Bile Acid Recognition by NAPE-PLD
The membrane-associated enzyme NAPE-PLD (N-acyl phosphatidylethanolamine specific-phospholipase D) generates the endogenous cannabinoid arachidonylethanolamide and other lipid signaling amides, including oleoylethanolamide and palmitoylethanolamide. These bioactive molecules play important roles in several physiological pathways including stress and pain response, appetite and lifespan. Recently, we reported the crystal structure of human NAPE-PLD and discovered specific binding sites for the bile acid deoxycholic acid. In this study we demonstrate that in the presence of this secondary bile acid, the stiffness of the protein measured by elastic neutron scattering increases, and NAPE-PLD results ~7 times faster to catalyze the hydrolysis of the more unsaturated substrate N-arachidonyl-phosphatidylethanolamine, compared with N-palmitoyl-phosphatidylethanolamine. Chenodeoxycholic acid and glyco- or tauro-dihydroxy conjugates can also bind to NAPE-PLD and drive its activation. The only natural monohydroxy bile acid, lithocholic acid, shows an affinity of ~20 ÎŒM and acts instead as a reversible inhibitor (IC(50) â 68 ÎŒM). Overall, these findings provide important insights into the allosteric regulation of the enzyme mediated by bile acid cofactors, and reveal that NAPE-PLD responds primarily to the number and position of their hydroxyl groups
Evidence of coexistence of change of caged dynamics at Tg and the dynamic transition at Td in solvated proteins
Mossbauer spectroscopy and neutron scattering measurements on proteins
embedded in solvents including water and aqueous mixtures have emphasized the
observation of the distinctive temperature dependence of the atomic mean square
displacements, , commonly referred to as the dynamic transition at some
temperature Td. At low temperatures, increases slowly, but it assume
stronger temperature dependence after crossing Td, which depends on the
time/frequency resolution of the spectrometer. Various authors have made
connection of the dynamics of solvated proteins including the dynamic
transition to that of glass-forming substances. Notwithstanding, no connection
is made to the similar change of temperature dependence of obtained by
quasielastic neutron scattering when crossing the glass transition temperature
Tg, generally observed in inorganic, organic and polymeric glass-formers.
Evidences are presented to show that such change of the temperature dependence
of from neutron scattering at Tg is present in hydrated or solvated
proteins, as well as in the solvents used unsurprisingly since the latter is
just another organic glass-formers. The obtained by neutron scattering at
not so low temperatures has contributions from the dissipation of molecules
while caged by the anharmonic intermolecular potential at times before
dissolution of cages by the onset of the Johari-Goldstein beta-relaxation. The
universal change of at Tg of glass-formers had been rationalized by
sensitivity to change in volume and entropy of the beta-relaxation, which is
passed onto the dissipation of the caged molecules and its contribution to
. The same rationalization applies to hydrated and solvated proteins for

Thermal motions in bacteriorhodopsin at different hydration levels studied by neutron scattering: correlation with kinetics and light-induced conformational changes.
Bacteriorhodopsin (BR) is a transmembrane protein in the purple membrane (PM) of Halobacterium salinarum. Its function as a light-driven proton pump is associated with a cycle of photointermediates which is strongly hydration-dependent. Using energy-resolved neutron scattering, we analyzed the thermal motions (in the nanosecond-to-picosecond time range) in PM at different hydration levels. Two main populations of motions were found that responded differently to water binding. Striking correlations appeared between these "fast" motions and the "slower" kinetic constants (in the millisecond time range) of relaxations and conformational changes occurring during the photocycle
Enzyme activity and dynamics: xylanase activity in the absence of fast anharmonic dynamics.
The activity and dynamics of a simple, single subunit enzyme, the xylanase from Thermotoga maritima strain Fj SS3B.1 have been measured under similar conditions, from -70 to +10 degrees C. The internal motions of the enzyme, as evidenced by neutron scattering, undergo a sharp transition within this temperature range; they show no evidence for picosecond-timescale anharmonic behaviour (e.g. local diffusive motions or jumps between alternative conformations) at temperatures below -50 degrees C, whereas these motions are strongly activated at higher temperatures. The activity follows Arrhenius behaviour over the whole of the temperature range investigated, -70 to +10 degrees C. The results indicate that a temperature range exists over which the enzyme rate-limiting step is independent of fast anharmonic dynamics
Segregation in aqueous methanol enhanced by cooling and compression
Molecular segregation in methanol-water mixtures is studied across a wide
concentration range as a function of temperature and pressure. Cluster
distributions obtained from both neutron diffraction and molecular dynamics
simulations point to significantly enhanced segregation as the mixtures are
cooled or compressed. This evolution toward greater molecular heterogenity in
the mixture accounts for the observed changes in the water-water radial
distribution function and there are indications also of a change in the
topology of the water clusters. The observed behavior is consistent with an
approach to an upper critical solution point. Such a point would appear to be
``hidden'' below the freezing line, thereby precluding observation of the

Methanol-water solutions: A bi-percolating liquid mixture
An extensive series of neutron diffraction experiments and molecular dynamics
simulations has shown that mixtures of methanol and water exhibit extended
structures in solution despite the components being fully miscible in all
proportions. Of particular interest is a concentration region(methanol mole
fraction between 0.27 and 0.54) where {\em both} methanol and water appear to
form separate, percolating networks. This is the concentration range where many
transport properties and thermodynamic excess functions reach extremal values.
The observed concentration dependence of several of these material properties
of the solution may therefore have a structural origin.Comment: 12 pages, 6 figure
Enzyme Activity and Flexibility at Very Low Hydration
Recent measurements have demonstrated enzyme activity at hydrations as low as 3%. This raises the question of whether hydration-induced enzyme flexibility is important for activity. Here, to address this, picosecond dynamic neutron scattering experiments are performed on pig liver esterase powders at 0%, 3%, 12%, and 50% hydration by weight and at temperatures ranging from 120 to 300 K. At all temperatures and hydrations, significant quasielastic scattering intensity is found in the protein, indicating the presence of anharmonic, diffusive motion. As the hydration increases, a temperature-dependent dynamical transition appears and strengthens involving additional diffusive motion. The implication of these results is that, although the additional hydration-induced diffusive motion in the protein detected here may be related to increased activity, it is not required for the enzyme to function
