    A rapid culture independent methodology to quantitatively detect and identify common human bacterial pathogens associated with contaminated high purity water

    Background: Water and High Purity Water (HPW) distribution systems can be contaminated with human pathogenic microorganisms. This biocontamination may pose a risk to human health as HPW is commonly used in the industrial, pharmaceutical and clinical sectors. Currently, routine microbiological testing of HPW is performed using slow and labour intensive traditional microbiological based techniques. There is a need to develop a rapid culture independent methodology to quantitatively detect and identify biocontamination associated with HPW. Results: A novel internally controlled 5-plex real-time PCR Nucleic Acid Diagnostics assay (NAD), was designed and optimised in accordance with Minimum Information for Publication of Quantitative Real-Time PCR Experiments guidelines, to rapidly detect, identify and quantify the human pathogenic bacteriaStenotrophomonas maltophilia, Burkholderia species, Pseudomonas aeruginosa and Serratia marcescenswhich are commonly associated with the biocontamination of water and water distribution systems. The specificity of the 5-plex assay was tested against genomic DNA isolated from a panel of 95 microorganisms with no cross reactivity observed. The analytical sensitivities of the S. maltophilia, B. cepacia, P. aeruginosa and the S. marcescens assays are 8.5, 5.7, 3.2 and 7.4 genome equivalents respectively. Subsequently, an analysis of HPW supplied by a Millipore Elix 35 water purification unit performed using standard microbiological methods revealed high levels of naturally occurring microbiological contamination. Five litre water samples from this HPW delivery system were also filtered and genomic DNA was purified directly from these filters. These DNA samples were then tested using the developed multiplex real-time PCR NAD assay and despite the high background microbiological contamination observed, both S. maltophilia andBurkholderia species were quantitatively detected and identified. At both sampling points the levels of both S. maltophilia and Burkholderia species present was above the threshold of 10 cfu/100 ml recommended by both EU and US guidelines. Conclusions: The novel culture independent methodology described in this study allows for rapid (<5 h), quantitative detection and identification of these four human pathogens from biocontaminated water and HPW distribution systems. We propose that the described NAD assay and associated methodology could be applied to routine testing of water and HPW distribution systems to assure microbiological safety and high water quality standards

    Estimation of the Health Impact and Cost-Effectiveness of Influenza Vaccination with Enhanced Effectiveness in Canada

    INTRODUCTION: The propensity for influenza viruses to mutate and recombine makes them both a familiar threat and a prototype emerging infectious disease. Emerging evidence suggests that the use of MF59-adjuvanted vaccines in older adults and young children enhances protection against influenza infection and reduces adverse influenza-attributable outcomes compared to unadjuvanted vaccines. The health and economic impact of such vaccines in the Canadian population are uncertain. METHODS: We constructed an age-structured compartmental model simulating the transmission of influenza in the Canadian population over a ten-year period. We compared projected health outcomes (quality-adjusted life years (QALY) lost), costs, and incremental cost-effectiveness ratios (ICERs) for three strategies: (i) current use of unadjuvanted trivalent influenza vaccine; (ii) use of MF59-adjuvanted influenza vaccine adults ≥65 in the Canadian population, and (iii) adjuvanted vaccine used in both older adults and children aged < 6. RESULTS: In the base case analysis, use of adjuvanted vaccine in older adults was highly cost-effective (ICER = 2111/QALYgained),butsuchaprogramwas"dominated"byaprogramthatextendedtheuseofadjuvantedvaccinetoincludeyoungchildren(ICER = 2111/QALY gained), but such a program was "dominated" by a program that extended the use of adjuvanted vaccine to include young children (ICER = 1612/QALY). Results were similar whether or not a universal influenza immunization program was used in other age groups; projections were robust in the face of wide-ranging sensitivity analyses. INTERPRETATION: Based on the best available data, it is projected that replacement of traditional trivalent influenza vaccines with MF59-adjuvanted vaccines would confer substantial benefits to vaccinated and unvaccinated individuals, and would be economically attractive relative to other widely-used preventive interventions

