    Predominant utilization of V beta 8+ T cell receptor genes in the H-2Ld- restricted cytotoxic T cell response against the immediate-early protein pp89 of the murine cytomegalovirus

    Cytotoxic T cell responses to the murine Cytomegalovirus (MCMV) were elicited in BALB/c mice (H-2d) by infectious virus. Eight days after infection, MCMV-primed local lymph node T cells were either depleted for T cells expressing a V beta 8+ TCR or separated into V beta 8+ and V beta 8- subpopulations by a cell sorter using the mAb F23.1. T cells were then expanded in vitro under limiting dilution conditions in the presence of IL-2 and in the absence of viral Ag to avoid selection by Ag in vitro. Frequencies of CTL precursors specific for the Immediate- Early-Ag 1 of MCMV and restricted to H-2Ld were determined. L cells of the endogenous haplotype H-2k cotransfected with the genes for MCMV-IE 1 and H-2Ld were used as target cells. Detection of a CTL response required previous priming of the animals by infection in vivo (less than 1/10(6) for nonimmunized animals). In primed animals CTL precursors of this specificity and restriction were three to fivefold more frequent in the V beta 8+ population (1/9.900 to 1/22.300) than in the V beta 8- population (1/57.000 to 1/87.200). Control experiments showed that frequencies were not influenced by the treatment with the anti-V beta 8-antibody and the fluorescein-labeled anti-Ig itself. V beta 8+ and V beta 8- T cells did not reveal any frequency differences when several other responses were determined (TNP-specific self- restricted CTL precursor; Th cells specific for keyhole limpet hemocyanin or Listeria monocytogenes)

    Non-Markovian master equation for a damped oscillator with time-varying parameters

    We derive an exact non-Markovian master equation that generalizes the previous work [Hu, Paz and Zhang, Phys. Rev. D {\bf 45}, 2843 (1992)] to damped harmonic oscillators with time-varying parameters. This is achieved by exploiting the linearity of the system and operator solution in Heisenberg picture. Our equation governs the non-Markovian quantum dynamics when the system is modulated by external devices. As an application, we apply our equation to parity kick decoupling problems. The time-dependent dissipative coefficients in the master equation are shown to be modified drastically when the system is driven by π\pi pulses. For coherence protection to be effective, our numerical results indicate that kicking period should be shorter than memory time of the bath. The effects of using soft pulses in an ohmic bath are also discussed

    Alloreactive cytotoxic T lymphocytes generated in the presence of viral- derived peptides show exquisite peptide and MHC specificity

    The nature of alloreactivity to MHC molecules has been enigmatic, primarily because of the observation that allogeneic responses are considerably stronger than syngeneic responses. To better determine the specificity potential of allogeneic responses, we have generated alloreactive CTL specific for exogenous, viral-derived peptide ligands. This approach allowed us to critically evaluate both the peptide- and MHC-specificity of these alloreactive T cells. Exploiting the accessibility of the H-2Ld class I molecule for exogenous peptide ligands, alloreactive CTL were generated that are specific for either murine cytomegalovirus (MCMV) or lymphocytic choriomeningitis virus (LCMV) peptides bound by Ld alloantigens. Peptide specificity was initially observed in bulk cultures of alloreactive CTL only when tested on peptide-sensitized T2.Ld target cells that have defective presentation of endogenous peptides. Subsequent cloning of bulk alloreactive CTL lines generated to MCMV yielded CTL clones that had exquisitely specific MCMV peptide recognition requirement. All of the MCMV/Ld alloreactive CTL clones were also exquisitely MHC-specific in that none of the CTL clones lysed targets expressing MCMV/Lq complexes, even though Lq differs from Ld by only six amino acid residues and Lq also binds the MCMV peptide. This observation clearly demonstrates that alloreactive CTL are capable of the same degree of specificity for target cell recognition as are syngeneic CTL in MHC-restricted responses

