    Polycystic ovary syndrome (PCOS), previously referred to as Stein-Leventhal syndrome, is a worldwide disorder affecting about one fifth of women in their reproductive years. It causes disturbances in reproductive, endocrine and metabolic functions. PCOS is the focus of a great deal of research and studies indicate that its prevalence is on the increase. The main characteristics of PCOS are ovulatory dysfunction, hyperandrogenism, insulin resistance and obesity but one need to investigate and exclude other functional disorders which may resemble PCOS.     Can Anyone Climb? The Skills of a Non-specialized Toad and its Bearing on the Evolution of New Niches

    Several studies of arboreal anuran species show morphological specializations for clinging onto narrow substrates. However, little is known about these capacities in non-specialized anurans, which is crucial to understand the initial phases of adaptation to a new niche. To assess the functional requirements related to the evolution of arboreality in anurans we analyzed climbing performance, and correlated anatomical traits, in the terrestrial toad Rhinellaarenarum, a species choose as a proxy for the ancestral condition regarding the evolution of this specialized niche. We studied the impact of a substrate of wooden rods with different diameters, arrangements, and slopes on locomotion, grasping, and climbing with a comparative framework. Animals were confronted with climbing tests, video recording their behaviors. Preserved specimens were dissected to assess limb myology, osteology, and tendons’ characteristics. Our results show that how terrestrial toad R.arenarum climbs is different from those displayed by specialized tree frogs. Animals flexed their fingers and toes, grasping the substrate displaying hookings and partial graspings. The palm was scarcely involved in the grip, as in specialized anurans. These actions were performed although flexor and extensor muscles of the digits are highly conserved and generalized. Further, we formally assess the evolutionary history of ecological and anatomical traits related to climbing among Rhinella species to improving the comprehension of the relation between morphofunctional patterns and behavioral climbing skills. Our experiments revealed that this terrestrial toad possesses unexpected climbing capacities, suggesting a way in which evolution of new niches could have developed in the evolution of anurans.Fil: Vassallo, Aldo Iván.     Unravelling the DNA sequences carried by Streptomyces coelicolor membrane vesicles

    Membrane vesicles (MVs) are spherical particles with nanoscale dimensions and characterized by the presence of diverse cargos, such as nucleic acids, proteins, lipids, and cellular metabolites. Many examples of (micro)organisms producing MVs are reported in literature. Among them, bacterial MVs are of particular interest because they are now considered as the fourth mechanism of horizontal gene transfer. Streptomyces bacteria are well-known for their ecological roles and ability to synthesize bioactive compounds, with Streptomyces coelicolor being the model organism. It was previously demonstrated that it can produce distinct populations of MVs characterized by different protein and metabolite cargos. In this work we demonstrated for the first time that MVs of S. coelicolor carry both DNA and RNA and that their DNA content represents the entire chromosome of the bacterium. These findings suggest that MV DNA could have a role in the evolution of Streptomyces genomes and that MVs could be exploited in new strain engineering strategies

    The Histidine Biosynthetic Genes in the Superphylum Bacteroidota-Rhodothermota-Balneolota-Chlorobiota: Insights into the Evolution of Gene Structure and Organization

    One of the most studied metabolic routes is the biosynthesis of histidine, especially in enterobacteria where a single compact operon composed of eight adjacent genes encodes the complete set of biosynthetic enzymes. It is still not clear how his genes were organized in the genome of the last universal common ancestor community. The aim of this work was to analyze the structure, organization, phylogenetic distribution, and degree of horizontal gene transfer (HGT) of his genes in the Bacteroidota-Rhodothermota-Balneolota-Chlorobiota superphylum, a group of phylogenetically close bacteria with different surviving strategies. The analysis of the large variety of his gene structures and organizations revealed different scenarios with genes organized in more or less compact—heterogeneous or homogeneous—operons, in suboperons, or in regulons. The organization of his genes in the extant members of the superphylum suggests that in the common ancestor of this group, genes were scattered throughout the chromosome and that different forces have driven the assembly of his genes in compact operons. Gene fusion events and/or paralog formation, HGT of single genes or entire operons between strains of the same or different taxonomic groups, and other molecular rearrangements shaped the his gene structure in this superphylum

    Sir, Human giardiasis, caused by Giardia intestinalis, a agellate protozoan parasite that colonizes the small bowel, is a worldwide infection (1). Giardia infection is usually asymptomatic but intestinal illness may occur (2–5). Several reports describe the association of allergy with increased levels of total serum IgE antibodies and of speci c IgE antibodies against food allergens in patients aVected by giardiasis, and Giardia infection may determine altered absorption of food antigens causing allergic sensitization (6). Cutaneous signs may be virtually indistinguishable from those of atopic dermatitis (7, 8). Acute reactions such as urticaria or asthma have also been described (9–11).     Geometry and evolution of a fault-controlled Quaternary basin by means of TDEM and single-station ambient vibration surveys: The example of the 2009 L'Aquila earthquake area, central Italy