    On Exceptional Vertex Operator (Super) Algebras

    We consider exceptional vertex operator algebras and vertex operator superalgebras with the property that particular Casimir vectors constructed from the primary vectors of lowest conformal weight are Virasoro descendents of the vacuum. We show that the genus one partition function and characters for simple ordinary modules must satisfy modular linear differential equations. We show the rationality of the central charge and module lowest weights, modularity of solutions, the dimension of each graded space is a rational function of the central charge and that the lowest weight primaries generate the algebra. We also discuss conditions on the reducibility of the lowest weight primary vectors as a module for the automorphism group. Finally we analyse solutions for exceptional vertex operator algebras with primary vectors of lowest weight up to 9 and for vertex operator superalgebras with primary vectors of lowest weight up to 17/2. Most solutions can be identified with simple ordinary modules for known algebras but there are also four conjectured algebras generated by weight two primaries and three conjectured extremal vertex operator algebras generated by primaries of weight 3, 4 and 6 respectively.Comment: 37 page

    We note that Witten's proposed duality between extremal c=24k CFTs and three-dimensional anti-de Sitter gravity may possibly be extended to central charges that are multiples of 8, for which extremal self-dual CFTs are known to exist up to c=40. All CFTs of this type with central charge 24 or higher, provided that they exist, have the required mass gap and may serve as candidate duals to three-dimensional gravity at the corresponding values of the cosmological constant. Here, we compute the genus one partition function of these theories up to c=88, we give exact and approximate formulas for the degeneracies of states, and we determine the genus two partition functions of the theories up to c=40.Comment: 17 pages, harvmac; v2: references added, version accepted in JHE

    On the Relationship between the Uniqueness of the Moonshine Module and Monstrous Moonshine

    We consider the relationship between the conjectured uniqueness of the Moonshine Module, Vâ™®{\cal V}^\natural, and Monstrous Moonshine, the genus zero property of the modular invariance group for each Monster group Thompson series. We first discuss a family of possible ZnZ_n meromorphic orbifold constructions of Vâ™®{\cal V}^\natural based on automorphisms of the Leech lattice compactified bosonic string. We reproduce the Thompson series for all 51 non-Fricke classes of the Monster group MM together with a new relationship between the centralisers of these classes and 51 corresponding Conway group centralisers (generalising a well-known relationship for 5 such classes). Assuming that Vâ™®{\cal V}^\natural is unique, we then consider meromorphic orbifoldings of Vâ™®{\cal V}^\natural and show that Monstrous Moonshine holds if and only if the only meromorphic orbifoldings of Vâ™®{\cal V}^\natural give Vâ™®{\cal V}^\natural itself or the Leech theory. This constraint on the meromorphic orbifoldings of Vâ™®{\cal V}^\natural therefore relates Monstrous Moonshine to the uniqueness of Vâ™®{\cal V}^\natural in a new way.Comment: 53 pages, PlainTex, DIAS-STP-93-0

    Somatosensory Training Improves Proprioception and Untrained Motor Function in Parkinson's Disease

    Background: Proprioceptive impairment is a common feature of Parkinson's disease (PD). Proprioceptive function is only partially restored with anti-parkinsonian medication or deep brain stimulation. Behavioral exercises focusing on somatosensation have been promoted to overcome this therapeutic gap. However, conclusive evidence on the effectiveness of such somatosensory-focused behavioral training for improving somatosensory function is lacking. Moreover, it is unclear, if such training has any effect on motor performance in PD.Objective: To investigate, whether proprioception improves with a somatosensory focused, robot-aided training in people with PD (PWPs), and whether enhanced proprioception translates to improved motor performance.Method: Thirteen PWPs of mild-moderate clinical severity were assessed and trained ON medication using a robotic wrist exoskeleton. Thirteen healthy elderly participants served as controls. Training involved making increasingly accurate, continuous, precise small amplitude wrist flexion/extension movements. Wrist position sense acuity, as a marker of proprioception function, and spatial error during wrist pointing, as a marker of untrained motor performance, were recorded twice before and once after training. Functional hand writing kinematics exhibited during training were evaluated in the PD group for determining training-induced changes.Results: Training improved position sense acuity in all PWPs (mean change: 28%; p &lt; 0.001) and healthy controls (mean change: 23%; p &lt; 0.01). Second, 10/13 PD participants and 10/13 healthy control participants had reduced spatial movement error in the untrained wrist pointing task after training. Third, spatial error for the functional handwriting tasks (line tracing and tracking) did not improve with training in the PD group.Conclusion: Proprioceptive function in mild to moderate PD is trainable and improves with a somatosensory-focused motor training. Learning showed a local transfer within the trained joint degree-of-freedom as improved spatial accuracy in an unpracticed motor task. No learning gains were observed for the untrained functional handwriting task, indicating that training may be specific to the trained joint degree-of-freedom

