    Las tareas cotidianas realizadas por los agentes de seguridad aeroportuaria incluyen procedimientos informales o semi-formales basados en la experiencia adquirida, los cuales complementan a los mecanismos formales utilizados en los puntos de inspección y registro previos al embarque en el Aeropuerto Internacional de Ezeiza. Sin embargo, tales procedimientos que comprenden ciertas técnicas de detección de comportamientos sospechosos, no han sido examinados sistemáticamente, ni documentados, e incluso escasas veces han sido sometidos a discusión en nuestro país. Con el objeto de comprender en profundidad la manera en que se aplican las medidas de seguridad a nivel local, de analizar las ventajas y desventajas de los procedimientos que actualmente se llevan a cabo en los aeropuertos, y de generar propuestas para optimizar la labor de los responsables de la seguridad aeroportuaria argentina, el trabajo expuesto en este congreso reúne las discusiones actuales en torno a la problemática de las metodologías empleadas para identificar sospechosos y consecuentemente someterlos a controles adicionales, y esboza propuestas de análisis de la misma en el contexto argentino.Facultad de Ingenierí

    SerpinB2 regulates stromal remodelling and local invasion in pancreatic cancer

    Pancreatic cancer has a devastating prognosis, with an overall 5-year survival rate of ~8%, restricted treatment options and characteristic molecular heterogeneity. SerpinB2 expression, particularly in the stromal compartment, is associated with reduced metastasis and prolonged survival in pancreatic ductal adenocarcinoma (PDAC) and our genomic analysis revealed that SERPINB2 is frequently deleted in PDAC. We show that SerpinB2 is required by stromal cells for normal collagen remodelling in vitro, regulating fibroblast interaction and engagement with collagen in the contracting matrix. In a pancreatic cancer allograft model, co-injection of PDAC cancer cells and SerpinB2(-/-) mouse embryonic fibroblasts (MEFs) resulted in increased tumour growth, aberrant remodelling of the extracellular matrix (ECM) and increased local invasion from the primary tumour. These tumours also displayed elevated proteolytic activity of the primary biochemical target of SerpinB2-urokinase plasminogen activator (uPA). In a large cohort of patients with resected PDAC, we show that increasing uPA mRNA expression was significantly associated with poorer survival following pancreatectomy. This study establishes a novel role for SerpinB2 in the stromal compartment in PDAC invasion through regulation of stromal remodelling and highlights the SerpinB2/uPA axis for further investigation as a potential therapeutic target in pancreatic cancer

    Semaphorin 3A Suppresses Tumor Growth and Metastasis in Mice Melanoma Model

    <div><h3>Background</h3><p>Recent understanding on cancer therapy indicated that targeting metastatic signature or angiogenic switch could be a promising and rational approach to combat cancer. Advancement in cancer research has demonstrated the potential role of various tumor suppressor proteins in inhibition of cancer progression. Current studies have shown that axonal sprouting inhibitor, semaphorin 3A (Sema 3A) acts as a potent suppressor of tumor angiogenesis in various cancer models. However, the function of Sema 3A in regulation of melanoma progression is not well studied, and yet to be the subject of intense investigation.</p> <h3>Methodology/Principal Findings</h3><p>In this study, using multiple <em>in vitro</em> and <em>in vivo</em> approaches we have demonstrated that Sema 3A acts as a potent tumor suppressor <em>in vitro</em> and <em>in vivo</em> mice (C57BL/6) models. Mouse melanoma (B16F10) cells overexpressed with Sema 3A resulted in significant inhibition of cell motility, invasiveness and proliferation as well as suppression of <em>in vivo</em> tumor growth, angiogenesis and metastasis in mice models. Moreover, we have observed that Sema 3A overexpressed melanoma clone showed increased sensitivity towards curcumin and Dacarbazine, anti-cancer agents.</p> <h3>Conclusions</h3><p>Our results demonstrate, at least in part, the functional approach underlying Sema 3A mediated inhibition of tumorigenesis and angiogenesis and a clear understanding of such a process may facilitate the development of novel therapeutic strategy for the treatment of cancer.</p> </div

    Co-Crystal Structures of Inhibitors with MRCKβ, a Key Regulator of Tumor Cell Invasion

