    Eindämmung des Erregers der Anthraknose der Lupine im ökologischen Landbau durch Anbau weniger anfälliger Lupinensorten der drei landwirtschaftlich wichtigen Lupinenarten

    Das Projekt befasste sich mit einem Sortenscreening von 23 Lupinus albus-, 14 L. luteus- und 21 L. angustifolius-Sorten auf die Reaktion gegenüber einer Infektion mit Colletotrichum lupini var. setosum. Für die Infektion der Samen wurde der Substrattest, eine Methode, die die Primärinfektion des Pathogens simuliert, genutzt. Der Einfluß der Infektion auf verschiedene Merkmale, wie Überlebensrate der Pflanzen, Symptome, Befallsverlauf und auf den Ertrag wurden bewertet. Die drei Lupinenarten zeigten unterschiedliche Krankheitssymptome. Während die Symptome bei L. luteus und L.angustifolius oft untypisch waren, entwickelten sich bei L. albus bekanntere Anthraknosesymptome. Unterschiedliche Verhaltensweisen der drei Lupinenarten waren auch im Befallsverlauf zu finden. Die Versuche lieferten interessante Ergebnisse zum Verhalten der einzelnen Lupinenarten und -sorten gegenüber der Infektion mit dem Anthraknoseerreger. Die höchste Absterberate der Pflanzen aller 3 Lupinenarten war während des Keimlings- und Jungpflanzenstadiums. Der Entwicklungsverlauf der Mykose und die Symptomausprägung waren jedoch bei den Lupinenarten unterschiedlich. Die Überlebensrate der Pflanzen war stark abhängig von der jeweiligen Sorte. Die Ergebnisse sind eine fundierte Materialgrundlage zur Krankheit Anthraknose. Die Sortenunterschiede lassen ein erhebliches Toleranzpotential gegenüber dem Pilz Colletotrichum lupini var. setosum erkennen, so daß entsrechend unterschiedlicher Fragestellungen Sorten anhand der Projektergebnisse auswählt werden können

    Observation of the thermal Casimir force

    Quantum theory predicts the existence of the Casimir force between macroscopic bodies, due to the zero-point energy of electromagnetic field modes around them. This quantum fluctuation-induced force has been experimentally observed for metallic and semiconducting bodies, although the measurements to date have been unable to clearly settle the question of the correct low-frequency form of the dielectric constant dispersion (the Drude model or the plasma model) to be used for calculating the Casimir forces. At finite temperature a thermal Casimir force, due to thermal, rather than quantum, fluctuations of the electromagnetic field, has been theoretically predicted long ago. Here we report the experimental observation of the thermal Casimir force between two gold plates. We measured the attractive force between a flat and a spherical plate for separations between 0.7 μ\mum and 7 μ\mum. An electrostatic force caused by potential patches on the plates' surfaces is included in the analysis. The experimental results are in excellent agreement (reduced χ2\chi^2 of 1.04) with the Casimir force calculated using the Drude model, including the T=300 K thermal force, which dominates over the quantum fluctuation-induced force at separations greater than 3 μ\mum.     Milliarcsecond Localization of the Repeating FRB 20201124A

    Very long baseline interferometric (VLBI) localizations of repeating fast radio bursts (FRBs) have demonstrated a diversity of local environments: from nearby star-forming regions to globular clusters. Here we report the VLBI localization of FRB 20201124A using an ad hoc array of dishes that also participate in the European VLBI Network (EVN). In our campaign, we detected 18 bursts from FRB 20201124A at two separate epochs. By combining the visibilities from both epochs, we were able to localize FRB 20201124A with a 1 sigma uncertainty of 2.7 mas. We use the relatively large burst sample to investigate astrometric accuracy and find that for greater than or similar to 20 baselines (greater than or similar to 7 dishes) we can robustly reach milliarcsecond precision even using single-burst data sets. Subarcsecond precision is still possible for single bursts, even when only similar to 6 baselines (four dishes) are available. In such cases, the limited uv coverage for individual bursts results in very high side-lobe levels. Thus, in addition to the peak position from the dirty map, we also explore smoothing the structure in the dirty map by fitting Gaussian functions to the fringe pattern in order to constrain individual burst positions, which we find to be more reliable. Our VLBI work places FRB 20201124A 710 +/- 30 mas (1 sigma uncertainty) from the optical center of the host galaxy, consistent with originating from within the recently discovered extended radio structure associated with star formation in the host galaxy. Future high-resolution optical observations, e.g., with Hubble Space Telescope, can determine the proximity of FRB 20201124A\u27s position to nearby knots of star formation

    Importance of exposure route for behavioural responses in Lumbriculus variegatus Müller (Oligochaeta: Lumbriculida) in short-term exposures to Pb

    Abstract Goal, Scope and Background Lumbriculus variegatus Müller (Oligochaeta), a common freshwater sediment-dweller, has frequently been used in toxicokinetic studies, although has been less used in ecotoxicity tests. Methods For the first time the Multispecies Freshwater Biomonitor® (MFB) was applied in a short-term whole-sediment toxicity test. The MFB automatically and quantitatively recorded the spontaneous locomotory behaviour of Lumbriculus variegatus in exposures with two compartments, water and sediment. The study questioned, whether the animals altered their locomotion depending on the compartment which was spiked with lead (Pb). Results and Discussion As in the exposures to Pb-contaminated water/clean sediment, the animals exposed to Pb-contaminated sediment/clean water showed higher activities in intermediate Pb-concentrations. This indicates, that spontaneous locomotory activity is affected by Pb-concentrations at sublethal levels regardless of whether the Pb-concentration is found in the water or in the sediment, because these animals use both environmental compartments simultaneously. However, within the same Pb-levels, the animals showed higher locomotory activity in contaminated water compared with contaminated sediment. This indicates a possible tendency to withdraw from (‘avoidance’) contaminated water into the clean sediment compartment, whereas there was no withdrawal from contaminated sediment into clean water. The latter might be explained by the fact that withdrawal from sediment to water might increase the risk of predation and drift in nature, whereas retracting to sediment might provide shelter. Conclusions The study showed that spontaneous locomotory responses of L. variegatus to Pb depend on whether the water or sediment is contaminated. The study also concluded that the Multispecies Freshwater Biomonitor® can be applied effectively in sediment toxicity testing. Recommendations and Perspectives More emphasis should be given to the interactions of water/sediment in sediment ecotoxicity tests to better simulate field conditions and increase ecological realism in risk assessment, especially as quantitative recording methods exisit

