Thawing condition is one the most important factors affecting the re-animation of the spermatozoon in order to fertilise the oocyte. For that matter, we aimed to evaluate the morphological features of the head and midpiece of cryopreserved spermatozoa thawed at different temperatures and for various durations, with using CASA (Computer aided sperm analyser). Frozen semen samples belonging to the same batch, collected from three different bulls were grouped as; control group thawed for 20 seconds at 37 ℃; and experimental groups were thawed for 30, 40, 50 and 60 s at 25 ℃ and 37 ℃; for 10, 15, 20 and 25 s at 40 ℃; for 3, 6, 9, 12 s at 70 ℃. Morphometrical features of the samples were evaluated by using CASA system with nine repetitions. As a result, straws thawed at 25℃ for 40 s had the highest average length of head (6.22 ± 0.09 μm), and the width of midpiece (0.68 ± 0.01 μm). It was concluded that the thawing temperature and duration has affected/altered the morphometry of the sperm head and midpiece, although the results were not statistically significant (p> 0.005).Suni tohumlamada kullanılan dondurulmuş spermanın çözüm sonu parametrelerini belirleyen faktörlerin başında çözdürme koşulları gelmektedir.

    Get PDF
The number of people with speech and language disorders is increasing every day (McLeod and McKinnon, 2007). Preschool teachers play a key role in diagnosing and treatment of these disorders since children acquire speech and language abilities rapidly in the preschool period. Speech and language therapy is a new discipline in Turkey. Therefore, the aim of this study is to determine the knowledge levels of preschool teachers in Turkey about children with speech and language disorders. The sample consisted of 153 preschool teachers from Kocaeli, Turkey and a self-administered Teacher Knowledge of Speech/Language Impairment Questionnaire was completed by the participants. Results revealed that knowledge and awareness levels of preschool teachers about children with speech/language impairment are limited. While 46% of the participants report that they have little or no experience with speech/language impaired children and do not feel themselves adequate in teaching children with severe language problems. Workshops designed for preschool teachers about speech and language disorders may offer opportunities to learn about the resources and classroom strategies that are proven successful in teaching students with speech and language disorders in Turkey

    Get PDF
Aim: To evaluate the susceptibility of the serotonin transporter-associated polymorphic region (5-HTTLPR) and complement factor H (CFH) gene variants to smoking status. Method: Smokers and non-smokers were included in the study. The smoking amount was assessed based on the scores on the Fagerström Test for Nicotine Dependence (FTND). DNA is extracted from blood samples. 5-HTTLPR and CFH Y402H variants were analyzed by polymerase chain reaction (PCR) and/or restriction fragment length polymorphism (RFLP) methods. The results were evaluated statistically. Results: 5-HTTLPR and CFH Y402H genotype and allele distribution did not differ significantly between smokers and non-smokers (p>0.05). There was no deviation from Hardy-Weinberg equilibrium (HWE) for these variants in the groups. Conclusion: Nicotine addiction is a complex phenomenon in which both genetic and environmental factors play a role. The identification of genes that play a role in addiction is important to elucidate the pathogenesis. The results of this study showed that 5-HTTLPR and CFH Y402H variants had no effect on nicotine addiction. However, these results need to be validated in larger sample groups and in different ethnic communities.   &nbsp

This study examines the serial multiple mediation of social appearance anxiety, positive perfectionism, and negative perfectionism in the relationship between self-compassion and resilience to observe the psychosocial development of children attending secondary school. The model of this study, which examines the role of social appearance anxiety, positive perfectionism, and negative perfectionism in the relationship between secondary school children's self-compassion and resilience, is a relational research type, which is one of the quantitative research types. Relational research method is used to determine the relationships between variables and to predict possible outcomes. A total of 497 children, 283 girls (59.1%) and 196 (40.9%) boys were reached for the study group. 55 (11.5%) of the children stated that they had a low socioeconomic level, 399 (83.3%) had a medium socioeconomic level, and 25 (5.2%) had a high socioeconomic level. Children's ages range from 10 to 14 years old. In this study, Self-Compassion Scale-Short Form, Psychological Resilience Scale, Social Appearance Anxiety Scale, Positive-Negative Perfectionism Scale, and Personal Information Form were used to collect data. Firstly, descriptive statistics for all variables were calculated, and the relations between variables were determined using Pearson Product Moments Correlation analysis. Mediation analyses were performed in line with the recommendations of Preacher and Hayes. This method is used by loading the "Multiple Mediation (INDIRECT)" macro into the IBM SPSS Statistic 22 software. Model 6, one of the serial multiple mediator variable models, was used in the analysis. Finally, according to the results of serial multiple analysis, it was observed that the mediation role of social appearance anxiety, positive perfectionism, and negative perfectionism was significant in the relationship between self-compassion and psychological resilience. The model shows that children with high self-compassion will experience less social appearance anxiety, have more positive and less negative perfectionism, and thus have higher psychological resilience

