    A German Perspective on Legal and Political Problems of Coalition Governments

    In September and October 1998 Professor von Münch was a visiting fellow of the New Zealand Institute of Public Law.  During an extremely busy visit, Professor von Münch gave a number of seminars on aspects of German Constitutional and Electoral Law.  These seminars, given by both a leading Constitutional and Electoral Law academic and a former deputy prime minister of the State of Hamburg and former member of the Bundesrat, or German Senate, were timely given the trials and tribulations of New Zealand's first MMP Coalition Government which had then just ended in the sacking of the minor party's leader as Deputy Prime Minister.  In contrast to much of the contemporary gloom at the perceived failed hope of MMP, Professor von Münch presented a hopeful view of both the electoral system that New Zealand had imported from Germany and of the possibilities of Coalition Government.     Methodology to compare costs of sanitation options for low-income peri-urban areas in Lusaka, Zambia

    Urban slums and low-income peri-urban areas in developing countries are characterised by a lack of infrastructure. The absence of sustainable sanitation systems is one of the causes that can lead to a high level of water-borne diseases in these areas, especially during the rainy season. This paper presents a methodology for cost comparisons of sanitation system options with a focus on excreta management (a sanitation system consists of the household toilets, collection and transport of excreta, treatment and storage, and transport of sanitised excreta to reuse sites). Greywater collection and treatment are excluded from the analysis for simplicity reasons. We used three low-income peri-urban areas in Lusaka, Zambia, to demonstrate our proposed methodology. The population density in the three peri-urban areas ranges from 104 to 244 people/ha. Unlined pit latrines are the most common form of excreta management, even though drilled boreholes and shallow wells are used as sources for drinking water in the same areas. Based on four selection criteria (no use of water for transporting the waste, low costs, waste sanitisation, and no contribution to groundwater pollution from stored excreta), we have short-listed two options which meet most or all of the criteria: A conventional low-cost option (Option 5: VIP latrines with downstream processing) and an ecological sanitation option (Option 6: urine-diversion dehydrating (UDD) toilets with downstream processing). The concept designs for both options are based on the entire peri-urban population in Lusaka of approximately 1.23 m. people, and on the assumption that 12 residents who live on the same plot (or ‘compound’) would share one toilet. The paper details the assumptions used to create a set of default model input parameters which are used in the cost equations to calculate capital costs, annual operating costs and net present values (NVP). Based on this basic financial analysis, we calculated the following indicative costs: capital costs of 31 €/cap and 39 €/cap for Option 5 and Option 6, respectively. Annual operating costs per capita were estimated to be 2.3 €/a·cap and 2.1 €/a·cap) for Option 5 and Option 6, respectively. The NPV for Option 6 is about 14% higher than for Option 5 but the difference is not significant, given the accuracy of the cost estimate (about ± 25%). Overall, this paper shows that the two options are difficult to differentiate based on cost alone. The financial model allows examination of the relative contributions of the different components to the overall cost of the sanitation system. For example, the costs of urine storage and transport are significant contributors to the capital and operating costs of the Ecosan option, and ways to reduce these costs should be investigated.Keywords: NPV, millennium development goals, groundwater, ecological sanitation, Ecosan, VIP latrine, UDD toilet, financial model, reuse, excret

    Use of long-term microdialysis subcutaneous glucose monitoring in the management of neonatal diabetes - A first case report

    In neonatal diabetes mellitus (NDM), a rare genetic disorder, insulin therapy is required but the management is difficult. Frequent blood glucose determinations are necessary in most cases. Microdialysis subcutaneous glucose monitoring (MSGM) is feasible in neonates and has been proposed to reduce painful blood sampling and blood loss. We have applied long-term MSGM to a small-fordate female newborn with transient NDM. We found a good correlation of subcutaneous and blood glucose concentration over a wide range of values. MSGM enabled a reduction in blood glucose determinations during optimization of intravenous insulin treatment and initiation of continuous subcutaneous insulin infusion. We conclude that long-term MSGM is feasible and may reduce painful blood sampling and blood loss in NDM. Furthermore, long-term MSGM may hold a potential for avoiding hypoglycemic episodes and earlier discharge. Copyright (C) 2006 S. Karger AG, Basel

    Hydrogen Dynamics in Superprotonic CsHSO4

    We present a detailed study of proton dynamics in the hydrogen-bonded superprotonic conductor CsHSO4 from first-principles molecular dynamics simulations, isolating the subtle interplay between the dynamics of the O--H chemical bonds, the O...H hydrogen bonds, and the SO4 tetrahedra in promoting proton diffusion. We find that the Grotthus mechanism of proton transport is primarily responsible for the dynamics of the chemical bonds, whereas the reorganization of the hydrogen-bond network is dominated by rapid angular hops in concert with small reorientations of the SO4 tetrahedra. Frequent proton jumping across the O--H...O complex is countered by a high rate of jump reversal, which we show is connected to the dynamics of the SO4 tetrahedra, resulting in a diminished CsHSO4/CsDSO4 isotope effect. We also find evidence of multiple timescales for SO4 reorientation events, leading to distinct diffusion mechanisms along the different crystal lattice directions. Finally, we employ graph-theoretic techniques to characterize the topology of the hydrogen-bond network and demonstrate a clear relationship between certain connectivity configurations and the likelihood for diffusive jump events.Comment: 12 pages, 10 figure

    NIPA Defines an SCF-Type Mammalian E3 Ligase that Regulates Mitotic Entry

    SummaryThe regulated oscillation of protein expression is an essential mechanism of cell cycle control. The SCF class of E3 ubiquitin ligases is involved in this process by targeting cell cycle regulatory proteins for degradation by the proteasome, with the F-box subunit of the SCF specifically recruiting a given substrate to the SCF core. Here we identify NIPA (nuclear interaction partner of ALK) as a human F-box-containing protein that defines an SCF-type E3 ligase (SCFNIPA) controlling mitotic entry. Assembly of this SCF complex is regulated by cell-cycle-dependent phosphorylation of NIPA, which restricts substrate ubiquitination activity to interphase. We show nuclear cyclin B1 to be a substrate of SCFNIPA. Inactivation of NIPA by RNAi results in nuclear accumulation of cyclin B1 in interphase, activation of cyclin B1-Cdk1 kinase activity, and premature mitotic entry. Thus, SCFNIPA-based ubiquitination may regulate S-phase completion and mitotic entry in the mammalian cell cycle

    ProdoNet: identification and visualization of prokaryotic gene regulatory and metabolic networks

    ProdoNet is a web-based application for the mapping of prokaryotic genes and the corresponding proteins to common gene regulatory and metabolic networks. For a given list of genes, the system detects shared operons, identifies co-expressed genes and deduces joint regulators. In addition, the contribution to shared metabolic pathways becomes visible on KEGG maps. Furthermore, the co-occurrence of genes of interest in gene expression profiles can be added to the visualization of the global network. In this way, ProdoNet provides the basis for functional genomics approaches and for the interpretation of transcriptomics and proteomics data. As an example, we present an investigation of an experimental membrane subproteome analysis of Pseudomonas aeruginosa with ProdoNet. The ProdoNet dataset on transcriptional regulation is based on the PRODORIC Prokaryotic Database of Gene Regulation and the Virtual Footprint tool.