    A graphical vector autoregressive modelling approach to the analysis of electronic diary data

    <p>Abstract</p> <p>Background</p> <p>In recent years, electronic diaries are increasingly used in medical research and practice to investigate patients' processes and fluctuations in symptoms over time. To model dynamic dependence structures and feedback mechanisms between symptom-relevant variables, a multivariate time series method has to be applied.</p> <p>Methods</p> <p>We propose to analyse the temporal interrelationships among the variables by a structural modelling approach based on graphical vector autoregressive (VAR) models. We give a comprehensive description of the underlying concepts and explain how the dependence structure can be recovered from electronic diary data by a search over suitable constrained (graphical) VAR models.</p> <p>Results</p> <p>The graphical VAR approach is applied to the electronic diary data of 35 obese patients with and without binge eating disorder (BED). The dynamic relationships for the two subgroups between eating behaviour, depression, anxiety and eating control are visualized in two path diagrams. Results show that the two subgroups of obese patients with and without BED are distinguishable by the temporal patterns which influence their respective eating behaviours.</p> <p>Conclusion</p> <p>The use of the graphical VAR approach for the analysis of electronic diary data leads to a deeper insight into patient's dynamics and dependence structures. An increasing use of this modelling approach could lead to a better understanding of complex psychological and physiological mechanisms in different areas of medical care and research.</p

    A systems analysis of the chemosensitivity of breast cancer cells to the polyamine analogue PG-11047

    <p>Abstract</p> <p>Background</p> <p>Polyamines regulate important cellular functions and polyamine dysregulation frequently occurs in cancer. The objective of this study was to use a systems approach to study the relative effects of PG-11047, a polyamine analogue, across breast cancer cells derived from different patients and to identify genetic markers associated with differential cytotoxicity.</p> <p>Methods</p> <p>A panel of 48 breast cell lines that mirror many transcriptional and genomic features present in primary human breast tumours were used to study the antiproliferative activity of PG-11047. Sensitive cell lines were further examined for cell cycle distribution and apoptotic response. Cell line responses, quantified by the GI<sub>50 </sub>(dose required for 50% relative growth inhibition) were correlated with the omic profiles of the cell lines to identify markers that predict response and cellular functions associated with drug sensitivity.</p> <p>Results</p> <p>The concentrations of PG-11047 needed to inhibit growth of members of the panel of breast cell lines varied over a wide range, with basal-like cell lines being inhibited at lower concentrations than the luminal cell lines. Sensitive cell lines showed a significant decrease in S phase fraction at doses that produced little apoptosis. Correlation of the GI<sub>50 </sub>values with the omic profiles of the cell lines identified genomic, transcriptional and proteomic variables associated with response.</p> <p>Conclusions</p> <p>A 13-gene transcriptional marker set was developed as a predictor of response to PG-11047 that warrants clinical evaluation. Analyses of the pathways, networks and genes associated with response to PG-11047 suggest that response may be influenced by interferon signalling and differential inhibition of aspects of motility and epithelial to mesenchymal transition.</p> <p>See the related commentary by Benes and Settleman: <url>http://www.biomedcentral.com/1741-7015/7/78</url></p

    Direct Identification of the Meloidogyne incognita Secretome Reveals Proteins with Host Cell Reprogramming Potential

    The root knot nematode, Meloidogyne incognita, is an obligate parasite that causes significant damage to a broad range of host plants. Infection is associated with secretion of proteins surrounded by proliferating cells. Many parasites are known to secrete effectors that interfere with plant innate immunity, enabling infection to occur; they can also release pathogen-associated molecular patterns (PAMPs, e.g., flagellin) that trigger basal immunity through the nematode stylet into the plant cell. This leads to suppression of innate immunity and reprogramming of plant cells to form a feeding structure containing multinucleate giant cells. Effectors have generally been discovered using genetics or bioinformatics, but M. incognita is non-sexual and its genome sequence has not yet been reported. To partially overcome these limitations, we have used mass spectrometry to directly identify 486 proteins secreted by M. incognita. These proteins contain at least segmental sequence identity to those found in our 3 reference databases (published nematode proteins; unpublished M. incognita ESTs; published plant proteins). Several secreted proteins are homologous to plant proteins, which they may mimic, and they contain domains that suggest known effector functions (e.g., regulating the plant cell cycle or growth). Others have regulatory domains that could reprogram cells. Using in situ hybridization we observed that most secreted proteins were produced by the subventral glands, but we found that phasmids also secreted proteins. We annotated the functions of the secreted proteins and classified them according to roles they may play in the development of root knot disease. Our results show that parasite secretomes can be partially characterized without cognate genomic DNA sequence. We observed that the M. incognita secretome overlaps the reported secretome of mammalian parasitic nematodes (e.g., Brugia malayi), suggesting a common parasitic behavior and a possible conservation of function between metazoan parasites of plants and animals
