Antidepressive and anxiolytic effects of ostruthin, a TREK-1 channel activator

We screened a library of botanical compounds purified from plants of Vietnam for modulators of the activity of a two-pore domain K+ channel, TREK-1, and we identified a hydroxycoumarin-related compound, ostruthin, as an activator of this channel. Ostruthin increased whole-cell TREK-1 channel currents in 293T cells at a low concentration (EC50 = 5.3 ÎĽM), and also activity of the TREK-2 channel (EC50 = 3.7 mM). In contrast, ostruthin inhibited other K+ channels, e.g. human ether-Ă -go-go-related gene (HERG1), inward-rectifier (Kir2.1), voltage-gated (Kv1.4), and two-pore domain (TASK-1) at higher concentrations, without affecting voltage-gated potassium channel (KCNQ1 and 3). We tested the effect of this compound on mouse anxiety- and depression-like behaviors and found anxiolytic activity in the open-field, elevated plus maze, and light/dark box tests. Of note, ostruthin also showed antidepressive effects in the forced swim and tail suspension tests, although previous studies reported that inhibition of TREK-1 channels resulted in an antidepressive effect. The anxiolytic and antidepressive effect was diminished by co-administration of a TREK-1 blocker, amlodipine, indicating the involvement of TREK-1 channels. Administration of ostruthin suppressed the stress-induced increase in anti-c-Fos immunoreactivity in the lateral septum, without affecting immunoreactivity in other mood disorder-related nuclei, e.g. the amygdala, paraventricular nuclei, and dorsal raphe nucleus. Ostruthin may exert its anxiolytic and antidepressive effects through a different mechanism from current drugs

Building Comprehensive Sex Education Plans for Teenagers: Groundwork-Based Research Design Application

Although sexual education is an important part of a child’s education and upbringing, in Vietnam, sex education is not an official subject in school, and young people’s awareness of Comprehensive Sex Education (CSE) is still limited. While sex education at school is necessary, it is not the only way nor sufficient enough to change teenage sexual behavior. A parent’s role could involve communicating about values, providing a positive family environment, and monitoring their children’s behavior. This topic explores the awareness of teenagers about CSE and the availability of teaching CSE in Vietnamese schools and families. Data was collected via an online survey of 89 teenagers and 119 parents whose children are teenagers. The findings revealed that the target group is not fully aware of CSE and in particular, they also feel the importance of the family in this regard but are afraid to share gender issues with their parents and tend to find information on their own through the internet. LEARNING IDIOMS FOR ENGLISH MAJORS: VIETNAMESE STUDENTS’ PERCEPTIONS OF DIFFICULTIES AND LEARNING STRATEGIES

Learning idioms play an influential role in language generally and in English notably. Comprehending idioms assists language learners in integrating culture, enhancing skills, and ameliorating English levels. Numerous studies have analyzed the function of idioms in second language acquisition (Cieślicka, 2015). This study investigates the difficulties and strategies used in learning idioms by English-majored students at a regional public university (PU) in the south of Vietnam. This paper furnishes data showing learners’ perceptions of facing complications and learning methods. The samples consisted of 150 undergraduate EFL students from English-medium instruction programs. The data was analyzed by utilizing descriptive and inferential statistics. The findings reveal that students struggle to understand idiomatic terms without specific, understandable contexts. Furthermore, the results indicate that the most frequently employed strategies are guessing the meaning of idioms, learning idioms through keywords, and learning from a range of sources, particularly via media. The findings also mentioned that low-proficiency and high-proficiency students encounter identical challenges, with no significant differences. The study's results revealed that the majority of students have difficulty acquiring, recognizing, and interpreting idioms. The findings indicated that guessing the implication of idioms is the most used strategy.  Article visualizations

General sensitivity analysis in data assimilation

    Get PDF
International audienceThe problem of variational data assimilation for a nonlinear evolution model is formulated as an optimal control problem to nd the initial condition function (analysis). The operator of the model, and hence the optimal solution, depend on the parameters which may contain uncertainties. A response function is considered as a functional of the solution after assimilation. Based on the second-order adjoint techniques, the sensitivity of the response function to the parameters of the model is studied. The gradient of the response function is related to the solution of a non-standard problem involving the coupled system of direct and adjoint equations. The solvability of the non-standard problem is studied. Numerical algorithms for solving the problem are developed. The results are applied for the 2D hydraulic and pollution models. Numerical examples on computation of the gradient of the response function are presented

Groundwork-Based Research to Design Application SCC - Building a Sustainable Community for Children in Mountainous Area

Children’s clothing is a prime example of fast fashion, as their continuous growth requires frequent purchases. This not only impacts the environment but also puts financial strain on parents. Concurrently, many highland children lack essential material and educational resources, contributing to poverty in the region. To address these challenges, local authorities urgently require a sustainable solution that supports children, communities, and localities, fostering comprehensive development. The project development criteria are evaluated based on the United Nations’ 17 sustainable development goals (SDGs). Data was collected through an online survey of 50 married individuals, 162 non-married individuals, and interviews with those who have organized events for children in the highlands. Additionally, insights were obtained from three children from disadvantaged areas. The results indicate that all target groups show a keen interest in social activities for children. Parents facing difficulties accessing charities, and the lack of effective collaboration between charities and local authorities, hinders sustainable development efforts. Large displacements of FGSW beams in thermal environment using a finite element formulation

    Get PDF
The large displacements of functionally graded sandwich (FGSW) beams in thermal environment  are studied using a finite element formulation. The beams are composed of three layers, a homogeneous core and two functionally graded face sheets with volume fraction of constituents following a power gradation law. The material properties of the beams are considered to be temperature-dependent.  Based on Antman beam model and the total Lagrange formulation, a two-node nonlinear beam element taking the effect of temperature rise into account  is formulated and employed in the study. The element with explicit expressions for the internal force vector and tangent stiffness matrix is derived using linear interpolations and reduced integration technique to avoid the shear locking. Newton-Raphson based iterative algorithm is employed in combination with the arc-length control method to compute the large displacement response of a cantilever FGSW beam subjected to end forces.  The accuracy of the formulated element is confirmed through a comparison study. The effects of the material inhomogeneity, temperature rise and layer thickness ratio on the large deflection response of the beam are examined and highlighted
