    T cells and T cell tumors efficiently generate antigen-specific cytotoxic T cell immunity when modified with an NKT ligand

    Various Invariant NKT (iNKT) cell ligands have been shown as potent adjuvants in boosting T cell reactivates to antigens on professional APC. Non-professional APC, such as T cells, also co-expressing MHC class I and CD1d, have been unattractive cell vaccine carriers due to their poor immunogenicity. Here, we report that T cells as well as T cell lymphoma can efficiently generate antigen-specific cytotoxic T lymphocytes (CTL) responses in mice in vivo, when formulated to present iNKT ligand α-galactosylceramide (αGC) on their surface CD1d. Vaccination with αGC-pulsed EG-7 T-cell lymphoma induced tumor-specific CTL response and suppressed the growth of EG-7 in a CD8 T cell-dependent manner. Injection of αGC-loaded CD4 T cells in mice efficiently activated iNKT cells in vivo. While T cells loaded with a class I-restricted peptide induced proliferation but not effector differentiation of antigen-specific CD8 T cells, injection of T cells co-pulsed with αGC strongly induced IFNγ and Granzyme B expression in T cells and complete lysis of target cells in vivo. Presentation of αGC and peptide on the same cells was required for optimal CTL response and vaccinating T cells appeared to directly stimulate both iNKT and cytotoxic CD8 T cells. Of note, the generation of this cytotoxic T cell response was independent of IL-4, IFNγ, IL-12, IL-21 and costimulation. Our data indicate that iNKT cell can license a non-professional APC to directly trigger antigen-specific cytotoxic T cell responses, which provides an alternative cellular vaccine strategy against tumors

    Preparation of tetrabutylammonium salt of a mono-Ru(III)-substituted α-Keggin-type silicotungstate with a 4,4′-bipyridine ligand and its electrochemical behaviour in organic solvents

    The tetrabutylammonium (TBA) salt of a mono-ruthenium(III)-substituted α-Keggin-type silicotungstate with a 4,4′-bipyridine (bipy) ligand, TBA5[α-SiW11O39RuIII(bipy)] (1), which is soluble in various organic solvents, was prepared by a cation exchange reaction of Cs5[α-SiW11O39RuIII(bipy)] with tetrabutylammonium bromide. Compound 1 was characterised using IR, 1H-NMR, elemental analysis, single crystal X-ray analysis, X-ray absorption near-edge structure (XANES) analysis (Ru L3-edge), electron spin resonance (ESR), cyclic voltammetry (CV) and UV-Vis. Single crystal X-ray analysis of 1 revealed that the RuIII unit was incorporated into the α-Keggin-type silicotungstate framework and coordinated by a bipy molecule through a Ru–N bond. CV indicated that the incorporated RuIII-bipy was reversibly oxidised to the RuIV-bipy derivative and reduced to the RuII-bipy derivative in organic solvents. The redox potential of RuIV/III-bipy was found to be affected by organic solvents.     Field measurements of absolute gravity in East Antarctica

    This paper reports the results of field-based absolute gravity measurements aimed at detecting gravity change and crustal displacement caused by glacial isostatic adjustment. The project was initiated within the framework of the 53rd Japanese Antarctic Research Expedition (JARE53). Absolute gravity measurements, together with GPS measurements, were planned at several outcrops along the Prince Olav Coast and Sôya Coast of East Antarctica, including at Syowa Station. Since the icebreaker Shirase (AGB 5003) was unable to moor alongside Syowa Station, operations were somewhat restricted during JARE53. However, despite this setback, we were able to complete measurements at two sites: Syowa Station and Langhovde. The absolute gravity value at the Syowa Station IAGBN (A) site, observed using an FG-5 absolute gravimeter (serial number 210; FG-5 #210), was 982 524 322.7±0.1 μGal, and the gravity change rate at the beginning of 2012 was –0.26 μGal∙a-1. An absolute gravity value of 982 535 584.2±0.7 μGal was obtained using a portable A-10 absolute gravimeter (serial number 017; A-10 #017) at the newly located site AGS01 in Langhovde

    Size Segregation and Convection of Granular Mixtures Almost Completely Packed in the Rotating Thin Box

