Functional plasticity in the type IV secretion system of Helicobacter pylori.
Helicobacter pylori causes clinical disease primarily in those individuals infected with a strain that carries the cytotoxin associated gene pathogenicity island (cagPAI). The cagPAI encodes a type IV secretion system (T4SS) that injects the CagA oncoprotein into epithelial cells and is required for induction of the pro-inflammatory cytokine, interleukin-8 (IL-8). CagY is an essential component of the H. pylori T4SS that has an unusual sequence structure, in which an extraordinary number of direct DNA repeats is predicted to cause rearrangements that invariably yield in-frame insertions or deletions. Here we demonstrate in murine and non-human primate models that immune-driven host selection of rearrangements in CagY is sufficient to cause gain or loss of function in the H. pylori T4SS. We propose that CagY functions as a sort of molecular switch or perhaps a rheostat that alters the function of the T4SS and "tunes" the host inflammatory response so as to maximize persistent infection

Production of ethanol from refinery waste gases. Phase 3. Engineering development. Annual report, April 1, 1995--May 15, 1996
Refineries discharge large volumes of H2, CO, and CO 2 from cracking, coking, and hydrotreating operations. This R&D program seeks to develop, demonstrate, and commercialize a biological process for converting these waste gases into ethanol for blending with gasoline. A 200,000 BPD refinery could produce up to 38 million gallons ethanol per year. The program is being conducted in 3 phases: II, technology development; III, engineering development; and IV, demonstration. Phase I, exploratory development, has been completed. The research effort has yielded two strains (Isolates O-52 and C-01) which are to be used in the pilot studies to produce ethanol from CO, CO2, and H2 in petroleum waste gas. Results from single continuous stirred tank reactor (CSTR) laboratory tests have shown that 20-25 g/L ethanol can be produced with < 5 g/L acetic acid byproduct. Laboratory studies with two CSTRs in series have yielded ethanol concentrations of 30-35 g/L with 2-4 g/L acetic acid byproduct. Water recycle from distillation back to the fermenter shows that filtration of the water before distillation eliminates the recycle of toxic materials back to the fermenter. Product recovery in the process will use direct distillation to the azeotrope, followed by adsorption to produce neat ethanol. This is less energy intensive than e.g. solvent extraction, azeotropic distillation, or pervaporation. Economic projections are quite attractive; the economics are refinery stream dependent and thus vary depending on refinery location and operation

Production of ethanol from refinery waste gases. Final report, April 1994--July 1997
The objective of this program was to develop a commercial process for producing ethanol from refinery waste gases. this report presents results from the development phases. The major focus of this work was the preparation of the prototype design which will demonstrate this technology in a 2.5 lb/hr ethanol production facility. Additional areas of focus included efforts in obtaining an industrial partner to help finance the prototype, and advanced engineering experiments concentrating on process optimization in various areas needing future development and optimization. The advanced engineering experiments were performed in the laboratory in these areas: treatment and use of recycle water from distillation back to fermentation; alternative methods of removing cells from the fermentation broth; the fermentation of streams containing CO{sub 2}/H{sub 2} alone, with little to no CO present; dealing with methanogen contaminants that are capable of fermenting CO{sub 2} and H{sub 2} to methane; and acetate tolerance by the culture. Results from the design, industrial partner search and the laboratory R&D efforts are discussed in this report
The Re-Emerging Role of the State in Contemporary Russia
I examine ownership structure of Russian firms during the 1998-2006 period, where a greater emphasis is placed on motivations behind increased government ownership in the latter years, when oligarchs' opportunistic influence on the firm diminished as state ownership correspondingly increased. As this phenomenon is also correlated with improved corporate growth during the period, I argue that state participation in corporate governance acted as an effective substitute mechanism to constrain wealth-tunnelling behaviour of corporate insiders and local bureaucrats in a country defined by a weak property rights system. © 2012 Springer-Verlag

