    Access to Barrier Perches Improves Behavior Repertoire in Broilers

    Restriction of behavioral opportunities and uneven use of space are considerable welfare concerns in modern broiler production, particularly when birds are kept at high densities. We hypothesized that increased environmental complexity by provision of barrier perches would help address these issues by encouraging perching and enhancing use of the pen space across a range of stocking densities. 2,088 day-old broiler chicks were randomly assigned to one of the following barrier and density treatment combinations over four replications: simple barrier, complex barrier, or control (no barrier) and low (8 birds/m2), moderate (13 birds/m2), or high (18 birds/m2) density. Data were collected on focal birds via instantaneous scan sampling from 2 to 6 weeks of age. Mean estimates per pen for percent of observations seen performing each behavior, as well as percent of observations in the pen periphery vs. center, were quantified and submitted to an analysis of variance with week as the repeated measure. Barrier perches, density and age affected the behavioral time budget of broilers. Both simple and complex barrier perches effectively stimulated high perching rates. Aggression and disturbances were lower in both barrier treatments compared to controls (P<0.05). Increasing density to 18 birds/m2 compared to the lower densities suppressed activity levels, with lower foraging (P<0.005), decreased perching (P<0.0001) and increased sitting (P = 0.001) earlier in the rearing period. Disturbances also increased at higher densities (P<0.05). Use of the central pen area was higher in simple barrier pens compared to controls (P<0.001), while increasing density above 8 birds/m2 suppressed use of the central space (P<0.05). This work confirms some negative effects of increasing density and suggests that barrier perches have the potential to improve broiler welfare by encouraging activity (notably by providing accessible opportunities to perch), decreasing aggression and disturbances, and promoting more even distribution of birds throughout the pen space

    The objective of this study was to discern whether providing enrichment in the form of barrier perches across a range of densities might improve leg and foot health and promote behavioral expression and more even use of space in broilers. To investigate this, 2,088 day-old broiler chicks were randomly assigned to one of three barrier treatments at one of three densities. Effects on behavior, space use, foot and hock health, tibia fluctuating asymmetry, fear and production were subsequently assessed. Higher densities appeared to compromise broiler welfare, seen by increased tibia length asymmetry, poorer foot and hock health, suppression of activity, increased disturbances, and decreased use of space. Conversely, barrier perches - particularly simple barriers - appeared to improve footpad quality, promote increased perching and activity, decrease aggression and disturbances, and improve use of the central pen space, all without negatively impacting production traits

It should also place a greater duty of care on owners of captive decapods, considering their needs and avoiding unnecessary suffering. The recognition of decapod sentience should also have an impact on their protection as research animals, although research with a potential to cause suffering may be needed to better understand decapods’ capacity to suffer

    Public Views of Dairy Calf Welfare and Dairy Consumption Habits of American Youth and Adults

    The primary objective of this study was to explore views of dairy calf welfare and dairy product consumption habits among youth and adults. The secondary objective was to explore views of dairy calf welfare and dairy product consumption habits among a subset of parent-child pairs. Participants 5–17 years of age (n = 463) and 18 years old or greater (n = 1,310) completed an in-person survey at the Minnesota State Fair (St. Paul, MN, USA) in summer 2018. A subset of these data was comprised of parent-child pairs (n = 188). The survey was administered via Qualtrics using iPads and included multiple-choice questions about demographics and calf welfare, an open-ended question on “what dairy calves need to have a good life,” and multiple-choice questions about participants' consumption of dairy products and nondairy alternative products. Content analysis was used for responses to the open-ended question, and concepts to describe dairy calf welfare views were identified. Fisher's exact test and Cohen's Kappa were used to investigate the relationships between parent-child pair responses about dairy calf welfare. In addition to these methods, prevalence-adjusted and bias-adjusted kappa (PABAK) were used to investigate the relationships between parent-child pair responses about consumption habits. The median age of all youth participants was 11 years and 61% were female, 82% were urban residents, and 63% did not have prior experience handling agricultural animals but 83% had visited a farm in the past. Most youth participants (94.4%) indicated that they consumed dairy products, while 47.1% consumed nondairy alternatives products. Median age range of all adult participants was 45–54 years, 65% were female, 82% urban residents, and 81% did not have prior experience handling agricultural animals but 63% had visited a farm in the past. Most adult participants (94%) indicated that they consumed dairy products and 47% indicated that they consumed nondairy alternative products. In response to “what dairy calves need to have a good life,” youth and adults most commonly focused on issues related to biological functioning (82 and 70% of youth and adults mentioning this concept, respectively), followed by natural living (44 and 50%, respectively), humane care (30 and 20%, respectively), and affective states (5% of both youth and adults). For the natural living concept of animal welfare, parent and child responses were slightly associated (Kappa = 0.19; P = 0.01; overall agreement = 61%). Almost all participants reported consuming dairy products, therefore, the agreement is high between parents and children because in most households (90%), both parents and children consume dairy products. However, child consumption was observed to be lower (75%, 9/12) when parents do not consume dairy than when parents do consume dairy (95%, 167/176), leading to a Kappa of 0.20 (P = 0.006, PABAK = 0.81) and a slight association between parents and children. The results suggest that biological functioning is highly valued by the public and views of parents and their children related to natural living in dairy calves are slightly associated

    Short-Term Impact of Point-Source Enrichment on the Behavior of Gestating Sows Housed in Groups

