    Tuberculosis in Man, Dog and Cat: An Investigation into the Inter-Relation of Tuberculosis between Humans and Their Domestic Pets

    1. An account is given of an investigation, extending over eight years, into the inter-relation of tuberculosis between man, and his domestic pets, the dog and cat; with particular reference to the ecological aspects of the infection, whether or not it was a zoonosis, and if so, its epidemiological significance. 2. The method of investigation was by a combined medical and veterinary approach, aimed at ascertaining the incidence of tuberculosis among apparently healthy animals in contact with active tuberculosis in humans, and among humans -in contact with animals dying of tuberculosis. 3. The investigation included an attempt to refine existing methods of diagnosis of tuberculosis in dogs and cats, by the development of B.C.G. as a test of immunity, in place of P.P.D. tuberculin, which was found to be both ineffective and unreliable. 4. A questionnaire was devised to elicit the required ecological details; and then distributed by the Feline Advisory Bureau to 750 individuals in the British Isles, Western Europe and the United States of America. 5. M. tuberculosis of human origin, was recovered from 4 dogs and 3 cats in a group of 48 animals, exposed to active tuberculosis in their owners, and from one cat in a further, similar group of 22 animals. The presence of these bacilli, which were of normal virulence, was not accompanied by pathological changes typical of tuberculosis; and it was concluded that the bacilli were probably present as commensals in highly resistant animals. 6. The atypical pathological changes, described as sinus catarrh, in isolated glands of the animals producing growths of M. tuberculosis, suggested further studies to determine if these changes might be related to sarcoidosis or an early, latent, phase of a tuberculous infection in a highly resistant animal. 7. The dangers of the apparently healthy domestic pet, excreting viable and virulent tubercle bacilli, indicated the advisability of proscribing the animal for a period of several months, following the detection of an active case among the human members of the household. 8. Three quarters of the human contacts of 31 dogs dying of tuberculosis were traced, half of them to the owner, or a member of his family, but in seven cases to a neighbour or a previous owner. Of the total of 354 human contacts, 41 (11.86%) were found to have had active tuberculosis at a material time in their association with the sick animal. 9. While the definition of the exact hazard of the tuberculous animal to its owner needed a more comprehensive, and inflexibly applied study of the human contacts, including Mantoux testing, and a new method of identifying identical growths obtained from owner and pet, the present investigation made it clear that the diagnosis of tuberculosis in a domestic animal, should be subject to the same statutory notification to the Public Health Authority, as was required in the case of a tuberculous human. 10. The intradermal injection of 0.1 ml. of B.C.G., producing a papule which ulcerated before 14 days (average interval from injection to ulceration, 9 days for cats and 11 days for dogs), was interpreted as the early, immune, 'positive' reaction of the infected animal; whereas a papule ulcerating after 14 days (average interval from injection to ulceration, 22 days for cats and 24 days for dogs), was taken as the late, non-immune, 'negative' reaction of the un-infected animal. The earlier the appearance of the ulcer, i.e., an accelerated, early, immune, 'positive' reaction, the more likely the animal to be suffering from an active tuberculosis infection.     Resolved Spectroscopy of the Narrow-Line Region in NGC 1068. II. Physical Conditions Near the NGC 1068 ``Hot-Spot''

