    The plight of camels eating plastic waste

    Embargo until 23 October 2022Ecological impacts of plastic pollution are widespread, in all biomes and geographies. Here, we report the ingestion of anthropogenic waste, primarily plastic bags and rope by dromedary camels (Camelus dromedarius) in the United Arab Emirates (UAE), which has led to a regional mortality rate of 1%. We define the ingested waste as a polybezoar, a collection of tightly packed indigestible materials which can include plastics, ropes, other litter and salt deposits trapped in the stomach or digestive tract forming a large stone-like mass. In the Central Veterinary Research Laboratory (CVRL) in Dubai, UAE, of the more than 30,000 camels evaluated from the region since 2008, there have been 300 camels observed post-mortem with polybezoars in their stomach, from both camels in the CRVL or recovered from desiccated skeletons found in the desert. Here, we analyze a subset of five polybezoars ranging from 6.2 to 63.6 kg. Polybeozars lead to gastrointestinal blockages, sepsis from increased gut bacteria, dehydration and malnutrition. Due to high winds and the open desert environment, plastic bags and other film packaging escape open waste bins and landfills, traveling long distances; therefore we suggest improved waste management and alternative systems to package and deliver goods throughout the region.acceptedVersio

    There is accumulating evidence that birds of prey are susceptible to fatal infection with highly pathogenic avian influenza (HPAI) virus. We studied the antigenic, molecular, phylogenetic, and pathogenic properties of 2 HPAI H5N1 viruses isolated from dead falcons in Saudi Arabia and Kuwait in 2005 and 2007, respectively. Phylogenetic and antigenic analyses grouped both isolates in clade 2.2 (Qinghai-like viruses). However, the viruses appeared to have spread westward via different flyways. It remains unknown how these viruses spread so rapidly from Qinghai after the 2005 outbreak and how they were introduced into falcons in these two countries. The H5N1 outbreaks in the Middle East are believed by some to be mediated by wild migratory birds. However, sporting falcons may be at additional risk from the illegal import of live quail to feed them

    Use of a Western blot technique for the serodiagnosis of glanders

    <p>Abstract</p> <p>Background</p> <p>The in vivo diagnosis of glanders relies on the highly sensitive complement fixation test (CFT). Frequently observed false positive results are troublesome for veterinary authorities and cause financial losses to animal owners. Consequently, there is an urgent need to develop a test with high specificity. Hence, a Western blot assay making use of a partly purified lipopolysaccaride (LPS) containing antigen of three <it>Burkholderia mallei </it>strains was developed. The test was validated investigating a comprehensive set of positive and negative sera obtained from horses and mules from endemic and non endemic areas.</p> <p>Results</p> <p>The developed Western blot assay showed a markedly higher diagnostic specificity when compared to the prescribed CFT and therefore can be used as a confirmatory test. However, the CFT remains the test of choice for routine testing of glanders due to its high sensitivity, its feasibility using standard laboratory equipment and its worldwide distribution in diagnostic laboratories.</p> <p>Conclusions</p> <p>The CFT should be amended by the newly validated Western blot to increase the positive likelihood ratio of glanders serodiagnosis in non endemic areas or areas with low glanders prevalence. Its use for international trade of horses and mules should be implemented by the OIE.</p

    Comparison of commercial DNA preparation kits for the detection of Brucellae in tissue using quantitative real-time PCR

    <p>Abstract</p> <p>Background</p> <p>The detection of Brucellae in tissue specimens using PCR assays is difficult because the amount of bacteria is usually low. Therefore, optimised DNA extraction methods are critical. The aim of this study was to assess the performance of commercial kits for the extraction of <it>Brucella </it>DNA.</p> <p>Methods</p> <p>Five kits were evaluated using clinical specimens: QIAamp™ DNA Mini Kit (QIAGEN), peqGold™ Tissue DNA Mini Kit (PeqLab), UltraClean™ Tissue and Cells DNA Isolation Kit (MoBio), DNA Isolation Kit for Cells and Tissues (Roche), and NucleoSpin™ Tissue (Macherey-Nagel). DNA yield was determined using a quantitative real-time PCR assay targeting IS<it>711 </it>that included an internal amplification control.</p> <p>Results</p> <p>Kits of QIAGEN and Roche provided the highest amount of DNA, Macherey-Nagel and Peqlab products were intermediate whereas MoBio yielded the lowest amount of DNA. Differences were significant (p < 0.05) and of diagnostic relevance. Sample volume, elution volume, and processing time were also compared.</p> <p>Conclusions</p> <p>We observed differences in DNA yield as high as two orders of magnitude for some samples between the best and the worst DNA extraction kits and inhibition was observed occasionally. This indicates that DNA purification may be more relevant than expected when the amount of DNA in tissue is very low.</p
