radial and axial force, torque, lateral displacement angular velocity, linear spring stiffness and viscous damping in the time domain due to vibrations in the system transmission of a wind turbine doubly fed induction with fixed speed by the action of mechanical loads. To perform the modeling and simulation, a multi mass model of the transmission system will be use, where each component will have a degree of freedom and the contact forces are modeled with a linear spring and its respective rigidity. In addition, the single mass model is not sufficiently accurate for a vibrational analysis of this magnitude

    A comparison between plausible models in layered turbid media with geometrical variations applying a Bayesian selection criterion

    One possible application of Near Infrared techniques is to analyze human brain metabolic activity. Currently used models take into account the layered structure of the human head but, usually, they do not consider the non-planar surface of some of the boundaries, i.e. gray matter, which results in a much more complex structure, thus leading to more sophisticated models and longer calculation times. The main objective of this work is to determine if it is worth to replace a planar layered structure by a non-planar one. To this end we implement a Bayesian-based quantitative methodology for choosing between two competitive models describing light propagation in layered turbid media. Experiments of time-resolved diffuse reflectance measurements are performed in layered phantoms and complemented with numerical calculations. The resulting Distributions of Time of Flight of both models are compared using Bayesian model selection analysis. The non-planar interface was introduced in the simulations by a simple surface parametrization.     Theoretical investigation of photons partialpathlengths in multilayered turbid media

    Functional near infrared spectroscopy (fNIRS) is a valuable tool for assessing oxy- and deoxyhemoglobin concentration changes (∆[HbO] and ∆[HbR], respectively) in the human brain. To this end, photon pathlengths in tissue are needed to convert from light attenuation to ∆[HbO] and ∆[HbR]. Current techniques describe the human head as a homogeneous medium, in which case these pathlengths are easily computed. However, the head is more appropriately described as a layered medium; hence, the partial pathlengths in each layer are required. The current way to do this is by means of Monte Carlo (MC) simulations, which are time-consuming and computationally expensive. In this work, we introduce an approach to theoretically calculate these partial pathlengths, which are computed several times faster than MC simulations. Comparison of our approach with MC simulations show very good agreement. Results also suggest that these analytical expressions give much more specific information about light absorption in each layer than in the homogeneous case.Fil: García, Héctor Alfredo.     Membrane vesicles from the probiotic Nissle 1917 and gut resident Escherichia coli strains distinctly modulate human dendritic cells and subsequent T cell responses

    Extracellular membrane vesicles (MVs) released by gut microbiota are key players in the communication with the host. The aim of this study was to evaluate the immunomodulatory properties of MVs from the probiotic E. coli Nissle 1917 (EcN) in terms of DC-derived adaptive immune responses and to compare the effects with those elicited by commensal E. coli. The effects of MVs were analysed in monocyte-derived DCs by measuring cytokine expression and the ability of activated-DCs to differentiate CD4+ T cells towards specific effector subsets. EcN MVs derived intricate Th1/Th2/Th17/Th22/Treg responses consistent with the beneficial effects of this probiotic. Th2/Th17/Th22 responses were common to commensal E. coli-derived vesicles but specific differences were observed for Th1 and Treg responses. Since MVs activate DCs in a strain-specific manner, probiotic-derived MVs could be explored as a safe (bacteria-free) strategy to develop new functional food ingredients targeting gut microbiota balance or intestinal inflammation

    Ocular penetration of fluorometholone-loaded PEG-PLGA nanoparticles functionalized with cell-penetrating peptides

    Aim: Development of fluorometholone-loaded PEG-PLGA nanoparticles (NPs) functionalized with cellpenetrating peptides for the treatment of ocular inflammatory disorders. Materials & methods: Synthesized polymers and peptides were used for elaboration of functionalized NPs, which were characterized physicochemically. Cytotoxicity and ability to modulate the expression of proinflammatory cytokines were evaluated in vitro using human corneal epithelial cells (HCE-2). NP uptake was assayed in both in vitro and in vivo models. Results: NPs showed physicochemical characteristics suitable for ocular administration without evidence of cytotoxicity. TAT-NPs and G2-NPs were internalized and displayed anti-inflammatory activity in both HCE-2 cells and mouse eye. Conclusion: TAT-NPs and G2-NPs could be considered a novel strategy for the treatment of ocular inflammatory diseases of the anterior and posterior segment

    Extracellular vesicles and soluble factors secreted by Escherichia coli Nissle 1917 and ECOR63 protect against enteropathogenic E. coli-induced intestinal epithelial barrier dysfunction

