    VERIFICATION AND VALIDATION OF NUMERICAL ANALYSIS FOR CONSOLIDATION OF CLAY GROUND

    粘性土地盤の圧密変形問題に対して,数値解析の検証と妥当性確認の適用を試みる.数値解析手法は多孔質体理論に基づく土―水連成解析を用いる. はじめにコード検証として解析解との比較を行い,数値解が解析解と一致することを確認し,プログラミングエラーがないと判断された.次の解析検証として,メッシュサイズによる解析解に対する数値解の収束性にOrder of accuracyを適用した.この指標を用いてメッシュサイズの細分化により数値解が解析解に収束することを確認した.また,沈下量よりも間隙水圧の方がメッシュサイズの影響を受けやすいことを示した. 妥当性確認では,三軸試験より得た材料パラメーターの不確かさを正規分布と仮定し,その中から代表点を選択し81通りのパラメトリック解析を,粘性土地盤の圧密変形に関する遠心模型実験を対象として実施した.沈下量に対する材料パラメーターの感度を示すと共に,それらの解析結果と実験結果を比較した.実験値と数値解の分布が重なる部分もあるが,限界状態応力比によって変形モードが変化し沈下量を過大評価する場合も見られた.Verification and validation procedures were applied for numerical analysis of clay ground consolidation. The numerical analysis is a soil-water coupled analysis on finite deformation porous media theory. At the verification stage, the code and calculation verification was conducted based on an analytical solution and the concept of order of accuracy. The accuracy and convergence of numerical method were verified. At the validation stage, first some elasto-plastic material parameters were assumed as normal distributions from laboratory tests. Second the numerical analyses simulated centrifuge tests of clay ground consolidation using the probabilistic material parameters. By comparing the probability distributions of experimental and numerical results, the uncertainty in the clay ground deformation and validity of numerical method were discussed. The distribution of numerical results overestimated that of experimental results

    Update of the Spectro-polarimeter on the Domeless Solar Telescope at Hida Observatory

    A new spectro-polarimeter was installed on the Domeless Solar Telescope at Hida Observatory. Major update s from the previous system are as follows; a super achromatic dual beam polarimeter, communication interface between telescope and data acquisition PC for image scanning , flexible setup for observing a wavelength range of 500-1100 nm, and a high sensitivity infrared camera. The field of view of the system is 120arcsec along the slit of spectrograph, and with a slit width of 0.64 arcsec, the system can achieve a sensitivity of 3x10⁻⁴ in a few second in on disk observation. Details of the instruments, the data reduction flow and initial results obtained by the new system are presented. At the end, future prospect is also discussed

    Chronological urodynamic evaluation of changing bladder and urethral functions after robot-assisted radical prostatectomy

    Objective To examine chronological changes in urethral and bladder functions before, immediately after, and 1 year after robot-assisted radical prostatectomy (RARP), urodynamic studies were prospectively performed. Methods Sixty-three consecutive patients underwent pressure-flow studies, urethral pressure profiles, and abdominal leak point pressure (ALPP) tests 1-2 days before, immediately after, and 1 year after RARP. Results The mean bladder compliance was 28.3 mL/cm H2O before RARP; it worsened to 16.3 mL/cm H2O immediately after RARP and recovered to 27.1 mL/cm H2O at 1 year. The mean detrusor pressure at maximum flow rate was 61.9 cm H2O before RARP; it decreased to 34.3 cm H2O immediately after RARP and remained at 35.6 cm H2O at 1 year. The mean maximum urethral closure pressure was 84.2 cm H2O before RARP; it decreased to 33.4 cm H2O immediately after RARP and recovered to 63.0 cm H2O at 1 year. Intrinsic sphincter deficiency (ISD) evaluated by the ALPP test was observed in 53 patients immediately after RARP, although no patient showed ISD before RARP. ISD remained in 7 patients at 1 year. Both ALPP and maximum urethral closure pressure at 1 year were significant factors for continence in multivariate analysis. Conclusion Urethral sphincter and bladder function worsen immediately after RARP and recover over time. The bladder storage function after RARP returns to almost the same level before RARP, and the voiding function improves compared with the condition before RARP; however, the urethral sphincter function does not return to its preoperative level. Urethral sphincter dysfunction is considered the main factor for urinary incontinence after RARP. © 2015 Elsevier Inc.Embargo Period 12 month

    Application of a Simplified Method of Chloroplast Enrichment to Small Amounts of Tissue for Chloroplast Genome Sequencing

    Premise of the study: High-throughput sequencing of genomic DNA can recover complete chloroplast genome sequences, but the sequence data are usually dominated by sequences from nuclear/mitochondrial genomes. To overcome this deficiency, a simple enrichment method for chloroplast DNA from small amounts of plant tissue was tested for eight plant species including a gymnosperm and various angiosperms. Methods: Chloroplasts were enriched using a high-salt isolation buffer without any step gradient procedures, and enriched chloroplast DNA was sequenced by multiplexed high-throughput sequencing. Results: Using this simple method, significant enrichment of chloroplast DNA-derived reads was attained, allowing deep sequencing of chloroplast genomes. As an example, the chloroplast genome of the conifer Callitris sulcata was assembled, from which polymorphic microsatellite loci were isolated successfully. Discussion: This chloroplast enrichment method from small amounts of plant tissue will be particularly useful for studies that use sequencers with relatively small throughput and that cannot use large amounts of tissue (e.g., for endangered species)