    Size segregation of granular mixtures which are almost completely packed in a rotating drum is discussed with an effective simulation and a brief analysis. Instead of a 3D drum, we simulate 2D rotating thin box which is almost completely packed with granular mixtures. The phase inversion of radially segregated pattern which was found in a 3D experiment are qualitatively reproduced with this simulation, and a brief analysis is followed. Moreover in our simulation, a global convection appears after radial segregation pattern is formed, and this convection induces axially segregated pattern.Comment: 9 pages, 5 figures, PACS number(s): 45.70.-n, 45.70.M

    Myositis autoantibodies:recent perspectives

    PURPOSE OF REVIEW: To provide an overview of recent discoveries related to myositis-specific autoantibodies (MSAs) and assays used for their measurement.RECENT FINDINGS: New autoantibody specificities have been reported including a MSA directed against eukaryotic initiation factor 3 and a myositis-associated autoantibody directed against heat shock factor 1. The association of anti-TIF1γ with cancer-associated dermatomyositis dependent on age has been confirmed in several large cohorts. Despite MSAs being almost entirely mutually exclusive, several myositis autoantigens are overexpressed in regenerating muscle and do not correlate with the corresponding MSA in any one patient. Further mechanisms may determine the final MSA specificity and are likely to include the need for autoantigen processing and presentation with adaptive T-cell help. The presence of CD4-positive T cells specific for histidyl tRNA synthetase protein in bronchial lavage fluid from antisynthetase patients lends support to this view. Finally, it is widely held that MSA do play an important role in clinical practice among some evidence and concern about commercial assay reliability.SUMMARY: MSAs continue to provide important tools for clinical diagnosis and management as well as insights into disease mechanisms. Further improvement in the standardization and reliability of routine detection of MSAs is a high priority.</p

    s -process branching at W 185 revised

    The neutron capture cross section of the unstable s-process branching nucleus 185W has been derived from experimental data of the inverse 186W(g,n)185W photodisintegration taken with monochromatic photon beams from laser Compton scattering. The result of sigma = 553 +- 60 mb at kT = 30 keV leads to a relatively high effective neutron density in the classical s-process of N_n = 4.7 \times 10^8 cm^-3. A realistic model for the s-process in thermally pulsing AGB stars overestimates the abundance of 186Os significantly because of the relatively small neutron capture cross section of 185W.Comment: 10 pages, 3 figure

    Synthesis, Characterization and Magnetic Susceptibility of the Heavy Fermion Transition Metal Oxide LiV_{2}O_{4}

    The preparative method, characterization and magnetic susceptibility \chi measurements versus temperature T of the heavy fermion transition metal oxide LiV_{2}O_{4} are reported in detail. The intrinsic \chi(T) shows a nearly T-independent behavior below ~ 30 K with a shallow broad maximum at about 16 K, whereas Curie-Weiss-like behavior is observed above 50-100 K. Field-cooled and zero-field-cooled magnetization M measurements in applied magnetic fields H = 10 to 100 G from 1.8 to 50 K showed no evidence for spin-glass ordering. Crystalline electric field theory for an assumed cubic V point group symmetry is found insufficient to describe the observed temperature variation of the effective magnetic moment. The Kondo and Coqblin-Schrieffer models do not describe the magnitude and T dependence of \chi with realistic parameters. In the high T range, fits of \chi(T) by the predictions of high temperature series expansion calculations provide estimates of the V-V antiferromagnetic exchange coupling constant J/k_{B} ~ 20 K, g-factor g ~ 2 and the T-independent susceptibility. Other possible models to describe the \chi(T) are discussed. The paramagnetic impurities in the samples were characterized using isothermal M(H) measurements with 0 < H <= 5.5 Tesla at 2 to 6 K. These impurities are inferred to have spin S_{imp} ~ 3/2 to 4, g_{imp} ~ 2 and molar concentrations of 0.01 to 0.8 %, depending on the sample.Comment: 19 typeset RevTeX pages, 16 eps figures included, uses epsf; to be published in Phys.     Emergence of Antiferromagnetic Correlation in LiTi2-xVxO4 via 7Li NMR

    We report 7Li NMR studies of V-substitution effects on spinel oxide superconductor LiTi2O4 (Tc = 13.4 K). In LiTi2-xVxO4 (x = 0-0.4), the V substitution for the Ti site suppressed the relative volume fraction of superconductivity faster than Tc. From the observation of fairly homogeneous enhancement in a 7Li nuclear spin-lattice relaxation rate, we conclude that the V substitution changes electron correlation effects through electron carrier doping from quarter electron filling 3d0.5 to 3d1.5 and then the antiferromagnetic correlation emerges.Comment: 4 pages, 7 figure