Deciphering the Multifactorial Nature of Acinetobacter baumannii Pathogenicity
Background: Acinetobacter baumannii is an emerging bacterial pathogen that causes a broad array of infections, particularly in hospitalized patients. Many studies have focused on the epidemiology and antibiotic resistance of A. baumannii, but little is currently known with respect to its virulence potential. Methodology/Principal Findings: The aim of this work was to analyze a number of virulence-related traits of four A. baumannii strains of different origin and clinical impact for which complete genome sequences were available, in order to tentatively identify novel determinants of A. baumannii pathogenicity. Clinical strains showed comparable virulence in the Galleria mellonella model of infection, irrespective of their status as outbreak or sporadic strains, whereas a non-human isolate was avirulent. A combined approach of genomic and phenotypic analyses led to the identification of several virulence factors, including exoproducts with hemolytic, phospholipase, protease and iron-chelating activities, as well as a number of multifactorial phenotypes, such as biofilm formation, surface motility and stress resistance, which were differentially expressed and could play a role in A. baumannii pathogenicity. Conclusion/Significance: This work provides evidence of the multifactorial nature of A. baumannii virulence. Improving survey methods in sero-epidemiological studies of injecting drug users: a case example of two cross sectional surveys in Serbia and Montenegro
BACKGROUND: Little is known about the prevalence of HIV or HCV in injecting drug users (IDUs) in Serbia and Montenegro. We measured prevalence of antibodies to HIV (anti-HIV) and hepatitis C virus (anti-HCV), and risk factors for anti-HCV, in community-recruited IDUs in Belgrade and Podgorica, and determined the performance of a parallel rapid HIV testing algorithm.
METHODS: Respondent driven sampling and audio-computer assisted survey interviewing (ACASI) methods were employed. Dried blood spots were collected for unlinked anonymous antibody testing. Belgrade IDUs were offered voluntary confidential rapid HIV testing using a parallel testing algorithm, the performance of which was compared with standard laboratory tests. Predictors of anti-HCV positivity and the diagnostic accuracy of the rapid HIV test algorithm were calculated.
RESULTS: Overall population prevalence of anti-HIV and anti-HCV in IDUs were 3% and 63% respectively in Belgrade (n = 433) and 0% and 22% in Podgorica (n = 328). Around a quarter of IDUs in each city had injected with used needles and syringes in the last four weeks. In both cities anti-HCV positivity was associated with increasing number of years injecting (eg Belgrade adjusted odds ratio (AOR) 5.6 (95% CI 3.2-9.7) and Podgorica AOR 2.5 (1.3-5.1) for >or= 10 years v 0-4 years), daily injecting (Belgrade AOR 1.6 (1.0-2.7), Podgorica AOR 2.1 (1.3-5.1)), and having ever shared used needles/syringes (Belgrade AOR 2.3 (1.0-5.4), Podgorica AOR 1.9 (1.4-2.6)). Half (47%) of Belgrade participants accepted rapid HIV testing, and there was complete concordance between rapid test results and subsequent confirmatory laboratory tests (sensitivity 100% (95%CI 59%-100%), specificity 100% (95%CI 98%-100%)).
CONCLUSION: The combination of community recruitment, ACASI, rapid testing and a linked diagnostic accuracy study provide enhanced methods for conducting blood borne virus sero-prevalence studies in IDUs. The relatively high uptake of rapid testing suggests that introducing this method in community settings could increase the number of people tested in high risk populations. The high prevalence of HCV and relatively high prevalence of injecting risk behaviour indicate that further HIV transmission is likely in IDUs in both cities. Urgent scale up of HIV prevention interventions is needed
Stereochemical Insignificance Discovered in Acinetobacter baumannii Quorum Sensing
Stereochemistry is a key aspect of molecular recognition for biological systems. As such, receptors and enzymes are often highly stereospecific, only recognizing one stereoisomer of a ligand. Recently, the quorum sensing signaling molecules used by the nosocomial opportunistic pathogen, Acinetobacter baumannii, were identified, and the primary signaling molecule isolated from this species was N-(3-hydroxydodecanoyl)-l-homoserine lactone. A plethora of bacterial species have been demonstrated to utilize 3-hydroxy-acylhomoserine lactone autoinducers, and in virtually all cases, the (R)-stereoisomer was identified as the natural ligand and exhibited greater autoinducer activity than the corresponding (S)-stereoisomer. Using chemical synthesis and biochemical assays, we have uncovered a case of stereochemical insignificance in A. baumannii and provide a unique example where stereochemistry appears nonessential for acylhomoserine lactone-mediated quorum sensing signaling. Based on previously reported phylogenetic studies, we suggest that A. baumannii has evolutionarily adopted this unique, yet promiscuous quorum sensing system to ensure its survival, particularly in the presence of other proteobacteria