    We applied a joint survey approach integrating time domain electromagnetic soundings and single-station ambient vibration surveys in the Middle Aterno Valley (MAV), an intermontane basin in central Italy and the locus of the 2009 L’Aquila earthquake. By imaging the buried interface between the infilling deposits and the top of the pre-Quaternary bedrock, we reveal the 3-D basin geometry and gain insights into the long-term basin evolution. We reconstruct a complex subsurface architecture, characterized by three main depocenters separated by thresholds. Basin infill thickness varies from ~200–300m in the north to more than 450m to the southeast. Our subsurface model indicates a strong structural control on the architecture of the basin and highlights that the MAV experienced considerable modifications in its configuration over time. The buried shape of the MAV suggests a recent and still ongoing predominant tectonic control by the NW-SE trending Paganica-San Demetrio Fault System (PSDFS), which crosscuts older ~ENE and NNE trending extensional faults. Furthermore, we postulate that the present-day arrangement of the PSDFS is the result of the linkage of two previously isolated fault segments. We provide constraints on the location of the southeastern boundary of the PSDFS, defining an overall ~19 km long fault system characterized by a considerable seismogenetic potential and a maximum expected magnitude larger than M6.5. This study emphasizes the benefit of combining two easily deployable geophysical methods for reconstructing the 3-D geometry of a tectonically controlled basin.     Results from shallow geophysical investigations in the northwestern sector of the island of Malta

    We performed geophysical investigations in the northwestern sector of the island of Malta to reconstruct velocity-depth models and provide shear-wave velocity profiles. We have chosen two sites, one located in Rabat (Malta) and another in the Golden Bay area. We used both active (seismic and electrical 2D-tomography, Multichanel Analysis of Surface Waves – MASW) and passive (2D arrays and single-station measurements using ambient noise) geophysical methods. Consistently with previous studies performed in this part of Malta, we have found that both sites are characterised by site resonance in the frequency range 1-2 Hz as an effect of the local lithostratigraphic succession that shows an impedance contrast at about 60-90 m depth. This resonance effect can have important implications on both seismic hazard as well as seismic risk evaluation of the region since the amplified frequency range coincides with the resonance frequencies typical of 5–10 storey buildings which are very diffuse in the Maltese Islands, especially after intense recent urbanization.Published41-484T.     Site effects estimation and their effects on strong ground motion at Amatrice village (Central Italy)

    We present a summary of seismological and geophysical investigations at Amatrice (Central Italy), a village seated on an alluvial terrace and severely stroke by the Mw 6.0 event of August 24th 2016. The high vulnerability alone could not explain the heavy damage (X-XI MCS), whereas the vicinity of the seismic source and the peculiar site effects should be claimed to understand the ground motion variability. After the first mainshock, we investigated the Amatrice terrace for microzonation purposes together with several Italian institutions (Priolo et al., Bull. Earthquake Eng. 2019). In particular: (i) we installed 7 seismic stations as a part of the 3A network (DOI: 10.13127/SD/ku7Xm12Yy9; Cara et al., Sci. Data 2019); we performed (ii) an extensive campaign of 60 single-station ambient noise measurements (downtown stations recorded also few earthquakes), and (iii) several 2D passive seismic arrays aimed at obtaining Vs profiles down to a depth of few tens of meters (Milana et al., Bull. Earthquake Eng. 2019). Earthquake recordings were used to empirically evaluate ground motion amplification effects through spectral ratio approaches, and noise data were collected for defining the spatial distribution of the resonance frequencies. Data analysis reveals a diffuse amplification effect that reaches its maximum values in downtown area with a resonant frequency (f0) of about 2 Hz. Seismic amplification is also characterized by spatial variation and directional amplification, mainly in downtown to the west side of the alluvial terrace, and related to both stratigraphic and topographic effects. This effect tends to decrease and almost vanishes in the central part of the terrace, and it increases again moving towards its eastern edge with a clear shift of f0 towards higher frequencies. Empirical transfer functions were then used to recover the ground motion that could have hit the historical center of Amatrice during the August 24th mainshock, through the convolution with the only record in the vicinity (IT.AMT station experienced a PGA of 0.87 g). The reconstructed peak values are much greater than expected from ground motion models, showing that detailed studies on local site response can largely modify the seismic hazard assessment.PublishedSan Francisco, California (USA)4T.     Rolly Protein (ROLP)-Epb4.1/3: A Potential Protein-Protein Interaction Relevant for the Maintenance of Cell Adhesion

    We recently described Rolly Protein (ROLP), a small protein synthesized by substrate-adherent cells in a broad range of tissues. In a first set of experiments performed taking advantage of bone forming tibial cartilage as an experimental model we showed that ROLP transcription is associated to cells in an active proliferation state, whereas its downregulation is observed when cell proliferation decreases. Taking advantage of siRNA technology we also documented the expression modulation of some apoptosis-related genes in ROLP-silenced cells. In this work we search for the possible molecular interactors of ROLP by using both the antibody array approach as well as the co-immunoprecipitation approach. Results suggest the occurrence of an interaction of ROLP with Erythrocyte membrane Protein Band 4.1/3 (Epb4.1/3), an oncosuppressor downregulated in tumor development and in metastatic tissues; in addition we report experimental results that keep in line also with a potential interaction of ROLP with other PDZ-containing proteins. We also present experimental evidences supporting a role played by ROLP in cell adhesion thus supporting the existence of a biologically relevant link between ROLP and Epb4.1/3. We here suggest that ROLP might exert its biological role cooperating with Epb4.1/3, a protein that is involved in biological pathways that are often inhibited in tumor metastasis. Given the role of Epb4.1/3 in contrasting cancerogenesis we think that its cooperation with ROLP might be relevant in cancer studies and deserves further investigation
