Kulakov Ministry of Health care and Social Development of Russia, Moscow, Russia; e-mail: [email protected] Abstract More than 50% of pregnant women after the programs of assisted reproductive technologies (ART) face the problem of recurrent miscarriage (RMC), especially in the first trimester. Significant role in the development of RMC has infectious factor and chronic inflammation in the endometrium. The aim: to reveal the peculiarities of immune response mRNA genes of the inflammatory component expression in the period of the tentative implantation window (TIW) in women with RMC in ART programs. Material and methods. The main group consisted of 240 patients with RMC in ART programs; the control group included 100 conditionally healthy fertile women. On the ground of PCR reverse transcription, the mRNA of the IL-1β, IL-2, IL-10, Foxp3, TLR9, IL-2Rα cytokine genes was examined in endometrial samples obtained with the help of biopsy on the TIW day. Results. Analysis of the transcriptional profile of the immune response genes in the endometrium on TIW day revealed that the relative level of mRNA expression of the IL-1β, IL-2, Foxp3, TLR9, IL-2Rα genes did not differ significantly in the main and control groups. Statistically significant decrease in mRNA expression of IL-10 gene was observed in women with RPL. Conclusions. A feature of mRNA expression of the inflammatory component of the immune response in TIW period in women with RMC in ART programs is a decrease in the expression level of the IL-10 gene mRNA, which may be one of the reasons for the unfavorable outcomes of the onset pregnancy.     Anomalous Behavior Of The Complex Conductivity Of Y_{1-x}Pr_xBa_2Cu_3O_7 Observed With THz Spectroscopy

    We have measured the electrodynamic properties of Y_{1-x}Pr_xBa_2Cu_3O_7 single crystal thin films as a function of temperature using coherent THz-time-domain spectroscopy. We obtain directly the complex conductivity σ=σ1+iσ2\sigma=\sigma_1+i\sigma_2, the London penetration depth λL\lambda_L, the plasma frequency ωp\omega_p, and the quasiparticle scattering rate 1/τ1/\tau. We find that 1/τ1/\tau drops exponentially rapidly with TT below the critical temperature in {\em all} the superconducting samples, implying that this behavior is a {\em signature} of high-TcT_c superconductivity. The plasma frequency decreases with increasing Pr content, providing evidence that Pr depletes carriers, leaving the CuO planes {\em underdoped}. Both the conductivity in the THz region and the dc resistivity yield evidence for the opening of a spin gap {\em above} TcT_c.Comment: 9 pages, REVTEX 3.

    Investigation of lymphocyte proliferation rate and IL2 and IL2RA mRNA expression in concanavalin А-activated lymphocytes by real time PCR

    One of the traditional methods of lymphocytes functional activity research is blast transformation reaction (RBTL). The rate of PBMC proliferation in response to different stimuli can be evaluated in vitro by introduction of radioactive 3H-thymidine in cell DNA. The aim of our work was to search for alternative method of RBTL evaluation by PCR since clinical application of RBTL is limited due to radioactive labelling. The genes expressing an increased proteomic activity of their products in mitosis such as topoisomerase II alpha (gene tpa) and CDC2 (gene cdc2) were chosen as proliferation markers. In our work test systems for mRNA content estimation of tpa and cdc2 genes in concanavalin A activated lymphocytes were developed by means of real time PCR. Kinetics of mRNA level of il2 and il2ra genes in these cells was investigated. As a result the mRNA accumulation of the given genes was monitored from the stimulation starting up to 180 hours of post stimulation time. Common pattern of their mRNA expression in cells of healthy donors was identified, significant differences in kinetics of mRNA levels in comparison with lymphocytes stimulated by phytohemagglutinin were determined.The obtained results show that tpa and cdc2 mRNA are reliable proliferation markers in RBTL. Thus convenient test systems for lymphocytes functional activity evaluation in vitro free of radioactive labelling can be developed. The mRNA level of interleukin 2 gene can be used as marker of lymphocyte activation by concanavalin A in the first hours of stimulation.Реакция бластной транформации лимфоцитов (РБТЛ) представляет собой классический метод исследования функциональной активности лимфоцитов. В зтой реакции in vitro оценивают степень пролиферации мононуклеарных клеток периферической крови (РВМС) в ответ на различные стимулы с помощью включения меченого 3Н-тимидина в ДНК в сравнении с нестимулированными клетками, однако использование радиоактивной метки ограничивает применение этой методики на практике. Целью работы являлось исследование степени пролиферации мононуклеарных клеток периферической крови с помощью разработанной нами тест-системы на основе ПЦР «в реальном времени» в сравнении с классической методикой с использованием 3Н-тимидина. Маркерами реакции служили гены, активность продуктов которых возрастает при митозе: топоизомеразы II альфа (ген tpa) и белка CDC2 (ген cdc2). В ходе работы с использованием метода ПЦР в режиме «реального времени» была исследована кинетика накопления соответствующих мРНК в процессе стимуляции мононуклеарных клеток периферической крови неспецифическим митогеном конканавалином А (СопА). Также был проведен мониторинг изменения уровня мРНК генов интерлейкина 2 и рецептора интерлейкина 2a(il2 и il2ra, соответственно). Выявлены значимые различия в кинетике накопления мРНК исследованных генов по сравнению со стимуляцией фитогемагглютинином.Полученные результаты показывают, что мРНК генов tpa и cdc2 может служить надежным маркером пролиферации клеток в РБТЛ, что позволяет создать удобные тест-системы для оценки функциональной активности лимфоцитов in vitro без использования радиоактивной метки, а определение уровня мРНК интерлейкина 2 можно использовать как маркер активации клеток иммунной системы при стимуляции СопА в первые же часы после стимуляции

    Optical Properties of Layered Superconductors near the Josephson Plasma Resonance

    We study the optical properties of crystals with spatial dispersion and show that the usual Fresnel approach becomes invalid near frequencies where the group velocity of the wave packets inside the crystal vanishes. Near these special frequencies the reflectivity depends on the atomic structure of the crystal provided that disorder and dissipation are very low. This is demonstrated explicitly by a detailed study of layered superconductors with identical or two different alternating junctions in the frequency range near the Josephson plasma resonance. Accounting for both inductive and charge coupling of the intrinsic junctions, we show that multiple modes are excited inside the crystal by the incident light, determine their relative amplitude by the microscopic calculation of the additional boundary conditions and finally obtain the reflectivity. Spatial dispersion also provides a novel method to stop light pulses, which has possible applications for quantum information processing and the artificial creation of event horizons in a solid.Comment: 25 pages, 20 figures, submitted to Phys.