    Sunshine, Sea, and Season of Birth: MS Incidence in Wales

    Maternal sun exposure in gestation and throughout the lifetime is necessary for vitamin D synthesis, and living near the sea is a population level index of seafood consumption. The aim of this study was to estimate the incidence rate of multiple sclerosis (MS) in Wales and examine its association with sun exposure, coastal living, and latitude. The study used a database of MS hospital visits and admissions in Wales between 2002 and 2013. For the 1,909 lower layer super output areas (LSOAs) in Wales, coastal status, population, longitude/latitude, and average sunshine hours per day were obtained. Age-specific and age-standardised MS incidence were calculated and modelled using Poisson regression. The distribution of births by month was compared between MS cases and the combined England and Wales population. There were 3,557 new MS cases between 2002 and 2013, with an average annual incidence of 8.14 (95% CI: 7.69-8.59) among males and 12.97 (95% CI: 12.44-13.50) among females per 100,000 population. The female-to-male ratio was 1.86:1. For both sexes combined, the average annual incidence rate was 9.10 (95% CI: 8.80-9.40). All figures are age-standardized to the 1976 European standard population. Compared to the combined England and Wales population, more people with MS were born in April, observed-to-expected ratio: 1.21 (95% CI: 1.08-1.36). MS incidence varied directly with latitude and inversely with sunshine hours. Proximity to the coast was associated with lower MS incidence only in easterly areas. This study shows that MS incidence rate in Wales is comparable to the rate in Scotland and is associated with environmental factors that probably represent levels of vitamin D

    Quantification of amyloid fibril polymorphism by nano-morphometry reveals the individuality of filament assembly

    Amyloid fibrils are highly polymorphic structures formed by many different proteins. They provide biological function but also abnormally accumulate in numerous human diseases. The physicochemical principles of amyloid polymorphism are not understood due to lack of structural insights at the single-fibril level. To identify and classify different fibril polymorphs and to quantify the level of heterogeneity is essential to decipher the precise links between amyloid structures and their functional and disease associated properties such as toxicity, strains, propagation and spreading. Employing gentle, force-distance curve-based AFM, we produce detailed images, from which the 3D reconstruction of individual filaments in heterogeneous amyloid samples is achieved. Distinctive fibril polymorphs are then classified by hierarchical clustering, and sample heterogeneity is objectively quantified. These data demonstrate the polymorphic nature of fibril populations, provide important information regarding the energy landscape of amyloid self-assembly, and offer quantitative insights into the structural basis of polymorphism in amyloid populations

    Twenty four female gerbils were alloted into 4 groups and each group consisted of 6 animals. Group A was fed with standard diet used as control. Groups B, C and D were fed with diets respectively containing 2,5 %, 5 % and 10 % concentrations of dried ground cultures of Penicillium viridicatum. Three gerbils from each group were killed at the end of 5 and 12 week feeding period respectively. No clinical sign was observed in test groups, other than decrease of body weight. Gross lesions were observed in animals which were killed.at the end of 12 week feeding period. Lesions included hydropic degeneration, hemorrhages, and necrosis of perirenal fatty tissue. The renal parenchyma showed the feature of toxicosis. Microscopic lesions of degeneration, hemorrhages, necrosis and inflammatory cells infiltration of perirenal tissue. The kidney showed focal dilatation and collapse of tubulus