    The physical conditions near the optical continuum peak (``hot spot'') in the inner narrow line region (NLR) of the Seyfert 2 galaxy, NGC 1068. Spectra were taken with HST/STIS through the 0.1X52 arcsec slit, covering the full STIS 1200 to 10000 Angstrom waveband, and are from a region that includes the hot spot, extending 0.2, or ~ 14 pc (for H= 75 km/sec/Mpc). Perhaps the most striking feature of these spectra is the presence of strong coronal emission lines, including [S XII] 7611 which has hitherto only been identified in spectra of the solar corona. There is an apparent correlation between ionization energy and velocity of the emission lines with respect to the systemic velocity of the host galaxy, with the coronal lines blueshifted, most other high excitation lines near systemic, and some of the low ionization lines redshifted. From the results of our modeling, we find that the emission-line gas consists of three principal components: 1) one in which most of the strong emission-lines, such as [O III] 5007, [Ne V] 3426, C IV 1550, arise, 2) a more tenuous, highly ionized component, which is the source of the coronal-line emission, and 3) a component, which is not co-planar with the other two, in which the low ionization and neutral lines, such as [N II] 6548 and [O I] 6300, are formed. The first two components are directly ionized by the EUV-Xray continuum emitted by the central source, while the low ionization gas is ionized by a combination of highly absorbed continuum radiation and a small fraction of unabsorbed continuum scattered by free electrons associated with the hot spot. The combination of covering factor and Thomson optical depth of the high ionization components is insufficient to scatter the observed fraction of continuum radiation into our line-of-sight.Comment: 42 pages, Latex, includes 5 figures (postscript), to appear in the Astrophysical Journa

    Water in the electrical double layer of ionic liquids on graphene

    The performance of electrochemical devices using ionic liquids (ILs) as electrolytes can be impaired by water uptake. This work investigates the influence of water on the behavior of hydrophilic and hydrophobic ILs─with ethylsulfate and tris(perfluoroalkyl)trifluorophosphate or bis(trifluoromethyl sulfonyl)imide (TFSI) anions, respectively─on electrified graphene, a promising electrode material. The results show that water uptake slightly reduces the IL electrochemical stability and significantly influences graphene's potential of zero charge, which is justified by the extent of anion depletion from the surface. Experiments confirm the dominant contribution of graphene's quantum capacitance (CQ) to the total interfacial capacitance (Cint) near the PZC, as expected from theory. Combining theory and experiments reveals that the hydrophilic IL efficiently screens surface charge and exhibits the largest double layer capacitance (CIL ∼ 80 μF cm-2), so that CQ governs the charge stored. The hydrophobic ILs are less efficient in charge screening and thus exhibit a smaller capacitance (CIL ∼ 6-9 μF cm-2), which governs Cint already at small potentials. An increase in the total interfacial capacitance is observed at positive voltages for humid TFSI-ILs relative to dry ones, consistent with the presence of a satellite peak. Short-range surface forces reveal the change of the interfacial layering with potential and water uptake owing to reorientation of counterions, counterion binding, co-ion repulsion, and water enrichment. These results are consistent with the charge being mainly stored in a ∼2 nm-thick double layer, which implies that ILs behave as highly concentrated electrolytes. This knowledge will advance the design of IL-graphene-based electrochemical devices

    Development of a self-report measure of capability wellbeing for adults: the ICECAP-A

    Purpose The benefits of health and social care are not confined to patient health alone and therefore broader measures of wellbeing may be useful for economic evaluation.\ud This paper reports the development of a simple measure of capability wellbeing for adults (ICECAP-A).\ud Methods In-depth, informant-led, interviews to identify the attributes of capability wellbeing were conducted with 36 adults in the UK. Eighteen semi-structured, repeat interviews were carried out to develop a capability-based descriptive system for the measure. Informants were purposively selected to ensure variation in socio-economic status, age, sex, ethnicity and health. Data analysis was carried out inductively and iteratively alongside interviews, and findings were used to shape the questions in later interviews.\ud Results Five over-arching attributes of capability wellbeing were identified for the measure: ‘‘stability’’,‘‘attachment’’, ‘‘achievement’’, ‘‘autonomy’’ and ‘‘enjoyment’’. One item, with four response categories, was developed for each attribute for the ICECAP-A descriptive system.\ud Conclusions The ICECAP-A capability measure represents a departure from traditional health economics outcome measures, by treating health status as an influence over broader attributes of capability wellbeing. Further work is required to value and validate the attributes and test the sensitivity of the ICECAP-A to healthcare interventions

    Epigenetic and transcriptome profiling identifies a population of visceral adipose-derived progenitor cells with potential to differentiate into an endocrine pancreatic lineage

