HorTILLUS - a rich and renewable source of induced mutations for forward/reverse genetics and pre-breeding programs in barley (Hordeum vulgare L.)

TILLING (Targeting Induced Local Lesions IN Genomes) is a strategy used for functional analysis of genes that combines the classical mutagenesis and a rapid, high-throughput identification of mutations within a gene of interest. TILLING has been initially developed as a discovery platform for functional genomics, but soon it has become a valuable tool in development of desired alleles for crop breeding, alternative to transgenic approach. Here we present the HorTILLUS (Hordeum—TILLING—University of Silesia) population created for spring barley cultivar “Sebastian” after double-treatment of seeds with two chemical mutagens: sodium azide (NaN3) and N-methyl-N-nitrosourea (MNU). The population comprises more than 9,600 M2 plants from which DNA was isolated, seeds harvested, vacuum-packed, and deposited in seed bank. M3 progeny of 3,481 M2 individuals was grown in the field and phenotyped. The screening for mutations was performed for 32 genes related to different aspects of plant growth and development. For each gene fragment, 3,072–6,912 M2 plants were used for mutation identification using LI-COR sequencer. In total, 382 mutations were found in 182.2Mb screened. The average mutation density in the HorTILLUS, estimated as 1 mutation per 477 kb, is among the highest mutation densities reported for barley. The majority of mutations were G/C to A/T transitions, however about 8% transversions were also detected. Sixty-one percent of mutations found in coding regions were missense, 37.5% silent and 1.1% nonsense. In each gene, the missense mutations with a potential effect on protein function were identified. The HorTILLUS platformis the largest of the TILLING populations reported for barley and best characterized. The population proved to be a useful tool, both in functional genomic studies and in forward selection of barley mutants with required phenotypic changes. We are constantly renewing the HorTILLUS population, which makes it a permanent source of new mutations.We offer the usage of this valuable resource to the interested barley researchers on cooperative basis

Removal of the Crucifixes in 1958 in the Sandomierz poviat

The paper presents the course of decrucification action in Sandomierz County located in former Kielce voivodeship. The action was planned by communist authorities and legally based on the Circular no. 26 of the Ministry of Education dated 4 August 1958. In order to present the course of the action, the author needed a query of source materials: KP PZPR and Department of Education of PPRN in Sandomierz, security force act and local Bishop Curia. The author shows all stages of decrucification action carried out on a country level. The first one were preparations, consisting of meeting KP PZR executive, trainings for school directors and state institutions. Another stage was to remove crosses from common schools and state institutions, which did not take place without argument. At the end of the decrucification action, summaries and evaluation concluded that removal of crosses had gone without major problems. In order to present a full picture of the described action, the author presented an outline of the introduction process of Circular 26 by Ministry of Education on 4 August 1958 and discussed the school network of Sandomierz County. The paper is another attempt to show the ways communist authorities fought the Catholic church after the II World War

Surveillance of the Sandomierz diocese clergy in the light of a document of the Kielce Voivodeship Police Headquarters of 6 October 1961.

This source edition presents a document created by the officers of the Security Service at the Voivodeship Police Headquarters in Kielce in October 1961, containing information on the clergy of the Sandomierz diocese. It was kept as part of a case file entitled “Wisła” (surveillance of the diocesan curia and the seminary in Sandomierz). The document is a typical example of “counter-intelligence features of a case.” In addition, it contains a directional operational plan of undertakings”. An analysis of the source allows us to determine the scale of surveillance to which the priests of the diocese were subjected. The present edition of the document contains a number of footnotes, which explain the more difficult formulations used by security officers, briefly approximate the figures of the priests named in it and elaborate on the state/Church relations in the Polish People’s Republic

Zawiera on informacje na temat duchowieństwa diecezji sandomierskiej. Został on wytworzony w ramach teczki zagadnieniowej o kryptonimie „Wisła” (rozpracowanie Kurii diecezjalnej w Sandomierzu i seminarium duchownego w tymże mieście). Dokument ten jest typową dla tych teczek „kontrwywiadowczą charakterystyką zagadnienia”. Dodatkowo zawiera on „kierunkowy plan operacyjnych przedsięwzięć”.  Analiza tego źródła pozwala na określenie skali inwigilacji, której poddanie zostali kapłani wspomnianej diecezji.