    MRCKα and MRCKβ (myotonic dystrophy kinase-related Cdc42-binding kinases) belong to a subfamily of Rho GTPase activated serine/threonine kinases within the AGC-family that regulate the actomyosin cytoskeleton. Reflecting their roles in myosin light chain (MLC) phosphorylation, MRCKα and MRCKβ influence cell shape and motility. We report further evidence for MRCKα and MRCKβ contributions to the invasion of cancer cells in 3-dimensional matrix invasion assays. In particular, our results indicate that the combined inhibition of MRCKα and MRCKβ together with inhibition of ROCK kinases results in significantly greater effects on reducing cancer cell invasion than blocking either MRCK or ROCK kinases alone. To probe the kinase ligand pocket, we screened 159 kinase inhibitors in an in vitro MRCKβ kinase assay and found 11 compounds that inhibited enzyme activity >80% at 3 µM. Further analysis of three hits, Y-27632, Fasudil and TPCA-1, revealed low micromolar IC50 values for MRCKα and MRCKβ. We also describe the crystal structure of MRCKβ in complex with inhibitors Fasudil and TPCA-1 bound to the active site of the kinase. These high-resolution structures reveal a highly conserved AGC kinase fold in a typical dimeric arrangement. The kinase domain is in an active conformation with a fully-ordered and correctly positioned αC helix and catalytic residues in a conformation competent for catalysis. Together, these results provide further validation for MRCK involvement in regulation of cancer cell invasion and present a valuable starting point for future structure-based drug discovery efforts

    Streblo: the app prototype for managing stress in the construction industry

    High levels of stress at work, great responsibilities, hazards and lack of balance between tasks and deadlines are common in the daily lives of many construction workers. Changing the patterns of thinking and behaviours is not an easy mission, and it requires constant support, learning and determination. E-health applications can contribute to this change through their ability to foster continuous interaction with the user. Mobile phone apps have shown promising results in the field of ‘e-health and wellbeing’. Accordingly, an App is being designed as a self-help system for stress management which will enable construction workers to 1) detect the onset of stress quite early, 2) track their stress status, 3) empower persons to cope with stressful and/or demanding situations in an adaptive way, 4) improve and streamline the operability of job tasks, and 5) optimise efficient solutions for the construction industry. The development of this innovative app, known as Streblo, is part of a wider research that is studying stress management in the construction industry. Streblo’s blueprint will match personality traits with coping strategies in real- life situations. Its inputs are being generated from a field study that has commenced, where 23 structured interviews have been used to collect data from construction workers on their 1) personality and 2) behaviours while experiencing stress. Results of the data collection and analysis are being used to develop Streblo (an App) with IT experts. The paper reports the detail development and performance of Streblo’s prototype. Ultimately, users will be able to engage Streblo on electronic devices (mobile phones, tablets, and computers) through both text and image-based communication obtain real-time solutions and feedbacks on their stress status. Streblo will enhance and support attitude and behavioural changes in people who suffer from stress symptoms in the construction industry.Streblo is being developed within a European Commission (EC) project H2020-MSCA- IF-2015/H2020-MSCA-IF-2015, Grant Agreement: 703236 - ‘Inhibiting Stress in the Construction Industry’ (INSTINCT).     Kita Driven Expression of Oncogenic HRAS Leads to Early Onset and Highly Penetrant Melanoma in Zebrafish

    Melanoma is the most aggressive and lethal form of skin cancer. Because of the increasing incidence and high lethality of melanoma, animal models for continuously observing melanoma formation and progression as well as for testing pharmacological agents are needed.Using the combinatorial Gal4-UAS system, we have developed a zebrafish transgenic line that expresses oncogenic HRAS under the kita promoter. Already at 3 days transgenic kita-GFP-RAS larvae show a hyper-pigmentation phenotype as earliest evidence of abnormal melanocyte growth. By 2-4 weeks, masses of transformed melanocytes form in the tail stalk of the majority of kita-GFP-RAS transgenic fish. The adult tumors evident between 1-3 months of age faithfully reproduce the immunological, histological and molecular phenotypes of human melanoma, but on a condensed time-line. Furthermore, they show transplantability, dependence on mitfa expression and do not require additional mutations in tumor suppressors. In contrast to kita expressing melanocyte progenitors that efficiently develop melanoma, mitfa expressing progenitors in a second Gal4-driver line were 4 times less efficient in developing melanoma during the three months observation period.This indicates that zebrafish kita promoter is a powerful tool for driving oncogene expression in the right cells and at the right level to induce early onset melanoma in the presence of tumor suppressors. Thus our zebrafish model provides a link between kita expressing melanocyte progenitors and melanoma and offers the advantage of a larval phenotype suitable for large scale drug and genetic modifier screens