Two Infant Cases Admitted with Atypical Presentation and Diagnosed as Type IV Hiatal Hernia

    Get PDF
Type IV paraesophageal hiatal hernias are diaphragmatic hernias that are extremely rare in the pediatric age group in which the stomach and other intra-abdominal organs herniate from the esophageal hiatus of the diaphragm into the mediastinum. Since the defect in the hiatus is large, serious complications such as gastric volvulus may develop in these giant hernias as a result of the passage of a large part of the stomach into the thoracic cavity. Patients may present with atypical clinical manifestations such as growth retardation, not gaining weight, and recurrent pulmonary infections. In this report, two cases in the pediatric age group who were admitted with different clinical course and were diagnosed as type IV paraesophageal hiatal hernia who were successfully treated with surgical intervention are presented

Objectives: Schizophrenia (Sch) is a severe and chronic mental illness. Smoking prevalence is higher in patients with Sch than general population. We aimed to investigate the effects of MAOB gene A644G variant on nicotine dependence (ND) and Sch+ND risk in Turkish population and to evaluate by bioinformatic analysis. Methods: Present study included 161 individuals with ND, 223 patients with Sch+ND, and 96 non-smoker controls. MAOB A644G variant was analyzed using PCR-RFLP method. As the MAOB gene is located on the X chromosome, each gender was analysed separately. Results: The total distributions of AA, AG and GG genotypes of MAOB gene A644G were 44.7%, 22.4% and 32.9% in the ND group, 45.3%, 25.1% and 29.6% in the Sch+ND group and, 44.8, 22.9% and 32.3% in non-smoker controls. No significant differences were observed between groups for the MAOB A644G genotype and allele frequencies when female group compared to male group (p > 0.05). Examination of disease associations of SNPs from each miRNA gene region in GWAS databases yielded results for aging, bipolar disorder, autoimmune, and neurological diseases. Discussion: Our results indicate that the MAOB gene A644G variant is not associated with ND and/or Sch susceptibility in the Turkish population

Case report: Revealing the rare—a Brody Disease patient from Turkey expanding the phenotype

    Get PDF
Brody Disease is an exceptionally rare, autosomal recessive myopathy attributed to the pathogenic variants in the ATP2A1, which encodes the sarcoplasmic/endoplasmic reticulum Ca (2+) ATPase type 1 protein SERCA1. It was first described by Brody IA in 1969. To date, only thirty-three Brody families with forty-seven patients have been reported in the literature, and the disease prevalence is considered as 1 in 10 million, demonstrating the peculiarity of the disease. Clinical characteristics of Brody Disease include muscle stiffness after exercise, myalgia, and muscle cramps. Brody Disease patients generally have disease onset in the first decade, and genetic diagnosis is delayed as a consequence of both the rareness and the mild course of the disease. Here, we report a Turkish Brody Disease patient with a homozygous c.428G>A p.Arg143Gln (NM_004320.4) missense mutation in the ATP2A1. The male patient, whose symptoms started at the age of 14–15, is now 36 years old. His clinical manifestations are athletic appearance, exotropia, slightly elevated creatine kinase (CK), mild progressive proximal muscle weakness in the lower extremities, muscle cramps, pain and stiffness. The patient described here has a very mild progression with an onset in the second decade, expanding the Brody Disease phenotype. The study also implies that in the era of emerging genetic therapies, the routine testing of patients with myopathies is a prerequisite since not only future therapies will be designed on molecular findings, but also currently available symptomatic and palliative treatment options will be more precisely applied