    Background: Enteric pathogens have developed mechanisms to disrupt tight junctions and increase gut permeability. Many studies have analysed the ability of live probiotics to protect intestinal epithelial cells against tight junction damage caused by bacterial pathogens. Escherichia coli Nissle 1917 (EcN) is among the probiotics that positively modulates the intestinal epithelial barrier by regulating expression and distribution of tight junction proteins. We previously reported that regulation of ZO-1, claudin-14 and claudin-2 is mediated by EcN secreted factors, either free-released or associated with outer membrane vesicles (OMVs). Factors secreted by commensal ECOR63 elicited comparable effects in intact epithelial T-84 and Caco-2 cell monolayers. Results:Here we analyse the ability of OMVs and soluble secreted factors to protect epithelial barrier function in polarized T-84 and Caco-2 cells infected with enteropathogenic Escherichia coli (EPEC). Transepithelial electrical resistance, paracellular permeability, mRNA levels and subcellular distribution of tight junction proteins were monitored in the absence or presence of EcN and ECOR63 extracellular fractions. EPEC downregulated expression of ZO-1 ZO-2, occludin and claudin-14 and altered the subcellular localization of ZO-1, occludin and F-actin cytoskeleton. OMVs and soluble factors secreted by EcN and ECOR63 counteracted EPEC- altered transepithelial resistance and paracellular permeability, preserved occludin and claudin-14 mRNA levels, retained ZO-1 and occludin at tight junctions in the cell boundaries and ameliorated F-actin disorganization. Redistribution of ZO-1 was not accompanied by changes at mRNA level. Conclusions: This study provides new insights on the role of microbiota secreted factors on the modulation of intestinal tight junctions, expanding their barrier- protective effects against pathogen-induced disruption

    Topical Pioglitazone Nanoformulation for the Treatment of Atopic Dermatitis: Design, Characterization and E cacy in Hairless Mouse Model

    Pioglitazone (PGZ) is a drug used to treat type 2 diabetes mellitus that has been reported to show additional therapeutic activities on diverse inflammatory parameters. The aim of this study was to optimize a topical PGZ-loaded nanoemulsion (PGZ-NE) in order to evaluate its effectiveness for treating atopic dermatitis (AD). The composition of the nanoformulation was established by pseudo-ternary diagram. Parameters such as physical properties, stability, in vitro release profile, and ex vivo permeation were determined. The efficacy study was carried out using oxazolone-induced AD model in hairless mice. PGZ-NE released the drug following a hyperbolic kinetic. Additionally, its properties provided high retention potential of drug inside the skin. Therapeutic benefits of PGZ-NE were confirmed on diverse events of the inflammatory process, such as reduction of lesions, enhancement of skin barrier function, diminished infiltration of inflammatory cells, and expression of pro-inflammatory cytokines. These results were reinforced by atomic force microscope (AFM), which demonstrated the ability of the formulation to revert the rigidification caused by oxazolone and consequently improve the elasticity of the skin.     Spatial variation of picoplankton communities along a cascade reservoir system in Patagonia, Argentina

    In this study we explored how picoplankton community structure and diversity varied along three cascade oligo-mesotrophic reservoirs of the Limay River (Patagonia, Argentina): Alicura, Piedra del Águila and Ramos Mexía. We analyzed the spatial changes, covering lotic and lentic stretches along a gradient of 262 km from Andes to steppe, and we also sampled the main affluent of the Limay River (Collon Cura). In all sampling sites the main limnological variables were measured, and the picoplankton abundance (autotrophic and heterotrophic) was analyzed by flow cytometry. The bacterial biodiversity was assessed using high throughput sequencing Illumina MiSeq. We expected an increase in the trophic state along this series of cascade reservoirs, which would determine spatial differences in the structure of the picoplankton communities. We also hypothesized that the lotic and lentic conditions along the system would influence the bacterial composition. The results showed a slight increase in trophic state together with an increase in overall picoplankton abundance downstream, towards Ramos Mexía Reservoir. Picocyanobacteria were represented by phycoerythrin-rich cells all along the system, in accordance to the pattern described for oligotrophic aquatic ecosystems. Multivariate analyses based on bacterial OTU composition and environmental variables showed a spatial ordination of sites following the trend of increasing trophic state downstream. Molecular analyses of bacterial OTU diversity also showed an increase in richness and a decrease in evenness at the lotic stretches, and the opposite pattern in the reservoirs, suggesting that water retention time may play a role in structuring the community composition.Fil: Bernal, María Carolina.     Paediatricians provide higher quality care to children and adolescents in primary care: A systematic review

    Aim: The number of primary care paediatricians is decreasing in Europe without a justifiable reason. We aimed to compare the clinical practice of paediatricians and family doctors attending children and adolescents in primary care. Methods: MEDLINE, Embase, CENTRAL, TRIP and Google Scholar were searched from December 2008 to February 2018. No language or study design restrictions were applied. Three reviewers assessed eligibility of the studies. Seven pairs of reviewers performed the data extraction and assessed the methodological quality independently. Discrepancies were resolved by consensus. Results: Fifty-four, out of 1150 studies preselected, were included. We found that paediatricians show more appropriate pharmacology prescription patterns for the illness being treated; they achieve higher vaccination rates and have better knowledge of vaccines and fewer doubts about vaccine safety; their knowledge and implementation of different screening tests are better; they prescribe psychoactive drugs more cautiously and more in line with current practice guidelines; their evaluation and treatment of obesity and lipid disorders follow criteria more consistently with current clinical practice guidelines; and they perform fewer diagnostic test, show a more suitable use of the test and request fewer referrals to specialists. Conclusion: According to published data, in developed countries, paediatricians provide higher quality care to children than family doctors
