    Developmental Biology Courseware that Integrates Multimedia Technology into the Laboratory and Classroom Experience

    Biological Sciences (61) It is paramount to the learning of science that students participate in science as active learners and researchers. The use of multimedia technology to facilitate this goal offers an inexpensive way of making materials available to a large number of students and encourages students to be independent learners. This project is developing multimedia courseware that makes a wealth of resources available to undergraduate college students who are exploring the field of developmental biology. This field has gained tremendous importance recently because of its relevance to our daily lives through techniques such as cloning, genetic engineering, assisted reproductive techniques, and our realization of environmental impacts on development. It is important that college students gain an understanding of this field and that educational materials be available that make it both exciting and accessible. The multimedia courseware this project is developing consists of an interactive CD-ROM that interfaces with an interactive Web site. This CD-ROM/Web hybrid instructs students on techniques and the biology of organisms used in this field. The courseware is called Vade Mecum, Latin for go with me, which was a term used in the past to designate a manual or handbook, and is used here to refer to the portability of this courseware, aiding its usefulness as the student uses it to explore developmental biology. One of the strengths of Vade Mecum is that it is fully integrated with a laboratory manual and coordinated with a major textbook in the field. This creates a unified package that facilitates both teaching and learning. Vade Mecum uses QuickTime movies, QuickTime Virtual Reality modules, an extensive gallery of labeled photographs, interactive learning sequences, puzzles, questions, and Web links to communicate to students how to create their own investigative tools, how to work with various model organisms, and how to understand the complex life cycles and development of these organisms. Web modules allow and encourage students to post their own work and discussions in the Virtual Poster Session and Virtual Round Table sections. By showing students the methods for low-cost experiments, how to construct their own tools out of common, inexpensive materials, how to adapt an inexpensive microscope to have it perform as an expensive instrument, and by keeping the cost of the Vade Mecum CD to a minimum and making the Vade Mecum Web Site available to all, this course material will be useful to learning at all institutions regardless of their financial status

    Hypoxia-inducible factor (HIF) appears to function as a global master regulator of cellular and systemic responses to hypoxia. HIF-pathway manipulation is of therapeutic interest, however global, systemic upregulation of HIF may have as yet unknown effects on multiple processes. We utilized a mouse model of Chuvash polycythemia (CP), a rare genetic disorder which modestly increases expression of HIF target genes in normoxia, to understand what these effects might be within the heart. An integrated in and ex vivo approach was employed. In comparison to wild-type controls, CP mice had evidence (using in vivo MRI) of pulmonary hypertension, right ventricular hypertrophy, and increased left ventricular ejection fraction. Glycolytic flux (measured using 3H glucose) in the isolated, contracting, perfused CP heart was 1.8-fold higher. Net lactate efflux was 1.5-fold higher. Furthermore, in vivo 13C magnetic resonance spectroscopy (MRS) of hyperpolarized 13C1 pyruvate revealed a 2-fold increase in real-time flux through lactate dehydrogenase in the CP hearts, and a 1.6-fold increase through pyruvate dehydrogenase. 31P MRS of perfused CP hearts under increased workload (isoproterenol infusion) demonstrated increased depletion of phosphocreatine relative to ATP. Intriguingly, no changes in cardiac gene expression were detected. In summary, a modest systemic dysregulation of the HIF pathway resulted in clear alterations in cardiac metabolism and energetics. However, in contrast to studies generating high HIF levels within the heart, the CP mice showed neither the predicted changes in gene expression nor any degree of LV impairment. We conclude that the effects of manipulating HIF on the heart are dose-dependent. New and noteworthy This is the first integrative metabolic and functional study of the effects of modest HIF manipulation within the heart. Of particular note, the combination (and correlation) of perfused heart metabolic flux measurements with the new technique of real-time in vivo MR spectroscopy using hyperpolarized pyruvate is a novel development

    How risky are risk factors? An analysis of prenatal risk factors in patients participating in the congenital upper limb differences registry

