Reciprocal Sign Epistasis between Frequently Experimentally Evolved Adaptive Mutations Causes a Rugged Fitness Landscape
The fitness landscape captures the relationship between genotype and evolutionary fitness and is a pervasive metaphor used to describe the possible evolutionary trajectories of adaptation. However, little is known about the actual shape of fitness landscapes, including whether valleys of low fitness create local fitness optima, acting as barriers to adaptive change. Here we provide evidence of a rugged molecular fitness landscape arising during an evolution experiment in an asexual population of Saccharomyces cerevisiae. We identify the mutations that arose during the evolution using whole-genome sequencing and use competitive fitness assays to describe the mutations individually responsible for adaptation. In addition, we find that a fitness valley between two adaptive mutations in the genes MTH1 and HXT6/HXT7 is caused by reciprocal sign epistasis, where the fitness cost of the double mutant prohibits the two mutations from being selected in the same genetic background. The constraint enforced by reciprocal sign epistasis causes the mutations to remain mutually exclusive during the experiment, even though adaptive mutations in these two genes occur several times in independent lineages during the experiment. Our results show that epistasis plays a key role during adaptation and that inter-genic interactions can act as barriers between adaptive solutions. These results also provide a new interpretation on the classic Dobzhansky-Muller model of reproductive isolation and display some surprising parallels with mutations in genes often associated with tumors
Tau-dependent suppression of adult neurogenesis in the stressed hippocampus
uncorrected proofStress, a well-known sculptor of brain plasticity, is shown to suppress hippocampal neurogenesis in the adult brain; yet, the underlying cellular mechanisms are poorly investigated. Previous studies have shown that chronic stress triggers hyperphosphorylation and accumulation of the cytoskeletal protein Tau, a process that may impair the cytoskeleton-regulating role (s) of this protein with impact on neuronal function. Here, we analyzed the role of Tau on stress-driven suppression of neurogenesis in the adult dentate gyrus (DG) using animals lacking Tau (Tau-knockout; Tau-KO) and wild-type (WT) littermates. Unlike WTs, Tau-KO animals exposed to chronic stress did not exhibit reduction in DG proliferating cells, neuroblasts and newborn neurons; however, newborn astrocytes were similarly decreased in both Tau-KO and WT mice. In addition, chronic stress reduced phosphoinositide 3-kinase (PI3K)/mammalian target of rapamycin (mTOR)/glycogen synthase kinase-3 beta (GSK3 beta)/beta-catenin signaling, known to regulate cell survival and proliferation, in the DG of WT, but not Tau-KO, animals. Temporal variation in the biomass and nutrient status of Azolla filiculoides Lam. (Salviniaceae) in a small shallow dystrophic lake
AIM: This study determined the temporal variation of the biomass and the concentrations of carbon, nitrogen and phosphorus in Azolla filiculoides Lam. in a small (0.5 ha) shallow dystrophic lake located in the city of Rio Grande (Rio Grande do Sul State, Brazil). METHOD: Sampling was conducted monthly between November 2000 and October 2001. The macrophytes were collected randomly in three replicates with a circular collector 0.3 m in diameter and subsequently washed with tap water and oven-dried at 60 ºC for determination of the dry weight and the nutrient status (i.e., carbon, nitrogen and phosphorus). Primary productivity was estimated by the variation in biomass among successive samples. RESULTS: A. filiculoides was present in the lake throughout the year and occupied between 50 and 80% of the surface area. The biomass values ranged from 34.2 g DW.m-2, recorded in May (autumn), to 170.9 g DW.m-2 in January (summer). The highest rate of primary productivity was 3.3 g DW.m-2.d-1, observed in June. The concentrations of carbon, nitrogen and phosphorus in the plant ranged between 403 and 551 g.kg-1, 13.4 and 25.7 g.kg-1 and 0.5 and 1.9 g.kg-1, respectively. The water N:P ratio ranged between 19:1 and 368:1. CONCLUSION: The coverage of the surface of the lake by A. filiculoides throughout the study period and the nutritional status of the plant demonstrate the importance of the cycling of nutrients by macrophytes in this aquatic environment. The higher N:P ratio in the water column, compared with other neighboring environments without macrophytes, shows that the enrichment of the lake may result from the biological N-fixation activity produced by A. filiculoides
Aquatic macrophytes as indicators of water quality in subtropical shallow lakes, Southern Brazil
AIMS: We evaluated the potential of aquatic macrophyte communities as bioindicators in six small shallow lakes. METHODS: The sampling was conducted monthly for one year, during which all macrophytes were surveyed, and the water temperature, dissolved oxygen, pH, conductivity, total alkalinity, chlorophyll-a, suspended matter, total nitrogen (Nt) and total phosphorus (Pt) were measured. RESULTS: In total, forty-three species were recorded, and there were significant differences in the species richness and limnological conditions among the lakes studied. A canonical correspondence analysis showed that the concentration of nutrients (Nt and Pt), chlorophyll-a, suspended matter, dissolved oxygen and pH were the most important predictors of the distribution of macrophytes. Some emergents were related to the high concentration of nutrients, chlorophyll-a, and suspended matter. Moreover, the most submersed species were associated with environments with low nutrient concentrations and the lowest values chlorophyll-a and suspended matter. In addition, some species submerged and floating were related to low values pH, alkalinity and dissolved oxygen. Limnological differences between lakes may be cited as the main causes of the observed heterogeneous distribution of macrophytes. CONCLUSIONS: These results indicate the importance of limnological characteristics of the different environments in the macrophyte community composition and the potential role of this community as a bioindicator in shallow lakes in southern Brazil
O que as organizações entendem por gestão de talentos?
Este estudo objetiva discorrer sobre práticas que constituem a denominada Gestão de Talentos (GT) no universo corporativo. O tema que, segundo o estudo da SHRM (2013), ocupa, desde 2010, a primeira posição na lista de prioridades dos lÃderes de RH, aparece também no Brasil como tendência principal em polÃticas e práticas de Gestão de Pessoas (GP). A denominada guerra pelos talentos associa-se a um momento de transição da sociedade industrial para a pós-industrial. Apesar de ser um tema emergente e presente em muitos debates mundiais que abordam o futuro da GP, observa-se ainda confusão nas definições existentes sobre os termos talento e Gestão de Talentos, instigando o questionamento: o que especificamente é Gestão de Talentos? Embora reconheçam a carência de uma definição consistente sobre o termo, alguns autores afirmam que uma boa GT é muito importante para a estratégia da empresa. A Gestão de Talentos não tem ainda um significado claro. A multiplicidade de conceitos revela a necessidade de estudos que ofereçam avanços significativos no entendimento do assunto. Em que realmente acreditam as empresas que hoje utilizam a expressão Gestão de Talentos em seus discursos e práticas de gestão? Em que a GT se diferencia da GP? Qual o objetivo da identificação e/ou diferenciação dos talentos dentro de cada estrutura organizacional pesquisada? A presente pesquisa propõe, através de análise comparativa, uma reflexão sobre tais inquietações. Para isso foram entrevistados gestores de três organizações de natureza distinta, todas de grande porte, localizadas no estado do Rio Grande do Sul, região sul do Brasil, tendo elas um elemento em comum: a presença do termo Gestão de Talentos em suas retóricas. A análise comparativa dos resultados oferece avanços na compreensão do assunto e demonstra a confusão de conceitos ainda existente, possÃvel reflexo da atual miscelânea literária de definições