    Environmental enrichment is an important strategy to improve the welfare of farm animals. However, relatively little is known about enrichment for gestating sows, especially those raised on farms with slatted floors and for which provision of straw may be difficult. The objective of this study was therefore to investigate the short-term (4 d) impact of a point-source enrichment object on the behavior of gestating sows housed in group pens. Four pens of gestating sows on a university research farm were randomly allocated to receive either enrichment or no enrichment (control) in a 2 by 2 crossover design. Time budgets were established by video recording focal sows' behaviors (n = 10 focals per pen) every 15 min between 0800 and 1500 every day. Enrichment use was further characterized by continuous behavior sampling for a 1 h interval between 0830 and 0930 each day. The impact of parity, lameness and presence of stereotypical behavior such as sham chewing on enrichment use was evaluated. Over the course of the study, focal sows spent approximately 73% of observations inactive [either lying down (70%), standing (2%), or sitting (1%)]. Within the remaining observations, sows were most commonly observed sham-chewing (16%), followed by 3% exploring, 2% feeding, 2% walking and 1% interacting with the enrichment when it was available. Low-parity sows, moderately-lame sows, and sows observed sham chewing at baseline displayed more consistent enrichment use over the course of the study (p = 0.02, p < 0.01, p = 0.04, respectively). While no adverse behavioral effects (increased agonism or sham chewing) due to provision or removal of the enrichment object were observed and while 85% of sows were observed to interact with enrichment at least once, interest declined sharply after the first day. We conclude that further research is needed to identify effective and sustainable enrichment strategies for gestating sows

    The Application of 3D Landmark-Based Geometric Morphometrics towards Refinement of the Piglet Grimace Scale

    Proper methods of assessment to objectively identify pain are essential for welfare improvements of piglets undergoing surgical castration on commercial farms. The Piglet Grimace Scale (PGS) is used to identify changes in facial expressions caused by acute pain in piglets undergoing tail docking and castration. However, subjective scoring methods are a concern for the validation of the PGS. The objectives of this study were to evaluate and refine the PGS through 3D landmark geometric morphometrics. Male piglets (n = 88) were randomly allocated to one of two treatments: castration and sham-castration. Piglet facial images were taken at four time points (before treatment, immediately post-treatment, 1 h post-treatment, and 4 h post-treatment) using a photogrammetry rig. Images were scored by four raters using five facial action units (FAUs): orbital tightening, ear position, temporal tension, lip contraction, and nose bulge/cheek tension. Three-dimensional facial models were generated and landmarked denoting 3 FAUs (orbital tightening, lip contraction, and nose bulge/cheek tension). Results suggest that orbital tightening and ear position may be reliable FAUs for the PGS. However, neither the PGS nor 3D landmark-based geometric morphometrics were able (both p > 0.10) to reliably identify facial indicators of pain in piglets undergoing castration

    Pan-cancer Alterations of the MYC Oncogene and Its Proximal Network across the Cancer Genome Atlas

    Although theMYConcogene has been implicated incancer, a systematic assessment of alterations ofMYC, related transcription factors, and co-regulatoryproteins, forming the proximal MYC network (PMN),across human cancers is lacking. Using computa-tional approaches, we define genomic and proteo-mic features associated with MYC and the PMNacross the 33 cancers of The Cancer Genome Atlas.Pan-cancer, 28% of all samples had at least one ofthe MYC paralogs amplified. In contrast, the MYCantagonists MGA and MNT were the most frequentlymutated or deleted members, proposing a roleas tumor suppressors.MYCalterations were mutu-ally exclusive withPIK3CA,PTEN,APC,orBRAFalterations, suggesting that MYC is a distinct onco-genic driver. Expression analysis revealed MYC-associated pathways in tumor subtypes, such asimmune response and growth factor signaling; chro-matin, translation, and DNA replication/repair wereconserved pan-cancer. This analysis reveals insightsinto MYC biology and is a reference for biomarkersand therapeutics for cancers with alterations ofMYC or the PMN

    Overexpression of MicroRNAs from the miR-17-92 Paralog Clusters in AIDS-Related Non-Hodgkin's Lymphomas

    Individuals infected by HIV are at an increased risk for developing non-Hodgkin's lymphomas (AIDS-NHL). In the highly active antiretroviral therapy (HAART) era, there has been a significant decline in the incidence of AIDS-associated primary central nervous system lymphoma (PCNSL). However, only a modest decrease in incidence has been reported for other AIDS-NHL subtypes. Thus, AIDS-NHLs remain a significant cause of morbidity and mortality in HIV infected individuals. Recently, much attention has been directed toward the role of miRNAs in cancer, including NHL. Several miRNAs, including those encoded by the miR-17-92 polycistron, have been shown to play significant roles in B cell tumorigenesis. However, the role of miRNAs in NHL in the setting of HIV infection has not been defined.We used quantitative realtime PCR to assess the expression of miRNAs from three different paralog clusters, miR-17-92, miR-106a-363, and miR-106b-25 in 24 cases of AIDS-NHLs representing four tumor types, Burkitt's lymphoma (BL, n = 6), diffuse large B-cell lymphoma (DLBCL, n = 8), primary central nervous system lymphoma (PCNSL, n = 5), and primary effusion lymphoma (PEL, n = 5). We also used microarray analysis to identify a differentiation specific miRNA signature of naïve, germinal center, and memory B cell subsets from tonsils (n = 4). miRNAs from the miR-17-92 paralog clusters were upregulated by B cells, specifically during the GC differentiation stage. We also found overexpression of these miRNA clusters in all four AIDS-NHL subtypes. Finally, we also show that select miRNAs from these clusters (miR-17, miR-106a, and miR-106b) inhibited p21 in AIDS-BL and DLBCL cases, thus providing a mechanistic role for these miRNAs in AIDS-NHL pathogenesis.Dysregulation of miR-17-92 paralog clusters is a common feature of AIDS-associated NHLs