    Phytoplankton cell size: intra- and interspecific effects of warming and grazing

    Decreasing body size has been suggested as the third universal biological response to global warming after latitudinal/altitudinal range shifts and shifts in phenology. Size shifts in a community can be the composite result of intraspecific size shifts and of shifts between differently sized species. Metabolic explanations for the size shifts dominate in the literature but top down effects, i.e. intensified size-selective consumption at higher temperatures, have been proposed as alternative explanation. Therefore, we performed phytoplankton experiments with a factorial combination of warming and consumer type (protist feeding mainly on small algae vs. copepods mainly feeding on large algae). Natural phytoplankton was exposed to 3 (1st experiment) or 4 (2nd experiment) temperature levels and 3 (1st experiment: nano-, microzooplankton, copepods) or 2 (2nd experiment: microzooplankton, copepods) types of consumers. Size shifts of individual phytoplankton species and community mean size were analyzed. Both, mean cell size of most of the individual species and mean community cell size decreased with temperature under all grazing regimes. Grazing by copepods caused an additional reduction in cell size. Our results reject the hypothesis, that intensified size selective consumption at higher temperature would be the dominant explanation of decreasing body size. In this case, the size reduction would have taken place only in the copepod treatments but not in the treatments with protist grazing (nano- and microzooplankton)

    A variety of studies have demonstrated that motor disorders, parkinsonism and extrapyramidal motor symptoms (EPMS) are common in patients with Alzheimer's disease (AD). Several studies have reported an association of EPMS with severity, progression and poor prognosis of AD. The majority of these studies used clinical assessments for the rating of EPMS. In this study, kinematic handwriting analysis was used to quantify differences in fine hand motor function in patients with probable AD and mild cognitive impairment (MCl, as an assumed initial stage of AD) compared to depressed patients and healthy controls. Both patients with MCl and patients with probable AD exhibited loss of fine motor performance. Movements of AD patients were significantly less regular than those of healthy controls.     Kurzarbeit: Wichtige Stütze des Arbeitsmarktes in der Krise, aber keine Dauerlösung

    Kurzarbeit hat sich als ein wichtiges arbeitsmarktpolitisches Instrument bei der Bewältigung der Folgen der schwersten Wirtschaftskrise der Nachkriegszeit erwiesen. Dazu haben in erster Linie zeitlich befristete Neuregelungen zum Kurzarbeitergeld beigetragen, die diesen „Klassiker“ unter den arbeitsmarktpolitischen Instrumenten für die Unternehmen wieder deutlich attraktiver gemacht haben. Die Zahl der Kurzarbeiter erreichte im Mai des Jahres 2009 mit über 1,5 Millionen Personen einen Stand, der seit dem Jahr 1991 nicht mehr erreicht worden war. Bezogen auf 100 Arbeitslose standen 45 Arbeitnehmer in Kurzarbeit. Nur Mitte der 1970er Jahre und im Jahr 1991 wurde eine – bezogen auf die registrierten Arbeitslosen – höhere Entlastung durch Kurzarbeit erreicht. Die Zahl der Kurzarbeiter geht derzeit sowohl in Ost- als auch in Westdeutschland zurück. Bemerkenswert ist die Zunahme der Dauer von Kurzarbeit. Mittlerweile werden schon 43% der Konjunkturkurzarbeiter seit über einem Jahr durch dieses Instrument gestützt. Die Gefahr einer Verstetigung von „Langzeitkurzarbeit“ nimmt zu, wenn die maximale Bezugsdauer von Kurzarbeitergeld – wie zuletzt im November 2009 – weiter verlängert wird. Dadurch wird die Intention, mit Kurzarbeit einem temporären Arbeitsausfall zu begegnen, konterkariert. Generell ist zu bedenken, dass auch bei Kurzarbeit die Gefahr von Mitnahmeeffekten besteht. Vor dem Hintergrund einer deutlichen Aufhellung der Lage auf dem Arbeitsmarkt sollten nunmehr die großzügigen Regelungen für die Inanspruchnahme von Kurzarbeitergeld schrittweise zurückgenommen werden.