    Type 1 diabetes (T1D) is characterized by the loss of insulin-producing β-cells in the pancreas. T1D can be treated using cadaveric islet transplantation, but this therapy is severely limited by a lack of pancreas donors. To develop an alternative cell source for transplantation therapy, we carried out the epigenetic characterization in nine different adult mouse tissues and identified visceral adipose-derived progenitors as a candidate cell population. Chromatin conformation, assessed using chromatin immunoprecipitation (ChIP) sequencing and validated by ChIP-polymerase chain reaction (PCR) at key endocrine pancreatic gene promoters, revealed similarities between visceral fat and endocrine pancreas. Multiple techniques involving quantitative PCR, in-situ PCR, confocal microscopy, and flow cytometry confirmed the presence of measurable (2–1000-fold over detectable limits) pancreatic gene transcripts and mesenchymal progenitor cell markers (CD73, CD90 and CD105; >98%) in visceral adipose tissue-derived mesenchymal cells (AMCs). The differentiation potential of AMCs was explored in transgenic reporter mice expressing green fluorescent protein (GFP) under the regulation of the Pdx1 (pancreatic and duodenal homeobox-1) gene promoter. GFP expression was measured as an index of Pdx1 promoter activity to optimize culture conditions for endocrine pancreatic differentiation. Differentiated AMCs demonstrated their capacity to induce pancreatic endocrine genes as evidenced by increased GFP expression and validated using TaqMan real-time PCR (at least 2–200-fold relative to undifferentiated AMCs). Human AMCs differentiated using optimized protocols continued to produce insulin following transplantation in NOD/SCID mice. Our studies provide a systematic analysis of potential islet progenitor populations using genome-wide profiling studies and characterize visceral adipose-derived cells for replacement therapy in diabetes

    The natural history of chronic obstructive pulmonary disease

    Chronic obstructive pulmonary disease (COPD) is a leading cause of morbidity and mortality in the USA, and it remains one of the few diseases that continues to increase its numbers. The development and progression of COPD can vary dramatically between individuals. A low level of lung function remains the cornerstone of COPD diagnosis and is a key predictor of prognosis. Lung function, however, is not the only factor in determining morbidity and mortality related to COPD, with factors such as body mass index, exercise capability and comorbid disease being important predictors of poor outcomes. Exacerbations of COPD are additional important indicators of both quality of life and outcomes in COPD patients. Definitions of exacerbations can vary, ranging from an increase in symptoms to COPD-related hospitalisations and death. COPD exacerbations are more common in patients with lower levels of lung function and may lead to more rapid declines in lung function. Better understanding of the natural history of COPD may lead to better definitions of specific COPD phenotypes, better interventions and improved outcomes

    Atmospheric neutrino induced muons in the MACRO detector

    A measurement of the flux of neutrino-induced muons using the MACRO detector is presented. Different event topologies, corresponding to different neutrino parent energies can be detected. The upward throughgoing muon sample is the larger event sample. The observed upward-throughgoing muons are 26% fewer than expected and the zenith angle distribution does not fit with the expected one. Assuming neutrino oscillations, both measurements suggest maximum mixing and Dm2 of a few times 10-3 eV2. The other samples are due to the internally produced events and to upward-going stopping muons.     The Observation of Up-going Charged Particles Produced by High Energy Muons in Underground Detectors

    An experimental study of the production of up-going charged particles in inelastic interactions of down-going underground muons is reported, using data obtained from the MACRO detector at the Gran Sasso Laboratory. In a sample of 12.2 10^6 single muons, corresponding to a detector livetime of 1.55 y, 243 events are observed having an up-going particle associated with a down-going muon. These events are analysed to determine the range and emission angle distributions of the up-going particle, corrected for detection and reconstruction efficiency. Measurements of the muon neutrino flux by underground detectors are often based on the observation of through-going and stopping muons produced in νμ\nu_\mu interactions in the rock below the detector. Up-going particles produced by an undetected down-going muon are a potential background source in these measurements.