    PURPOSE: Risk factors for congenital upper limb differences (CoULDs) are often studied at the general population level. The CoULD registry provides a unique opportunity to study prenatal risk factors within a large patient sample. METHODS: All patients enrolled between June 2014 and March 2020 in the prospective CoULD registry, a national multicenter database of patients diagnosed with a CoULD, were included in the analysis. We analyzed self-reported, prenatal risk factors, including maternal smoking, alcohol use, recreational drug use, prescription drug use, gestational diabetes mellitus (GDM), and gestational hypertension. The outcome measures included comorbid medical conditions, proximal involvement of limb difference, bilateral involvement, and additional orthopedic conditions. Multivariable logistic regression was used to analyze the effect of the risk factors, controlling for sex and the presence of a named syndrome. RESULTS: In total, 2,410 patients were analyzed, of whom 72% (1,734) did not have a self-reported risk factor. Among the 29% (676) who did have at least 1 risk factor, prenatal maternal prescription drug use was the most frequent (376/2,410; 16%). Maternal prescription drug use was associated with increased odds of patient medical comorbidities (odds ratio [OR] = 1.43, CONCLUSIONS: Most caregivers (72%) did not report a risk factor during enrollment. However, reporting a risk factor was associated with patient medical and orthopedic comorbidities. Of note, GDM alone significantly increased the odds of both these outcome measures along with proximal limb differences. These findings highlight the ill-defined etiology of CoULDs but suggest that prenatal risk factors, especially GDM, are associated with a higher degree of morbidity.     Mindfulness-based school interventions: A systematic review of outcome evidence quality by study design.

    Objective: The purpose of this systematic review was to assess the current literature on mindfulness-based school interventions (MBSIs) by evaluating evidence across specific outcomes for youth. Methods: We evaluated 77 studies with a total sample of 12,358 students across five continents, assessing the quality of each study through a robust coding system for evidence-based guidelines. Coders rated each study numerically per study design as 1++ (RCT with a very low risk of bias) to 4 (expert opinion) and across studies for the corresponding evidence letter grade, from highest quality (\u27A Grade\u27) to lowest quality (\u27D Grade\u27) evidence. Results: The highest quality evidence (\u27A Grade\u27) across outcomes indicated that MBSIs increased prosocial behavior, resilience, executive function, attention and mindfulness, and decreased anxiety, attention problems/ADHD behaviors and conduct behaviors. The highest quality evidence for well-being was split, with some studies showing increased well-being and some showing no improvements. The highest quality evidence suggests MBSIs have a null effect on depression symptoms. Conclusion: This review demonstrates the promise of incorporating mindfulness interventions in school settings for improving certain youth outcomes. We urge researchers interested in MBSIs to study their effectiveness using more rigorous designs (e.g., RCTs with active control groups, multi-method outcome assessment, and follow-up evaluation), to minimize bias and promote higher quality-not just increased quantity-evidence that can be relied upon to guide school-based practice

    CHARIS Science: Performance Simulations for the Subaru Telescope's Third-Generation of Exoplanet Imaging Instrumentation

    We describe the expected scientific capabilities of CHARIS, a high-contrast integral-field spectrograph (IFS) currently under construction for the Subaru telescope. CHARIS is part of a new generation of instruments, enabled by extreme adaptive optics (AO) systems (including SCExAO at Subaru), that promise greatly improved contrasts at small angular separation thanks to their ability to use spectral information to distinguish planets from quasistatic speckles in the stellar point-spread function (PSF). CHARIS is similar in concept to GPI and SPHERE, on Gemini South and the Very Large Telescope, respectively, but will be unique in its ability to simultaneously cover the entire near-infrared JJ, HH, and KK bands with a low-resolution mode. This extraordinarily broad wavelength coverage will enable spectral differential imaging down to angular separations of a few λ/D\lambda/D, corresponding to ∼\sim0.\!\!''1. SCExAO will also offer contrast approaching 10−510^{-5} at similar separations, ∼\sim0.\!\!''1--0.\!\!''2. The discovery yield of a CHARIS survey will depend on the exoplanet distribution function at around 10 AU. If the distribution of planets discovered by radial velocity surveys extends unchanged to ∼\sim20 AU, observations of ∼\sim200 mostly young, nearby stars targeted by existing high-contrast instruments might find ∼\sim1--3 planets. Carefully optimizing the target sample could improve this yield by a factor of a few, while an upturn in frequency at a few AU could also increase the number of detections. CHARIS, with a higher spectral resolution mode of R∼75R \sim 75, will also be among the best instruments to characterize planets and brown dwarfs like HR 8799 cde and κ\kappa And b.Comment: 13 pages, 7 figures, proceedings from SPIE Montrea

    On Black Hole Detection with the OWL/Airwatch Telescope

    In scenarios with large extra dimensions and TeV scale gravity ultrahigh energy neutrinos produce black holes in their interactions with the nucleons. We show that ICECUBE and OWL may observe large number of black hole events and provide valuable information about the fundamental Planck scale and the number of extra dimensions. OWL is especially well suited to observe black hole events produced by neutrinos from the interactions of cosmic rays with the 3 K background radiation.
