    Multilevel Analysis of Oscillation Motions in Active Regions of the Sun

    We present a new method that combines the results of an oscillation study made in optical and radio observations. The optical spectral measurements in photospheric and chromospheric lines of the line-of-sight velocity were carried out at the Sayan Solar Observatory. The radio maps of the Sun were obtained with the Nobeyama Radioheliograph at 1.76 cm. Radio sources associated with the sunspots were analyzed to study the oscillation processes in the chromosphere-corona transition region in the layer with magnetic field B=2000 G. A high level of instability of the oscillations in the optical and radio data was found. We used a wavelet analysis for the spectra. The best similarities of the spectra of oscillations obtained by the two methods were detected in the three-minute oscillations inside the sunspot umbra for the dates when the active regions were situated near the center of the solar disk. A comparison of the wavelet spectra for optical and radio observations showed a time delay of about 50 seconds of the radio results with respect to optical ones. This implies a MHD wave traveling upward inside the umbral magnetic tube of the sunspot. Besides three-minute and five-minute ones, oscillations with longer periods (8 and 15 minutes) were detected in optical and radio records.Comment: 17 pages, 9 figures, accepted to Solar Physics (18 Jan 2011).     Erratum. Maternal ageing impairs mitochondrial DNA kinetics during early embryogenesis in mice

    STUDY QUESTION: Does ageing affect the kinetics of the mitochondrial pool during oogenesis and early embryogenesis? SUMMARY ANSWER: While we found no age-related change during oogenesis, the kinetics of mitochondrial DNA content and the expression of the factors involved in mitochondrial biogenesis appeared to be significantly altered during embryogenesis. WHAT IS KNOWN ALREADY: Oocyte mitochondria are necessary for embryonic development. The morphological and functional alterations of mitochondria, as well as the qualitative and quantitative mtDNA anomalies, observed during ovarian ageing may be responsible for the alteration of oocyte competence and embryonic development. STUDY DESIGN, SIZE, DURATION: The study, conducted from November 2016 to November 2017, used 40 mice aged 5-8 weeks and 45 mice aged 9-11 months (C57Bl6/CBA F(1)). A total of 488 immature oocytes, with a diameter ranging from 20 μm to more than 80 μm, were collected from ovaries, and 1088 mature oocytes or embryos at different developmental stages (two PN, one-cell, i.e. syngamy, two-cell, four-cell, eight-cell, morula and blastocyst) were obtained after ovarian stimulation and, for embryos, mating. PARTICIPANTS/MATERIALS, SETTING, METHODS: Mitochondrial DNA was quantified by quantitative PCR. We used quantitative reverse transcriptase PCR (RT-PCR) (microfluidic method) to study the relative expression of three genes involved in the key steps of embryogenesis, i.e. embryonic genome activation (HSPA1) and differentiation (CDX2 and NANOG), two mtDNA genes (CYB and ND2) and five genes essential for mitochondrial biogenesis (PPARGC1A, NRF1, POLG, TFAM and PRKAA). The statistical analysis was based on mixed linear regression models applying a logistic link function (STATA v13.1 software), with values of P < 0.05 being considered significant. MAIN RESULTS AND THE ROLE OF CHANCE: During oogenesis, there was a significant increase in oocyte mtDNA content (P < 0.0001) without any difference between the two groups of mice (P = 0.73). During the first phase of embryogenesis, i.e. up to the two-cell stage, embryonic mtDNA decreased significantly in the aged mice (P < 0.0001), whereas it was stable for young mice (young/old difference P = 0.015). The second phase of embryogenesis, i.e. between the two-cell and eight-cell stages, was characterized by a decrease in embryonic mtDNA for young mice (P = 0.013) only (young/old difference P = 0.038). During the third phase, i.e. between the eight-cell and blastocyst stage, there was a significant increase in embryonic mtDNA content in young mice (P < 0.0001) but not found in aged mice (young/old difference P = 0.002). We also noted a faster expression of CDX2 and NANOG in the aged mice than in the young mice during the second (P = 0.007 and P = 0.02, respectively) and the third phase (P = 0.01 and P = 0.008, respectively) of embryogenesis. The expression of mitochondrial genes CYB and ND2 followed similar kinetics and was equivalent for both groups of mice, with a significant increase during the third phase (P < 0.01). Of the five genes involved in mitochondrial biogenesis, i.e. PPARGC1A, NRF1, POLG, TFAM and PRKAA, the expression of three genes decreased significantly during the first phase only in young mice (NRF1, P = 0.018; POLGA, P = 0.002; PRKAA, P = 0.010), with no subsequent difference compared to old mice. In conclusion, during early embryogenesis in the old mice, we suspect that the lack of a replicatory burst before the two-cell stage, associated with the early arrival at the minimum threshold value of mtDNA, together with the absence of an increase of mtDNA during the last phase, might potentially deregulate the key stages of early embryogenesis. LARGE SCALE DATA: N/A. LIMITATIONS, REASONS FOR CAUTION: Because of the ethical impossibility of working on a human, this study was conducted only on a murine model. As superovulation was used, we cannot totally exclude that the differences observed were, at least partially, influenced by differences in ovarian response between young and old mice. WIDER IMPLICATIONS OF THE FINDINGS: Our findings suggest a pathophysiological explanation for the link observed between mitochondria and the deterioration of oocyte quality and early embryonic development with age.     RUSSIAN GRAIN FROM SIBERIA: EXPORT POTENTIAL, PRIORITY SALES MARKETS

