Mitochondria in tumor cells studied by laser scanning confocal microscopy

We present here a confocal fluorescence microscopy study of mitochondria in sensitive and resistant carcinoma cells by using two potentiometric probes of mitochondria, rhodamine 123 (R123) and dimethylaminostyryl-methylpyridiniumiodine. We have found that active mitochondria in sensitive MCF-7 and multidrug resistant MCF-7/DX carcinoma cells are very different in localization and morphology. In sensitive cells active mitochondria are found in the perinuclear region, whereas in the multidrug resistance (MDR) subline they are confined to the cell periphery. Interestingly, the MDR revertant verapamil has been found to restore in MCF-7/DX cells the same pattern of active mitochondria seen in sensitive cells. We have also studied R123 in human lung carcinoma A549 cells, which display a low responsivity to doxorubicin, and overexpress the lung resistance-related protein. In addition to perinuclear mitochondria, peripheral mitochondria with weaker fluorescence can be ssen in this cell line. Interestingly, in the two examined carcinoma lines we have been able to recognize by image analysis a common new star-lobed morphology. Oure results indicate that in resistant carcinoma cells two populations of mitochondria coexist with different localization, morphology, and activity. © 2004 Society of Photo-Optical Instrumentation Engineer

Social investment, labour market participation and public debt sustainability: An empirical analysis of European countries

This article explores the role of SI Stock, Flow and Buffer policies by shedding light on their relationships with active labour market participation and public debt sustainability for a panel of 22 European countries from 1997 to 2018. We find SI Stock, Flow and Buffer to be positively correlated with labour market participation and more sustainable public debt. When disaggregating the components of SI, we detect a small degree of heterogeneity, with Active Labour Market Policies (ALMPs) negatively associated with the activity rate and positively associated with the employment rate. This result is coherent with the idea that ALMPs make a significant contribution to increasing opportunities for those already in the labour market rather than creating new jobs for those excluded from the labour market, that is, inactive individuals. In this case, our findings indicate that measures to fight social exclusion and out-of-work expenditure (Buffer), as well as in-kind family benefits, are significantly associated with employability for those excluded from the labour market

Yritysvastuun yhteys yrityksen markkina-arvoon

Sijoittajan saatavilla on taloudellisen informaation lisäksi ei-taloudellista informaatiota. Yritysvastuu, joka koostuu alatasoilla ympäristö- ja sosiaalisesta suorituskyvystä sekä hyvästä hallintotavasta, on ei-taloudellista informaatiota. Tutkimuksen tavoitteena oli tarkastella yritysvastuun näkymistä yritysten markkina-arvossa. Yritysvastuun mittarina olivat yritysvastuuarvioinnit, joita myös sijoittajat hyödyntävät päätöksenteossa. Tutkimuksen otoksena oli OMX GES Sustainability -vastuullisuusindeksissä olevat suomalaiset pörssiyritykset ja ajankohta oli puoli vuotta ennen ja vuosi jälkeen indeksin julkaisun. Vastuullisuutta oli yrityskohtaisesti arvioitu GES Risk Rating -analyysissä. Arvioinnit yhdistettiin omavaraisuusasteeseen ja nettotulokseen, eli taloudelliseen informaatioon, Ohlsonin arvonmääritysmallissa. Markkina-arvon määrityksessä voi näin yhdistää kahta informaatiotyyppiä mallissa. Yritysten tarkastelussa huomioitiin toimialan vaikutus. Tulokseksi saatiin yritysvastuun negatiivinen suhde markkina-arvoon. Alatasoilla tulos oli toisenlainen. Vaikka ympäristösuorituskyky oli negatiivisesti yhteydessä markkina-arvoon, sosiaalisen suorituskyvyn vaikutus oli positiivinen. Lisää vahvistusta tuloksille haettiin tarkastelun ajan rajaamisella indeksin julkaisun jälkeiselle ajalle. Tulosten rajoitteena oli pieni otoskoko sekä huonosti yritysvastuuarvioinneissa sijoittuneiden yritysten puuttuminen. Tulokset kertovat yritysvastuun olevan arvorelevanttia ei-taloudellista informaatiota sijoittajille. Tämän ohella tulokset tukevat yritysvastuun tarkastelemista alatasoilla. Jos tarkastelun kohteena olisi ollut yritysvastuu kokonaisuutena, sosiaalisen suorituskyvyn positiivinen yhteys markkina-arvoon olisi jäänyt havaitsematta. Sosiaalinen suorituskyky voidaan edelleen jakaa työntekijöiden, asiakkaiden sekä yhteiskunnallisten asioiden käsittelyyn. Novel nontoxic mitochondrial probe for confocal fluorescence microscopy.

We propose a 2,5-Bis[1-(4-N-methylpyridinium)ethen-2-yl)]-N-methylpyrrole ditriflate (PEPEP) as a novel nontoxic, nonpotentiometric mitochondrial probe for confocal fluorescence microscopy. PEPEP is a representative chromophore of a large family of heterocyclic fluorescent dyes that show fluorescence emission in aqueous media and great DNA affinity. We check its cytotoxicity and intracellular localization in mammalian and yeast cell cultures. We demonstrate that PEPEP is a very efficient dye for fluorescence confocal microscopy and a valuable alternative to the most frequently used mitochondrial stains. © 2006 Society of Photo-Optical Instrumentation Engineers

Identification of N-terminal protein acetylation and arginine methylation of the voltage-gated sodium channel in end-stage heart failure human heart

The α subunit of the cardiac voltage-gated sodium channel, Naᵥ1.5, provides the rapid sodium inward current that initiates cardiomyocyte action potentials. Here, we analyzed for the first time the post-translational modifications of Naᵥ1.5 purified from end-stage heart failure human cardiac tissue. We identified R526 methylation as the major post-translational modification of any Naᵥ1.5 arginine or lysine residue. Unexpectedly, we found that the N terminus of Naᵥ1.5 was: 1) devoid of the initiation methionine, and 2) acetylated at the resulting initial alanine residue. This is the first evidence for N-terminal acetylation in any member of the voltage-gated ion channel superfamily. Our results open the door to explore Naᵥ1.5 N-terminal acetylation and arginine methylation levels as drivers or markers of end-stage heart failure

A multidimensional analysis of youth well-being

The phase of the life cycle of youth is characterised by a series of opportunities and challenges: the choice of whether or not to continue studying, the employment opportunities, the influence of the area and the social and relational context in which they live, the community and the role of institutions. The potential risks lurking in these transitions can have life-long effects on their well-being conditions. Following the methodological approach proposed by the OECD to calculate the Youth Multi-dimensional Deprivation Indicator (Y-MDI) (Oecd, 2017), in this study the well-being of young people is measured through the construction of a multi-deprivation indicator, similar in the calculation method, but different in the selection of relevant dimensions and indicators. The concept of deprivation is here understood, in line with the capability approach defined by Sen (Sen, 1998), such as the failure to achieve a plurality of factors (individual and context) acting in determining the well-being of young people. The approach proposed provides a synthetic (and easily representative) tool for the evaluation and monitoring of youth well-being, based on the conceptual framework of Equitable and Sustainable Well-being (Bes) developed in Italy by Istat (Istat, 2019). Overall, slightly less than half of the young people aged 18-34 years (47.8%) show no deprivation, while one out of three (33.5%) is deprived in one dimension of well-being and 18.7% (slightly less than 2 million young people) is multi-deprived, i.e. deprived in two or more dimensions