    The study identifies the possibilities of the grain complex of Siberia to supply grain for export. The role of the state and the main regulatory documents of the federal and regional levels, aimed at the development of the grain market and its export from Siberia, have been determined. Using data from the Federal Customs Service of Russia, an analysis of grain exports by regions of Siberia and grain crops for 2017-2019 was carried out. The factors hindering the export of Siberian grain have been warned. The study presents a forecast of grain exports by the regions of Siberia until 2025, as well as its promising directions and priority sales markets. To identify trends in the development of grain exports, the main commodity groups (wheat, rye, barley, oats, buckwheat) were considered. Priority sales markets have been identified, to which supplies should be increased, while solving transport and logistics problems, overcoming tariff and non-tariff barriers

    Drug related problems and pharmacist interventions in a geriatric unit employing electronic prescribing

    Background Computerised physician order entry (CPOE) and the integration of a pharmacist in clinical wards have been shown to prevent drug related problems (DRPs). Objectives The primary objective was to make an inventory of the DRPs and resident pharmacist on-ward interventions (PIs) identified in a geriatric acute care unit using CPOE system. The secondary objective was to evaluate the physicians\u27 acceptance of the proposed interventions. Setting A 26-bed geriatric ward of a 1,300-bed teaching hospital. Method A 6-month descriptive study with prescription analysis and recommendations to physicians by a resident pharmacist during five half days a week. Main outcome measures Patients\u27 characteristics, number of prescribed drugs per patient, nature and frequency of DRPs and PIs, physicians\u27 acceptance and drugs questioned. Results Resident pharmacist reviewed 311 patients and identified 241 DRPs. One hundred and fifty-two patients (49 %) had at least one DRP (mean +/- A SD age 87 +/- A 6 years, mean +/- A SD number of prescribed drugs 10.7 +/- A 3.4). Most frequent DRPs were: untreated indication (n = 58, 24.1 %), dose too high (n = 46, 19.1 %), improper administration (n = 31, 12.9 %) and drug interactions (n = 23, 9.5 %). The rate of physicians\u27 acceptance was 90.0 % (7.5 % refusals, 2.5 % not assessable). DRPs related to CPOE system misuse (n = 35, 14.5 %) appeared as a worrying phenomenon (e.g., errors in selecting dosage or unit, or duplication of therapy). Conclusion A resident pharmacist detected various DRPs. Most PIs were accepted. DRPs related to the misuse of the CPOE system appeared potentially dangerous and need particular attention by healthcare professionals. The description of the DRPs is an essential step for implementation of targeted clinical pharmacy services in order to optimize pharmacists\u27 job time

    Development of the concept of an innovative laboratory installation for the study of dust-forming surfaces

    Currently, the determination of the emission rate of suspended solids from a unit of the surface area of a man-made mass at various parameters of the wind flow is not sufficiently described. The analysis of the world experience of researchers shows that existing laboratory installations have various design features that do not allow to correctly determine the mass of the dust being flapped and wind-blown. Based on the analysis results, the concept of an innovative laboratory installation for the study of dust-forming surfaces has been developed. It takes into account the influence of wind shadows, the deturbulization of an artificially created air flow, the possibility of regulating not only the flow velocity mode, but also the creation of a vacuum or disturbance in the area of sample placement, as well as the formation of a certain angle of wind flow attack relative to the surface. The concept provides for the possibility of determining the volume of dust emissions by the values of the lost dust masses in the sample and by the values of dust concentrations in the outgoing stream. The calculation of the main basic elements of the installation using the ANSYS FLUENT software package was carried out. The model and configuration of the wind tunnel have been developed and calculated, the main geometric parameters and functional elements for the possibility of use in scientific work have been determined. For practical use of the empirical roughness value of the underlying surface, its values are recommended in a wide range – from zero for the water surface to 0.44 for large cities with tall buildings and skyscrapers

    Nouméa: a new multi-mission calibration and validation site for past and future altimetry missions?

    Today, monitoring the evolution of sea level in coastal areas is of importance, since almost 11 % of the world's population lives in low-lying areas. Reducing uncertainties in sea level estimates requires a better understanding of both altimetry measurements and local sea level dynamics. In New Caledonia, the Nouméa lagoon is an example of this challenge, as altimetry, coastal tide gauge, and vertical land motions from global navigation satellite systems (GNSSs) do not provide consistent information. The GEOCEAN-NC 2019 field campaign addresses this issue with deployments of in situ instruments in the lagoon (GNSS buoy, pressure gauge, etc.), with a particular focus on the crossover of one Jason-series track and two Sentinel-3A missions tracks. In this study, we propose a method to virtually transfer the Nouméa tide gauge at the altimetry crossover point, using in situ data from the field campaign. Following the philosophy of calibration and validation (Cal/Val) studies, we derive absolute altimeter bias time series over the entire Jason and Sentinel-3A periods. Overall, our estimated altimeter mean biases are slightly larger by 1–2 cm compared to Corsica and Bass Strait results, with inter-mission biases in line with those of Bass Strait site. Uncertainties still remain regarding the determination of our vertical datum, only constrained by the three days of the GNSS buoy deployment. With our method, we are able to re-analyse about 20 years of altimetry observations and derive a linear trend of −0.2 ± 0.1 mm yr−1 over the bias time series. Compared to previous studies, we do not find any significant uplift in the area, which is more consistent with the observations of inland permanent GNSS stations. These results support the idea of developing Cal/Val activities in the lagoon, which is already the subject of several experiments for the scientific calibration phase of the SWOT wide-swath altimetry mission.</p

    Lipidomics Reveals Triacylglycerol Accumulation Due to Impaired Fatty Acid Flux in Opa1-Disrupted Fibroblasts

    OPA1 is a dynamin GTPase implicated in mitochondrial membrane fusion. Despite its involvement in lipid remodeling, the function of OPA1 has never been analyzed by whole-cell lipidomics. We used a nontargeted, reversed-phase lipidomics approach, validated for cell cultures, to investigate OPA1-inactivated mouse embryonic fibroblasts ( Opa1 MEFs). This led to the identification of a wide range of 14 different lipid subclasses comprising 212 accurately detected lipids. Multivariate and univariate statistical analyses were then carried out to assess the differences between the Opa1 and Opa1 genotypes. Of the 212 lipids identified, 69 were found to discriminate between Opa1 MEFs and Opa1 MEFs. Among these lipids, 34 were triglycerides, all of which were at higher levels in Opa1 MEFs with fold changes ranging from 3.60 to 17.93. Cell imaging with labeled fatty acids revealed a sharp alteration of the fatty acid flux with a reduced mitochondrial uptake. The other 35 discriminating lipids included phosphatidylcholines, lysophosphatidylcholines, phosphatidylethanolamine, and sphingomyelins, mainly involved in membrane remodeling, and ceramides, gangliosides, and phosphatidylinositols, mainly involved in apoptotic cell signaling. Our results show that the inactivation of OPA1 severely affects the mitochondrial uptake of fatty acids and lipids through membrane remodeling and apoptotic cell signaling

    The mitochondrial DNA content of cumulus cells may help predict embryo implantation

    PURPOSE: The quantification of mtDNA in cumulus granulosa cells (CGCs) surrounding an oocyte has been positively linked with morphological embryonic quality. In the present study, we evaluated the link between the amount of mtDNA in CGCs surrounding an oocyte and the chances for the corresponding embryo of implanting and leading to an ongoing pregnancy. METHODS: This is an observational study, performed on 84 oocyte-cumulus-complexes (OCCs) having led to the replacement of an embryo in the maternal uterus, retrieved from 71 patients undergoing IVF with intracytoplasmic sperm. The OCCs were classified in two groups, one including 26 OCCs having led to an implanted embryo and the other including 58 OCCs having led to a non-implanted embryo. The average mtDNA content of CGCs was assessed by using a quantitative real-time PCR technique. RESULTS: Significantly higher mtDNA copy numbers in CGCs were associated with implanted embryos than with non-implanted embryos (mean 215 [sd 375] and 59 [sd 72], respectively; p &lt; 10). Multivariate analysis, taking into account the women\u27s age, the embryo quality, and the AMH level, suggests an independent relationship between the mtDNA content of CGCs and the potential of embryo implantation. CONCLUSION: During in vitro fertilization (IVF) procedures, the probability of the implantation of the embryo appears to be closely correlated to the mtDNA copy numbers in the CGCs. Our results highlight the interest of mtDNA quantification in GCGs as a biomarker of the potential of embryo implantation
